Abstract
Desoxycholic acid is isolated from a mixture of bile acids, by adding to the mixture a base in aqueous solution capable of causing preferential solution of the cholic acid. Any weak base have a dissociation constant 1 x 10-5 to 1 x 10-8 affords a good separation, e.g. ammonia, morpholine and triethanolamine. The base is added in an amount just sufficient to cause solution of the bile acids at an elevated temperature and on cooling the desoxycholic acid separates. When utilizing a volatile amine such as ammonia the base may be conveniently removed by evaporation depositing the desoxycholic acid. Any residual cholic acid in the deposited desoxycholic acid may be removed by treating with an aqueous isopropyl alcohol mixture made weakly basic by addition of 1 per cent of a weakly basic inorganic salt equal in chemical equivalent to sodium bicarbonate.
