Abstract

Novel compounds of the general formula <FORM:1068035/C2/1> (wherein A represents O or S, R represents a hydroxyalkyl, haloalkyl, alkoxycarbonylalkyl, thiocarbamoylalkyl, aminoalkyl, amino-substituted carboxyalkyl, phenylalkyl, nuclear carboxy-substituted phenylalkyl, alkenyl, cycloalkyl, aryl, carboxyphenyl, phenyl substituted by an alkoxycarbonyl or an amido group, halophenyl, alkylphenyl, alkoxyphenyl, alkanoyl, thioalkanoyl, benzoyl or 2-(g -L-glutamylamino)- 2 - (N - carboxymethylcarbamoyl) - alkyl radical, or, in bis-compounds, a carboxymethoxybenzoylalkyl, carboxymethylmercaptobenzoylalkyl, carboxymethoxybenzoylalkylmercapto-(C1- 5 alkyl) or carboxymethylmercaptobenzoylalkylmercapto -(C1- 5 alkyl) radical, R1 represents H or a C1- 5 alkyl radical, R2 represents H or a C1- 5 alkyl, trihalomethyl- or hydroxy-substituted C1- 5 alkyl, C1- 5 alkoxy or alkylthio, or cycloalkyl radical or a phenyl or phenoxy optionally carrying halogen or C1- 5 alkyl or alkoxy substituents, and each of R3, R4, R5 and R6 represents H, halogen or a trihalomethyl, C1- 5 alkyl or alkoxy, carboxyalkoxy, OH, NO2 or acylamino radical or two of them on adjacent ring carbon atoms are linked to form a second 5- or 6-membered carbocyclic ring are prepared by reacting a compound of the general formula <FORM:1068035/C2/2> with a mercaptan R*SH (wherein R* has any of the meanings of R except the last four), or, for the bis-compounds, with H2S or a C1- 5 alkanedithiol. The preparation of the starting materials by the process of Specification 998,835 is described. Pharmaceutical compositions, e.g. tablets, pills, capsules and injections, having diuretic, natriuretic and chloruretic action, comprise, in association with an inert diluent, carrier or coating, a compound of the first general formula above, which may be in the form of a salt. In an example the active ingredient is prepared in the presence of various additives, giving a powdered mixture to which pyrogen-free water is added before use, whilst in another example an injectable suspension is produced directly by carrying out the reaction for preparing the active ingredient in pyrogen-free water in the presence of the additives.
