1343213 3-[1-(4-Phenyl-4-oxobutyl)-4-hydroxy- 4 - piperidyl] - dihydro - 2(3 H ) - furanones SANDOZ Ltd 19 April 1971 [20 March 1970 2 Sept 1970] 24384/71 Heading C2C Novel 3-[1-(4-phenyl-4-oxobutyl)-4-hydroxy- 4-pyridyl]-dihydro-2(3H)-furanones of the general formula wherein R 1 is a hydrogen atom or C 1-3 alkyl group and R 2 is a hydrogen, fluorine, chlorine or bromine atom and acid addition salts thereof are prepared by condensing a dihydro-4-(4- hydroxy-4-piperidyl)-2(3H)-furanone and a butyrophenone derivative of the respective general formulµ wherein X is the acid radical of a reactive ester, in the presence of an acid-binding agent, followed optionally by salification of a resulting free base or conversion of a resulting salt to the free base. Pharmaceutical compositions having analgesic activity comprise, as active ingredient, a 3-[1-(4- phenyl - 4 - oxobutyl) - 4 - hydroxy - 4 - piperidyl]-dihydro-2(3H)-furanone of the first general formula above or a pharmaceutically acceptable acid addition salt thereof, in association with a pharmaceutical carrier or diluent.
