Abstract

Concn. of a protein in an antigen sample is quantitatively measured by immunoelectrophoresis. This includes selective protein migration by electrophoresis in one dimension in a thin layer of supporting medium. The antigen is then subjected to immunodiffusion with an antibody source contg. an antibody specific to the protein. The protein and antibody diffuse into reactive contact in another dimension through an area of the medium layer initially free from antigen and antibody, and they react to form an elongated pptn. zone. The zone is scaned optically under dark field illumination at a selected time after the diffusion starts. There are parallel, spaced scan paths that cross the zone transversely to develop electrical intensity signals (the intensity of scattered light at posns. on the path, partly in the zone and partly out). From the signals an electrical sum signal for the sum of the effective excess of each intensity within the zone over intensities outside the zone. This signal is compared with reference signals produced with controls in a similar procedure. The process facilitate distinction among pptes. formed by different proteins; so that quantitative and reliable results are obtd. Rapid, reliable and economic process is used for the analysis of serum, spinal fluid, tissue extracts etc. used in clinical diagnosis.
