Abstract

Granules containing procaine penicillin which are readily dispersible in water to give an injectable suspension, are prepared by grinding a mixture of procaine penicillin and a water-soluble salt of penicillin until it has a particle size between 50 and 150m , then adding thereto an organic solvent to form a paste, forcing the paste through a sieve and freeing the granules so formed from the organic solvent. The organic solvents are preferably non-solvents for the penicillin salts, e.g. acetone, methyl ethyl ketone, petroleum ether, n-hexane, dithyl ether, di-isopropyl ether, beta methoxyethanol, dioxan and diethyl carbonate. Optional ingredients which are incorporated in the penicillin salt mixture prior to grinding are (a) non-ionic surface active agents such as condensation products of alkyl phenols with ethylene oxide or of sorbitan mono-oleate, -stearate, -palmitate or -laurate with ethylene oxide; (b) buffers, e.g. sodium citrate, and (c) bacteriostatic agents, e.g. phenyl-mecuric acetate. In the case of (a) and (e), it is preferred to incorporate these substances in the presence of an organic solvent, which is a non-solvent for the penicillin salts, and to remove the solvent prior to the grinding operation.
