Abstract
1,208,398. Determination of creatine phosphokinase. CALBIOCHEM. 17 April, 1969 [14 June, 1968), No. 19765/69. Addition to 1,163,409. Heading B1X. A substantially anhydrous solid assay material for dissolving in water for determination of creatine phosphokinate (CPK) comprises a mixture of the substrates creatine phosphate and glucose, the enzymes hexokinase and glucose 6-phosphate dehydrogenase (G-6-PDH), the coenzymes adenosine diphosphate (ADP) and triphosphopyridine nucleotide (TPN), a buffer to maintain the pH between 6.5 and 7.5, a stabiliser which is a disulphide compound other than cysteine and optionally one or more of the stabilisers acacia, tris(hydroxymethyl) aminomethane-sulphate and ammonium sulphate, and an activator comprising a water soluble magnesium salt, for example magnesium aspartate. The disulphide stabiliser may be dithiothreitol or dithioerythritol. In the test system the reactions are:- creatine phosphate + ADP CPK creatine + ATP glucose + # ATP hexokinase glucose-6-P + ADP glucose # -6-P + TPN G-6-PDH 6-phosphogluconate + # TPNH. The rate of change of optional density in the system is measured.
