Abstract
1403883 Polymers containing morpholine groups; enzymes entrapped in polymers; separating proteins KOCH-LIGHT LABORATORIES Ltd 15 Sept 1972 [15 Sept 1971] 43093/71 Headings C3P C3H and C3U [Also in Division C2] The invention comprises a cross-linked polymer, at least some of the repeating units of the backbone chains of which, have attached to an atom constituting the backbone chain, a side group of the formula sufficient cross-links existing between the backbone chains to make the polymer capable of forming a gel in water and in chloroform. The polymer may be made by (a) polymerizing in the presence of a cross-linking agent, an ethylenic monomer containing a morpholine group attached to a carbon atom of the double bond either directly or via a methylene or carbonyl group, (b) reacting an addition polymer with morpholine or a morpholine group-containing compound and a cross-linking agent, to introduce groups of the above formulae or (c) reacting an already cross-linked addition polymer with such a morpholine compound. Method (a) is preferred, being exemplified by the copolymerization of N-acryloyl-morpholine and N,N<SP>1</SP>-methylene-bis-acrylamide. Other monomers which may be used are N-methacryloyl-, N-ethacryloyl- and N-propacryloylmorpholine, and N-allyl morpholine. Method (b) may be achieved using a poly-(allyl halide) or poly-(acryloyl halide) reacted with morpholine and a cross-linker, and method (c) using a cross-linked polyacrylamide and morpholine. The polymers may be used as a chromatographic medium for the separation of molecules of differing molecular weight and for the entrapment of molecules. In the examples copolymers of N-acryloyl morpholine and N,N<SP>1</SP>-methylene-bis-aorylamide are used to separate solutions containing (numbers refer to the Examples) (5) Blue dextran, dextran T.10 and glucose; dextran T.10 and glucose; blue dextran, raffinose and glucose and (12) blue dextran, NaCl and glucose. Also, in Example 7 the same polymer is used to entrap or immobilize the enzyme urease and in Example (12) a mixture of the proteins egg albumin and ribonuclease A is separated.
