Abstract
A pituitary gonadotropic hormone is obtained from menopausal or post-menopausal human urine by acidifying the urine to pH 4-4.5, adding thereto 0.5% by weight thereof of siliceous or diatomaceous earth and 1% by weight of kaolin and separating the solid by centrifugation after agitating for about an hour. Thereafter, the solids are washed (using acidified water of pH about 4.5) and the active material removed therefrom by elution with 1M aqueous ammonia to give an eluate of pH 11 to 11.3. This eluate is adjusted to pH 8.5 to give a precipitate which is removed and the pH of the supernatant is adjusted to 5-5.5 and about 2 volumes of acetone are added and the whole is left overnight. The precipitate obtained is washed (using acetone, ethanol and ether successively), dried and taken up in 70% aqueous alcohol containing about 10% ammonium acetate, from which a precipitate is obtained by addition of ethanol containing 10% ammonium acetate. The precipitate is dissolved in 0.05M phosphate buffer (pH 7.0) and stirred with diethylaminoethyl cellulose. The supernatant is acidified to pH 5.4 and passed through a synthetic alumino-silicate gel cation exchanger column from which the active material is subsequently eluted with 40% aqueous ethanol containing 10% ammonium acetate and precipitated by addition of 95% ethanol. Thereafter, the active material may optionally be further purified by passing an aqueous solution thereof through a "Sephadex" (Registered Trade Mark) column and collecting the first fractions of the effluent. The product comprises follicular stimulating hormone (FSH) and luteinizing hormone (LH) having an FSH:LH activity ratio of at least 3:1 and has a total gonadotropic activity (determined by the mouse uterus test) of above 80 mg. equivalent I.R.P. per mg.
