Abstract

Compounds of the formula <FORM:1071815/C2/1> <FORM:1071815/C2/2> and their mono- and di-quaternary ammonium salts formed by reaction with one or two moles of R5X where A is a C1- 2-alkylene residue B is straight or branched chain alkylene residue containing a straight chain of 3-5 C between the two N atoms, R1 is a C1- 4 alkyl group, R2 and R3 each represents a C1- 4 alkyl group or they may be joined to represent with the common N atom, a heterocyclic ring of 5-8 atoms and optionally containing a second hetero atom, R4 is H, C1- 4 alkyl, CF3, halogen or acetyl, R5 is a C1- 4 alkyl group and X is Cl, Br or I o R5SO4 are prepared either by reacting the tertiary base with R5X or by first forming the base by condensing a phenothiazine with an aminoalkylaminoalkanoyl halide, a haloalkanoylan aminoalkanoyl- or a haloalkylaminoalkanoylphenothiazine with an aminoalkylamine, a haloalkylamine or an amine respectively and optionally quaternating the tertiary base or by replacing the tertiary bases in the forgoing reaction by the corresponding quarternary bases. N - Methyl - N - 3 - methylaminopropylpiperidinium chloride is prepared from N-3-chloropropyl-N-methyl piperidinium chloride and methylamine. Pharmaceutical compositions comprise the mono- and di-quaternary ammonium salts of the invention and a carrier therefor. The compositions, which are useful in the treatment of bronchospasm and spasm of smooth muscle, are formulated for oral and parenteral administration and also in the form of an aerosol.
