Abstract
N-(2-pyridyl)-N-(p-chlorobenzyl)-N1, N1-dimethyl-ethylenediamine is prepared by reacting N - (2 - pyridyl) - N1,N1 - dimethyl - ethylenediamine with a reactive ester of p-chlorobenzyl alcohol, e.g., a halide ester, in the presence of an acid-binding agent, such as sodamide, which is capable of exchanging a hydrogen atom by a metal atom. In a modification, the same compound is made by reacting 2-(p-chlorobenzyl)-amino pyridine with a reactive ester of b -di-methylamino ethanol in the presence of the acid-binding agent, and in another modification, N-(p-chlorobenzyl)-N1,N1-dimethyl-ethylene diamine is heated with a 2-halogen-pyridine in the presence of an acid-binding agent such as pyridine, quinoline or potassium carbonate, or a catalyst such as copper-bronze, which promotes the splitting off of hydrochloric acid. In examples: (1) 2-(b -dimethylaminoethyl)-amino pyridine is first boiled with sodamide in the presence of toluene and the product then reacted with p-chlorobenzyl chloride; (2) 2-bromopyridine, N,N-dimethyl-N1-(p-chlorobenzyl)-ethylenediamine (prepared by boiling an alcoholic solution of p-chlorobenzylamine with dimethylamino-ethyl chloride) and pyridine are heated together; (3) N-(2-pyridyl)-N1,N1-dimethyl - ethylenediamine, p - chlorobenzyl chloride, sodamide and toluene are heated together; and (4) 2 - (p - chlorobenzylamino)-pyridine (produced by reductive condensation of 2-aminopyridine with p-chlorobenzaldehyde by means of formic acid) is first reacted with sodamide in the presence of benzene and then heated with b -dimethylamino ethyl chloride. The Specification as open to inspection under Sect. 91 comprises also a modification in which a reactive ester of N-(2-pyridyl)-N-(p-chlorobenzyl aminoethanol is reacted with dimethylamine. It describes a process in which 2-(p-chloro-benzyl)-aminopyridine is reacted with an a b -dihalogen ethane, a glycol di-aryl (or-alkyl)-sulphonic acid ester or an aryl sulphonic acid-b -halogen ethyl ester and the esterified b -hydroxyethyl group then converted to the b -dimethylamino-ethyl group; and also a process in which a 2-(b -hydroxyethyl)-aminopyridine is first formed from 2-aminopyridine and glycol chlorohydrin and then reacted with a reactive ester of p-chlorobenzyl alcohol. This subject-matter does not appear in the Specification as accepted.
