Abstract

1294463 Cellulose-antigen reaction products MILES LABORATORIES Inc 29 Oct 1969 [1 Nov 1968] 53071/69 Heading C3H [Also in Division B1] An immunological reagent comprises particles of a water-insoluble cellulose derivative having chemically linked thereto an antigen or antibody, itself having an acidic indicator material adsorbed on it. The cellulose derivative is preferably p-aminobenzyl or p-aminomethoxy benzyl cellulose and may be made reactive to immunological materials by diazotization. The indicator may be hematoxylin, fluorescein or titan yellow, and prior to its adsorption on to the antigen or antibody-treated particles any unreacted diazo groups on the particles may be treated with a dye-forming material, for example beta-naphthol. In use of the reagent, the immunological counterpart of the antigen or antibody in the reagent may become attached to the reagent particles, releasing the indicator into solution; alternatively the immunological counterpart may itself become attached to the indicator and the indicator colour remains with the particulate material. A spot of the reagent may be placed on a bibulous strip. The strip may then be "developed" with the sample under test or the sample may be placed on the reagent spot and the strip developed with saline. In the examples, the antigens or antibodies are human myoglobin, human haemoglobin, rabbit anti-HCG, HCG, rabbit anti-human haemoglobin, mule anti- HCG. Other antigens and antibodies are mentioned.
