Abstract

A proteolyzed preparation is obtained by subjecting mammalian liver material to proteolysis by the action of pancreatic enzymes at a temperature of 35-65 DEG C in an aqueous medium having a pH range of 5.0-7.0, heating the resulting aqueous mixture to destroy the enzymes and coagulate undigested material therein, and separating the aqueous liquor from the coagulated material. The enzymes may be in the form of pancreas gland material or of an enzyme preparation obtained from pancreas gland material. The resulting aqueous preparation may be purified, fat being precipitated by adding alcohol to produce a mixture containing 70 per cent alcohol, and after removing the precipitated material, the active material remaining in the liquid being precipitated by increasing the alcohol concentration to 95 per cent. The precipitated material may be dissolved in water to provide an active aqueous preparation. If desired the aqueous solution may be further purified by precipitating it by saturation with ammonium sulphate, dissolving the precipitate in aqueous alcohol and, after the removal of excess of sulphate, evaporating the alcohol to produce a concentrated active aqueous preparation. The aqueous proteolyzed preparation, with or without subsequent purification, as described, may be used for oral administration, or may be dried to produce a product suitable for oral administration. The aqueous preparation may be made suitable for parenteral administration by sterilization. The active preparation contains riboflavin pyridoxine, folic acid and nicotinic acid.
