Abstract

1377674 Tetracycline complex; piperazine biguanide-formaldehyde compound SPA-SOC PRODOTTI ANTIBIOTICI SpA 22 June 1973 [11 July 1972] 32367/72 Heading C2C A novel complex of equimolar amounts of tetracycline, formaldehyde and piperazine biguanide is prepared by reacting the component substances with each other, or by reacting (i) a piperazine biguanide-formaldehyde reaction product with tetracycline or (ii) a piperazine biguanide-tetracycline reaction product with formaldehyde. The reaction may be performed in a solvent (e.g. methanol or ethanol) at about 5-30‹ C. The said complex may be salified with one or two equivalents of an organic or inorganic acid to give a mono- or di-acid addition salt. Preparation of the intermediates referred to in (i) and (ii) above viz. a 1: 1 piperazine biguanide-formaldehyde compound, and a 1 - 1 piperazine biguanide-tetracycline compound) is described. Also the preparation of piperazine biguanide free base by treatment of the bihydrochloride salt with methanolic NaOH is described. The novel complexes and their salts may be admixed with a pharmaceutical carrier to provide antibiotic pharmaceutical compositions.
