Abstract
1,051,382. Stabilized red blood cell suspension. WARNER-LAMBERT PHARMACEUTICAL CO. Feb. 11, 1965 [March 10, 1964], No. 6075/65. Heading B1X. A stabilized red blood cell suspension comprises aldehyde-treated red blood cells in a buffered iso-tonic aqueous medium containing a macromolecular compound compatible with said cells, and a surface active agent. The cells may be derived from warm-blooded animals including chicken, sheep, bovine species, and monkeys, and from human beings; the aldehyde employed may be formaldehyde, pyruvaldehyde or paraldehyde; the buffer may be phosphate buffer; the macromolecular compound may be polyvinyl pyrrolidone, dextran, gum acacia, gum tragacanth or gelatin; and the surface active agent may be polyoxyethylene sorbitan monooleate. The invention also comprises a method of producing stabilized red blood cell suspensions comprising mixing in a phosphate buffer of pH7, a suspension of said cells in isotonic saline solution with an aldehyde, allowing to stand at 4-10‹C., washing with isotonic saline to free from aldehyde and resuspending in an aqueous medium comprising a macromolecular compound. In the Example, pyruvaldehyde, adjusted to pH7 with Na 2 CO 3 , is used, and sodium azide is added as a preservative with the surface active compound, to the final suspension.
