Abstract

<FORM:1069777/C2/1> An antibiotic Rhi 12-648, active against yeasts, fungi and tumours and also having sedative and cramp-inhibiting action, has a m.pt. of 197-199 DEG C.; [a ]22D = +20.7 DEG (C = 1.57 in CHCl3); U.V. maximum at 265 mn log e = 4.17; I.R. spectrum as in Fig. 2; elementary analysis C = 59.1, H = 4.6, O = 25.1 and Cl = 10.0%, probable formula C18H17 O6Cl, molecular weight 368; violet colour with addition of FeCl3 to methanol solution; yellowish red with NaOH and brownish violet with conc. H2SO4; soluble in ethyl acetate, benzene, butyl acetate, chloroform and butanol; forms a dimethyl ether by treatment with ethereal diazomethane; forms a diacetate by treating the antibiotic with acetic anhydride and anhydrous sodium acetate. Antibiotic Rhi 12-648 is produced by the cultivation of Rhizoctonia repens N Bernard NRRL 3183, NRRL 3184 and NRRL 3185, or an Rhi 12-648 producing mutant thereof, in a nutrient medium. The antibiotic is recovered from the clarified broth by (a) extraction with a solvent, e.g. ethyl acetate, (b) chromatography of the crude from (a) on activated alumina, silica gel or magnesium silicate, (c) counter current distribution of crude from (a). A pharmaceutical composition comprises antibiotic Rhi 12-648, its di-O-acyl or its di-O-alkyl derivatives and a carrier.
