Abstract

1,178,364. N-Acyl-peptides. FARBWERKE HOECHST A.G. 15 March, 1968 [18 March, 1967], No. 12772/68. Heading C2C. [Also in Division C3] A process for the production of N-acylpeptides comprises (i) reacting a salt of an N- acyl-amino acid or -peptide with a tertiary amine with a bromo- or chloro-acetylated crosslinked polystyrene at 0-100‹ C., (ii) separating in acid medium an N-acyl group from the resulting polymer and reacting this deacylated product with an N-acyl-amino acid or -peptide by means of dicyclohexylcarbodiimide and N- hydroxy-succinimide or with an N-acyl-amino acid or -peptide-hydroxysuccinimide ester, (iii) optionally repeating step (ii) as many times as desired and (iv) separating the N-acyl peptide from the polymer with sodium thiophenolate at room temperature optionally in the presence of sodium iodide. Examples describe the production of CBO- phe - leu - ala - ala - OH with CBO - phe - leu- O-N-hydroxysuccinimide ester as by-product and BOC - pro - pro - phe - phe - ala - ala - OH with BOC - pro - pro - phe - phe - O - N- hydroxysuccinimide as by-product.
