Abstract

1,210,719. #-N-tert-butyloxycarbonyl N- carboxy anhydrides of lysine and ornithine. MERCK & CO. Inc. 28 June, 1968 [30 June, 1967], No. 30947/68. Heading C2C. # - N - tert - butyloxycarbonyl - lysine - N- carboxy anhydride and the corresponding ornithine compound are prepared by reacting the corresponding (# or 8)-N-tert-butyloxycarbonyl amino acid respectively with phosgene at 15-40‹ C. in an inert liquid reaction medium in the presence of an HCl-acceptor. The HCl-acceptor is preferably an oxide or inorganic salt of Ag, Pb or Hg. The liquid medium is preferably a solvent for the HClacceptor but a non-solvent for the reaction product of HCl and the acceptor. In the Examples the HCl acceptors AgCN, Ag 2 O and Hg(CN) 2 and the solvents acetonitrile, dioxane and acetone are employed. The above compounds are used in the preparation of tert - butyloxycarbonyl - lysyl - isoleucine, lysyl - isoleucine, ornithyl - isoleucine and lysylisoleucyl-glycyl-valyl-glycine.
