Abstract
The invention comprises sulpha-pyrimidines of the formula <FORM:1022966/C2/1> wherein one of the symbols R1 and R2 represents a lower alkoxy-lower alkyl group (the word "lower" indicating a group of up to 6 carbon atoms) and the other is a hydrogen or halogen atom or a lower alkyl, lower alkoxy, lower alkoxy lower alkyl or lower alkoxy-lower alkoxy group, R3 is a halogen atom or a lower alkyl or alkoxy group and Z1 is an amino, nitro, azo or acylamino group, N1-acyl derivatives thereof and salts, e.g. alkali-metal and alkaline earth metal salts of the compounds wherein Z1 is an amino group; and processes for their production by reacting a benzenesulphonyl halide, p-substituted by an amino, nitro, azo or acylamino group, with the appropriate 4-aminopyrimidine, or reacting a benzenesulphonamide, p-substituted by an amino, nitro, azo or acylamino group, with a 4-halg or 4-trialkylammonium - substituted pyrimidine and, in either case, in any desired order, converting the p-nitro-, acylamino- or azo-substituent in the resulting compound, if present, to an amino group, by reduction or hydrolysis, splitting any resulting bis-benzenesulphonyl compound by hydrolysis, N1-acylating the product, and converting a resulting free compound into a salt or a resulting salt into a free compound. 2-R1-4-R2-5-R3-6-R7-Pyrimidines used as starting materials, in which R7 is a free hydroxy or amino group, an ammonium group such as a lower trialkylammonium group, or a halogen atom, are made by reacting an amidine R1.C(:NH)NH2 or a salt thereof, with a compound of the formula R8.CO.CH(R3).R9, in which R8 is a lower alkoxyalkyl group or, if R1 is a lower alkoxyalkyl group, R8 may be hydrogen, lower alkyl or etherified hydroxy, and R9 is an esterified carboxyl, thioamide or nitrile group or an oxo derivative thereof such as an enol ether or imine; and if desired, converting any hydroxy group in the 4- or 6-position of the resulting compound into a free amino group, and/or a halogen atom in the 6-position into a free amino group, and/or a halogen atom in the 4-position into a lower alkoxy or lower alkoxy-alkoxy group or replacing the halogen atom in the 4-position by hydrogen and/or converting a halogen atom in the 6-position into an ammonium group. Amidines of the formula R1.C(:NH).NH2 are made by converting a nitrile R1.CN into an imido ether and reacting this with ammonia. Pharmaceutical preparations having antibacterial activity comprise the above sulphanilamides of the invention (Z=NH2) and their physiologically tolerable salts, in admixture or conjunction with a pharmaceutical carrier. The preparations suitably take forms adapted to enteral, parenteral or topical administration, e.g. tablets, dragees, salines, creams, solutions, suspensions or emulsions.
