ESTA INVENCION SE REFIERE A METODOS PARA AISLAR LOS MUTANTES DE ANULACION DE VIBRIO CHOLERAE. EN UN METODO, LA ANULACION ES PREDETERMINADA POR LA DIGESTION CON ENDONUCLEASAS DE RESTRICCION DE ESPECIFICIDAD CONOCIDA. LAS ANULACIONES SE INSERTAN EN EL CROMOSOMA VIBRIO CHOLERAE MEDIANTE UNA RECOMBINACION IN VIVO ENTRE UN PLASMIDO QUE LLEVA LA ANULACION DESEADA, CON SECUENCIAS DE FLANQUEO ADYACENTES Y EL CROMOSOMA VIBRIO CHOLERAE. EN OTRO METODO, UN EVENTO DE RECOMBINACION IN VIVO INICIAL DE SECUENCIAS HOMOLOGAS DEL PLASMIDO RECOMBINANTE EN EL CROMOSOMA PROPORCIONA UN MARCADOR SELECCIONABLE EN ESTE LUGAR. UN SEGUNDO EVENTO DE RECOMBINACION IN VIVO ENTRE SECUENCIAS DE FLANQUEO HOMOLOGAS DA COMO RESULTADO UNA EXCISION DE LOS GENES COMPETENTES DEL CROMOSOMA CON EL PRODUCTO FINAL SIENDO UNA MUTACION DE ANULACION.
