Abstract

Pyrrolidinone derivs. of formula (I) (where R1 and R2 are linked so as to form with the pyrroline nucleus an isoindoline, 6,7-dihydro-5H-pyrrolo 3,4-b pyrazine, 2,3,6,7-tetrahydro-5H-oxathiino 1,4 2,3-c -pyrrole or 2,3,6,7-tetrahydro-5H-dithiino 1,4 2,3-c -pyrrole nucleus; Het is 2-pyridyl, 2-quinolyl or 1,8-naphthyridin-2-yl opt. substd. by 1-3 substituents from halogenM 1-4C alkyl, 1-4C alkoxy and CN; Z is O or S; and R is H, 1-4C alkyl (opt. substd. by 1-3 halogen atoms), 2-4C alkenyl, 2-4C alkynyl, 3-6C cycloalkyl, 1-4c alkoxy, phenyl (opt. substd. by 1-3 substituents from halogen, 1-4C alkoxy, 1-4C alkyl, NO2 and CF3), phenyl-(1-4C alkyl)(opt. substd. in the phenyl residue as indicated for phenyl), or to 5- or 6- membered heterocyclyl with 1 or 2 hetero atoms selected from N, O and S; and when R1 and R2 are linked so as to form an isoindoline residue this opt. substd. by 1-4 substituents from halogen, 1-4C alkyl, 1-4C alkoxy, NO2 and CF3), e.g. 3- (4-acryloyl-1-piperazinyl)carbonyloxy -2-(7-chloro-1,8-naphthy- ridin-2-yl)-1-isoindolinone, are new cpds. (I) are tranquillisers, anticonvulsants, hypnogenic agents or hypnotics active at doses of 0.1-100 mg/kg p.o. in the mouse. Toxicity is low (mouse LD50 generally >300 mg/kg p.o.).
