Abstract

Extn. of antigenic complexes comprises solubilising the walls of microorganisms (previously purified) by the simultaneous action of sodium desocycholate (0.5-5 mg per mg of proteins), lysozyme (1 mg/ml) and sodium ethylene-diaminotetracetate (25 mmoles). Pref. the solubilisation is effected at neutral pH and 20 degrees C for 12-24 hrs.; the extract is then centrifuged and the soluble supernatant fraction is used directly as antigen source without removal of the solubilising agent. Prod. obtd. comprises a pseudomicellar reassociation (formed by hydrophobic interaction) of hydrophobic proteins, lepopolysaccharides and peptidoglycons in the presence of sodium desoxycholate to give inter- and intra-specific antigenic complexes in which the antigenic sites are not denaturised. The sodium desoxycholate remaining in the prod. aids formation of the soluble, hydrophobic antigenic complexes and helps protect them against protease contaminants. Used for prepn. of vaccines e.g. upper respiratory and bronchial tract vaccines. The process allows large scale prepn. of antigen complexes using large scale homogenisers, centrifuges etc. The prod. is a new hybrid complex prepd. from known strains which preserves the structural integrity qf the antigen molecule and allows more rapid recognition by the specialised cells of the organism.
