Abstract
1,197,758. Tetrasodium salt of reduced nicotinamide - adenine - dinucleotide phosphate. ARZNEIMITTELWERK DRESDEN VEB. 6 Sept., 1967, No. 40806/67. Heading C2C. The tetrasodium salt of reduced nicotinamideadeninedinucleotide phosphate is prepared by applying a solution of reduced nicotinamideadeninedinucleotide phosphate to a column of DEAE cellulose and eluting with a volatile buffer medium having a pH of less than 8 and having a linear increasing gradient of a sodium halide which is soluble in alcohol, the pH of the eluate being then adjusted to about 10 by adding a dilute aqueous solution of sodium hydroxide. The elution is preferably carried out at about 4‹ C. Good results are obtained by using the system pyridine-acetic acid or ethylene diamine-acetic acid at a pH below 8, e.g. 7À6-7À8, as the volatile buffer and using NaI or NaBr as the sodium halide.
