Abstract
1470246 Antibiotic BN-109 MEIJI SEIKA KAISHA Ltd 8 July 1974 [17 July 1973] 30240/74 Heading C2A A process for producing the antibiotic BN-109 comprises cultivating an antibiotic BN-109 producing strain of the genus Bacillus polymyxa under aerobic conditions and recovering the antibiotic from the medium or broth. Antibiotic BN-109, which is effective in inhibiting the growth of gram-negative bacteria, has the following properties: Appearance: white powder as free base; Solubility: insoluble in common organic solvents and water; slightly soluble in dilute acid and alkali, acetic acid, dimethyl formamide and dimethyl sulphoxide, and soluble in acidic methanol and acidic ethanol; Mobility on filter paper electrophoresis: Behaves as an alkaline substance; Rf values on TLC: 0À68 (n butanol acetic acid-water (2 : 1: 1)); 0À38 (n butanol-acetic acid-water (4 : 1 : 5, upperlayer)),0-66(n-butanol-pyridine-acetic acetic-water (6 : 4 : 1 : 3)); [α]<SP>25</SP> (C=0À5, meth- D anol-acetic acid (1 : 1)): -4‹; Maximum UV absorption bands: Found at 279 mÁ (E1%= 1cm 34 in 0À1 N-NaOH, E1%=23 in 0À1 N-HCl); 1cm IR absorption bands: Found at 3275, 2590, 1632, 1535, 1408, 1110 (cm<SP>-1</SP>); Colour reactions: positive: ninhydrin, Ehrich and Dragendorff; negative: Sakaguchi, Pauly and Trevelian; Melting point: 186-188‹ C.; Elementary analysis: C 52À75%, H 7À27%, N 12À77%; Amino acid composition: α,γ-diaminobutyric acid (4), serine (2), glutamic acid (1), glycine (1), alanine (1), valine (3), phenylalanine (1), tryptophan (2), and an unknown amino acid; Fatty acids: contain 30% of acid ether-soluble fatty acids; Molecular weight: in the range of from 1900 to 2200. The antibiotic may be used as the effective ingredient in preparations used in the treatment and/or prevention of swine scours.
