Description
Field of the Invention: The present invention relates to the field of animal nutrition. In particular, the present invention relates to an antioxidant composition based on aerial parts of nettle (Urtica dioica) and apple pomace (Malus sylvestris or Pyrus malus) intended for animal feed. The present invention also relates to a food comprising said antioxidant composition, as well as its use in the feed of farm animals to contribute to the protection of their cellular constituents against oxidative damage.
STATE OF THE ART Oxidative stress is the imbalance between the production of free radicals and the antioxidant defenses. Under normal conditions, free radicals are produced continuously in the body of an animal / mammal and are destroyed by antioxidant defenses. This process is harmful to the body and is an important cause of damage to cell structures including lipids, membranes, proteins and DNA. Free radicals, involved in a large number of redox reactions, are regularly generated in the amount necessary and sufficient for the survival of the cell. However, in animals, in certain physiological situations (pregnancy, lactation, ...), pathological (infections, inflammation, hepatic steatosis, ...) or food (intensive diet, restriction phase, acidosis .. .), as well as under certain conditions of stress of environmental origin (climatic, physical, emotional, ...), excessive production of free radicals within the cell can occur. This increase can be very detrimental to the integrity of the various cellular constituents of the animal and, in particular, membrane lipids. The exposure of cells to these free radicals has led the body to develop a series of defense mechanisms. Mechanisms of defense against free radicals induced by oxidative stress include: prevention mechanisms, repair mechanisms, physical defenses and antioxidant defenses. 2 Antioxidant defenses, in particular enzymatic, include superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT). The main non-enzymatic antioxidants are ascorbic acid (vitamin C), a-tocopherol (vitamin E), glutathione (GSH), carotenoids, flavonoids and other polyphenols. Antioxidants, synthesized by the body or brought by the diet, are therefore necessary for homeostasis of the body to avoid an imbalance of free radicals, this imbalance resulting in the deterioration of the health of the animal, and this also causes a deterioration of the nutritional and organoleptic qualities of the meat and milk derived from the animal. In practice, the food of farm animals and in particular ruminants, does not contain enough micronutrients with antioxidant action and it is necessary to provide a complementation. To limit these excessive oxidation phenomena consecutive to one of the particular situations detailed above, it is recommended to add to the ration of the animals, additives capable of limiting the oxidation reactions, or by neutralizing the free radicals (before those it does not attack the cellular constituents), either by preventing the propagation of oxidation reactions once these have been initiated.
The antioxidant additive most used in animal nutrition is vitamin E, in particular because of its specific action (especially with regard to lipids) and its low cost. However, vitamin E intake has disadvantages because that used in animal nutrition is of synthetic origin. Its antioxidant activity has a threshold effect beyond which vitamin E can be not only less active, but instead be pro-oxidant. Then, vitamin E only intervenes in a limited number of stages of the peroxidation reaction chain. The invention therefore aims to provide an antioxidant composition for animals and in particular for ruminant, which is at least as effective as those existing on the market at present.
Description of the invention: For this purpose, the subject of the present invention is an antioxidant composition intended for the nutrition of farm animals, characterized in that it comprises aerial parts of nettle and apple pomace.
Preferably, the aerial parts of nettles are those of Urtica dioica and the apple pomace is derived from a wild apple tree, such as Malus sylvestris or Pyrus malus. The present Applicant has surprisingly discovered that the combination of a crushed aerial part of nettle (such as Urtica dioica) and apple pomace (such as Malus sylvestris, Pyrus malus, etc.) has superior antioxidant activity. to that corresponding to the sum of the activities of each of the components of this composition. This makes it possible to reduce the amount of free radicals, increase the antioxidant defenses and reduce the cellular damage (proteins, lipids, DNA) related to oxidative stress. The stinging nettle (Urtica dioica L.), also known as stinging nettle or common nettle suitable for the present invention, is a herbaceous perennial plant of the family Urticaceae and the genus Urtica. Nettle consists mainly of polyphenols, polysaccharides and minerals (quercetin glucosides and rutinosides, kaempferol and isorhamnetin, caffeoylmalic acid, chlorogenic acid, neochlorogenic acid, caffeic acid, scopoletin (coumarin), sitosterol (-3-O-glucoside)) . This plant is generally used for applications: food (salad, vegetable) both in humans and animals, agricultural (nettle leaf slurry to kill or repel insects and as fertilizer), industrial (widely used fiber) to make ropes, threads and clothes), or medicinal. Apple pomace, for example from Malus sylvestris or Pyrus malus apple trees, is a by-product of apple juice production. It is rich in fiber and polyphenols. It consists mainly of cellulose, and contains minerals, such as zinc, copper, manganese, potassium and lesser amounts of calcium, phosphorus and magnesium.

The present invention also relates to the use of the antioxidant composition as described above, to be distributed in addition to a food ration and / or a drink of a farm animal of the type polygastric (bovine, ovine, goat, ...), or monogastric (pork, poultry, rabbit, ...). As mentioned above and for convenience, the antioxidant composition may be in the form of a dietary supplement or a dietary supplement.
Another object of the invention relates to an animal feed, characterized in that it comprises the antioxidant composition as defined above.

The present applicant has analyzed the content of polyphenols in apple pomace which contains 209.8 mg of polyphenols (expressed in catechin equivalent) per 100 g of apple pomace extract, which corresponds to a value expected for this type of product.
Development and validation of an improved oxygen radical absorbance capacity assay using fluorescein as the fluorescent probe. J Agric Food Chem. 2001 Oct;49(10):4619-26). Comparative Example G The present applicant has demonstrated the antioxidant activity of said composition comprising ground mashed nettle and apple pomace, by Oxygen Radical Absorbance Capacity (ORAC) analysis of the antioxidant capacity. The composition analyzed corresponds to the composition of Example A, namely a composition comprising 35% of aerial part of nettle and 65% of apple pomace (by weight relative to the total weight of the composition according to the invention). The ORAC method allows a global measurement of the antioxidant power of pure or mixed substances as in finished products. The first step consists in extracting the active ingredients in a mixture of water and ethanol for the water-soluble molecules and in ethanol for the lipophilic ones. The test is carried out in spectrofluorimetry. It consists of a measurement of the protection exerted by a given molecule against the oxidation of fluorescein by a stable free radical, AAPH (2,2-azobis (dimethylaminopropane chlorohydrate). The results are expressed in relation to the protection exerted by a reference antioxidant, Trolox®, which is the water-soluble equivalent of vitamin E and reduced by gram of the test product. ORAC test is to propose a standardized and widely accepted measure of antioxidant power, in particular, the protocol that has been followed is that of the Brunswick, USA laboratory (reference laboratory in the field), II was published in the publication et al (Hampsch-Woodill M, Prior RL, Development and validation of an improved oxygen radical absorbance capacity using fluorescein as the fluorescent probe.) J Agric Food Chem., 2001 Oct; 49 (10): 461 9-26) Thus, 0.5 g of a first sample comprising aerial parts of nettles, 0.5 g of a second sample comprising apple pomace and 0.5 g of the composition according to Example A were each taken up in 20 ml of extraction buffer (acetone / water mixture, 1: 1). Then adequate dilutions of each sample were made so that it could be compared in the Trolox response range. The acquisition of the kinetics was made on a fluorescence microplate reader and the results obtained are expressed in pmol of Trolox equivalent / g of product. Samples Results (pmoles of Trolox® equivalents / g of sample) Aerial parts of nettles 1600 Marc of apple 170 Antioxidant composition (invention) 1960> 1770 Thus, the composition according to the invention has an antioxidant activity greater than that corresponding to the sum of the activities of each of the components of this composition, namely 1960 versus 1770 (pmoles of Trolox® equivalents / g of sample) when the effect of aerial parts of nettles and that of apple pomace is added. The present invention also aims at the use of the food as described above, to prevent the premature aging of cells and to protect them against the damage caused by free radicals in farm animals, in particular the ruminant. Indeed, the present applicant has found that the antioxidant composition or the feed comprising said composition is effective in ruminants, (in particular cattle, sheep and goats), since the products which are effective in the monogastric are not necessarily in the polygastric. The total amount of said antioxidant composition administered to the animal is preferably from 0.02 mg to 200 g per day and per kg of body weight. The antioxidant composition according to the invention thus has the advantage of effectively limiting the oxidation of cellular components (DNA, proteins, lipids) caused by free radicals and by oxidative stress, so as to preserve the zootechnical performances, the well-being and the health of the animal as well as the nutritional and organoleptic qualities of its products (meat, milk and dairy products, eggs).
Bien que l'invention ait été décrite en relation avec un mode de réalisation particulier, il est bien évident qu'elle n'y est nullement limitée et qu'elle comprend tous les équivalents techniques des moyens décrits ainsi que leurs combinaisons si celles-ci entrent dans le cadre de l'invention.25 Although the invention has been described in connection with a particular embodiment, it is obvious that it is not limited thereto and that it comprises all the technical equivalents of the means described and their combinations if they fall within the scope of the invention.
