Abstract
1,147,403. Sulphonyl-ureas and related oxazolidinones. F. HOFFMANN-LA ROCHE & CO. A.G. 4 Aug., 1967 [8 Aug.. 1966], No. 35845/67. Heading C2C. Novel compounds of the general formula, X-NHCONHSO 2 Y and of the general formula wherein X represents a 2-(OXO or hydroxy)- cyclohexyl or 2-(OXO or hydroxy)-cyclopentyl group, Y represents a phenyl group which may be substituted by one or more amino, halo, alkyl, alkoxy, alkylthio, acyl, acylamino, diacylamido, alkoxy-substituted benzamidoethyl, halo-substituted benzamido-ethyl and/or alkoxy-halo-substituted benzamido-ethyl substituents or Y is a nitrogen-linked 5- or 6- membered nitrogen - containing heterocyclic group and X<SP>2</SP> is a divalent radical derived from a 2-hydroxy-cyclohexyl or 2-hydroxycyclopentyl group by removal of the hydroxy group, are prepared by the interaction of a compound of genera] formula, and either a sulphonyl carbamic acid' HOOC.NHSO 2 Y, or a sulphonyl isocyanate, YSO 2 NCO, thus affording the novel cyclic compounds illustrated above and when either of the cyclic compounds illustrated above are treated with alkali the novel compounds, X.NHCONHSO 2 Y, result. These latter compounds are also prepared: (a) by reacting a reactive derivative of a carbamic acid Y.SO 2 NH.COOH, or a sulphonyl isocyanate, Y.SO 2 .NCO, with 2-amino-cyclohexanol, 2- amino-cyclohexanone, 2-amino-cyclopentanol or 2 - amino - cyclopentanone or (b) by reacting a compound of the general formula, X<SP>1</SP>NH.CONHSO 2 Y, wherein X<SP>1</SP> is a 2-hydroxycyclohexyl or 2-hydroxy-cyclopentyl group which has been acylated at the hydroxy group with the carbamic acid, Y.SO 2 NH.COOH, with 2-amino-cyclohexanol or 2-amino-cyclopentanol. In addition, if desired, ring-located keto groups may be reduced and hydroxy groups oxidized. N - (4 - Chlorobenzenesulphonyl) - 3,5 - dimethylpyrazole - (1) - carboxamide is obtained by the interaction of 3,5-dimethyl pyrazole and 4-chloro-benzenesulphonyl-isocyanate Pharmaceutical preparations having hypoglycaemic action contain as active ingredient the above novel compounds and may be administered orally.
