Abstract
1415080 7-Amino-3-cephem compounds and derivatives and intermediates therefor MERCK & CO Inc 5 Nov 1973 [13 Nov 1972] 51302/73 Headings C2C and C3P 7#-Aminocephalosporins of formula wherein R 1 is a blocking group and R<SP>1</SP> is a heterocyclic or an optionally substituted phenyl group, are prepared by reacting an acid with a novel 7#-imino compound of Formula III wherein R 2 is the residue of the ketone or aldehyde, which is obtained by epimerizing the novel 7α-imino compound of Formula II which is obtained from 7α-aminocephalosporin of Formula IB by reaction with R 2 =O. The 7α-aminocephalosporins may be obtained via the intermediates VI, VII and VIII shown as outlined in the reaction scheme B The 7#-aminocephalosporins may be acylated to give 7#acylamino-cephalosporins. The preparation of a compound IV used in reaction scheme B, together with various phosphonate intermediates, is also described, each R in the compound IV being ethyl and R 1 being pmethoxy benzyl.
