Abstract

1358511 Enzyme assay test device GEOMET Inc 5 May 1972 21005/72 Heading G1B An enzyme assay test device comprises a support and a plurality of test spots on the support, the test spots comprising (i) at least one (control) spot containing all reagents for the assay together with a standard quantity of the enzyme, (ii) a (blank) spot containing all reagents for the assay except the substrate for the enzyme, (iii) a (test) spot containing all reagents for the assay, each spot comprising a plurality of discrete porous laminae 16, 18, 26 impregnated with the reagents, the laminae in each spot being in a rigidly confined stacked array, for example by means of retaining ring 20 and polystyrene slides 10, 12, the array including a filter disc 24, and the support allowing visual inspection of the lowermost lamina of each stacked array. Discs 16, 18 may be of glass fibre and disc 26 of cellulose acetate. An Example relates to assay of creatine phosphokinase (CPK), when zones A, B, C contain CPK standards representing normal, high and elevated levels of CPK, zone D provides a blank and zone E the assay reagents. In use of the assembled unit, a drop of water is applied to spots A, B, C and a drop of whole blood to spots D and E, and the unit is inverted for observation of colour development.
