Abstract

Thymus factor polypeptide hormone (I) is a new cpd. (I) is a solid with (i) isoelectric point at pH 7.5 plus-or-minus 0.1; (ii) no affinity for active charcoal; (iii) mol. wt. 700-3000 as determined by molecular sieve and membrane filtrations; (iv) is bound to carboxymethylcellulose (CMC) at pH below 7; (v) is bound to diethylaminoethyl-cellulose at pH above 9; (vi) has rf value on cellulose plates of 0.32 (20:20:9 methanol/CHCl3/NH4OH); 0.8 (0.01N acetic acid and 1.0 (distilled water); (vii) contains no aromatic amino acids and (vii) on 'Sephadex' (RTM) G-25 elutes after 2.1 dead-space volumes with pH 7.3 0.2M phosphate buffer. (I) has the formula Pyroglutamyl-Ala-Lys-Ser-Gln-Gly-Gly-Ser-Asn. and is prepd. from defibrinated blood by first dialysing through a polyacrylonitrile membrane, then dialysis and successive chromatography. (I) is used in treatment of autoimmune disorders, esp. lupus erythematosus, and also in geriatric medicine for selective stimulation of T-cell activity. It is administered by intravenous or intramuscular injection pref. at 10-4 to 10-2 mg./kg./day.
