Abstract

1299488 Critical point drying IMPERIAL CHEMICAL INDUSTRIES Ltd 25 May 1971 [9 June 1970] 27838/70 Heading A2D [Also in Division F4] The water in a biological sample 26 is replaced by ethyl alcohol (which is miscible with liquid carbon dioxide), the sample 26- is placed in a pressure vessel 10 (which is then sealed by valves 14 and 16), the vessel 10 is cooled to a temperature (e.g. O‹C by means of an ice-water bath or coils carrying cooling fluid) below the critical temperature (31‹C) of carbon dioxide, carbon dioxide gas is admitted from a cylinder 15 (at 20-25‹C) to the vessel 10 so that it condenses, fills the vessel 10, and replaces the alcohol in the sample, the vessel 10 is warmed to a temperature (e.g. 40‹C) above the critical temperature of carbon dioxide so that carbon dioxide gas is formed without a phase boundary passing through the sample, and the carbon dioxide gas is allowed to escape from the vessel 10 at a controlled rate, to produce the dried sample. The wet sample 26 (mounted on a slide) is initially passed through a series of water-alcohol baths of increasing alcohol content, placed in a depression 25, and covered with alcohol. The warming of the vessel 10 may be with a hot water bath, or coils carrying heating fluid, or electrical means.
