Abstract
Penicillin salts are purified by alternate extractions from aqueous solutions into an aliphatic alcohol of 4 or 5 carbon atoms or a mixture of these compounds and from organic solvent solutions into water at a pH of 4.5-8.0, preferably in the range 6.0-8.0 The process may suitably be applied to the ammonium, substituted ammonium, sodium, potassium and rubidium salts of penicillin. The process may be used to effect partial separation of salts of the individual penicillins, e.g. on extraction from an aqueous phase to an organic solvent phase penicillin X remains in the aqueous phase and on re-extraction with water penicillin K remains in the organic solvent. The extraction may be performed by continuous counter-current methods. When the solvent is butyl alcohol a ratio of butyl alcohol to aqueous phase of between 1 : 1 and 4 : 1 suitably between 2.0 : 1 and 1 : 1 is used for extraction from the aqueous phase and a ratio of water to butyl alcohol of less than 1 : 1, preferably between .03 : 1 and 1 : 1 is used for extraction from the butyl alcohol phase. In examples, solutions of sodium, ammonium, potassium and calcium penicillins are extracted successively with butyl alcohol and water giving in some cases partial separation of the various penicillins. The Provisional Specification describes also the use in the process of the present invention of organic solvents in general.
