Abstract

Derivatives of the tetracyclines with D(-)a -aminobenzyl-penicillin of the general formula <FORM:1092198/C2/1> in which the tetracyclic fraction may be tetracycline itself (R = H; R1 = H; R11 = CH3) or a derivative thereof such as hydroxytetracycline (R = OH; R1 = H; R11 = CH3), chlorotetracycline (R = H; R1 = Cl; R11 = CH3) or dimethylchlorotetracycline (R = H; R1 = Cl; R11 = H) and X is H, an organic base, e.g. procaine or an amine or an alkali or alkaline earth metal, are prepared by reacting D(-)a -aminobenzylpenicillin in the form of the free base or salt with the appropriate tetracycline in the presence of formaldehyde. When the acid tetracycline derivative is formed initially (D(-)a -aminobenzylpenicillin free base used), the process may include the additional step of salt formation. The reaction is conducted in the presence of a solvent at room temperature.
