1500464 Applying a reagent to molecular separation media MARINE COLLOIDS Inc 29 March 1976 12587/76 Heading G1B [Also in Divisions B1 and C7] A device for the application of a measured quantity of water-soluble or water dispersible reagent to a water containing solid medium usable in molecular diffusion or affinity separation procedures, e.g. chromatography or electrophoresis, consists of a solid film comprising a film-forcing solid organic polymeric binder soluble in water (as defined) to the extent of at least 1 wt. per cent at 20‹C and incorporated within the binder film a measured quantity of reagent, the film being of a size and shape so that when it is placed in contact with the medium, the reagent and binder diffuse completely into the medium. Reagents which may be incorporated into the film of binder may be any of the water-soluble or water dispersible reagents employed in assay procedures, e.g. antibodies, antigens, enzymes, enzyme substrates, precipitin brightners, stains, precipitants, buffers, salt, microbiological organisms, as well as radio-actively tagged or fluorescent reagents of the foregoing types. The device may include a supporting backing of water insoluble synthetic plastics bonded to one face of the film. The binder may be selected from dextran, polyacrylamide, polyacrylic acid, polyvinyl alcohol, polyethylene glycol, polyethylene oxide, polyvinyl pyrrolidone, guar, carboxymethyl cellulose, hydroxyethyl cellulose, methyl cellulose, algin, carrageenan, xanthan gum, starch, and copolymers of maleic anhydride with vinyl monomers.
