METODO PARA PRODUCIR Y PURIFICAR UBIQUITIN-HIDROLASA, ASI COMO PARA DISOCIAR CONJUGADOS DE UBIQUITINA-POLIPEPTIDO. SE TRANSFORMAN CELULAS HOSPEDANTES CON VECTORES DE EXPRESION QUE CODIFICAN UBIQUITIN-HIDROLASA, Y SE CULTIVAN DICHAS CELULAS PARA EXPRESAR HIDROLASA. SE HOMOGENEIZAN PASTAS DE CELULAS EUCARIOTICAS, RECUPERANDO LA PORCION CON ACTIVIDAD DE UBIQUITIN-HIDROLASA, SE SEPARA EL PRECIPITADO Y SE PONE EN CONTACTO CON RESINA INTERCAMBIADORA IONICA, Y DESPUES CON RESINA DE AFINIDAD HIDROFOBA, RECUPERANDO LA FRACCION ABSORBIDA A LA RESINA, Y SEGUIDAMENTE CON RESINA DE CROMATOGRAFIA, RECUPERANDO LA FRACCION ABSORBIDA A ESTA RESINA, Y NUEVAMENTE CON RESINA INTERCAMBIADORA IONICA, RECUPERANDO LA FRACCION CON ACTIVIDAD DE HIDROLASA Y ASILANDO IBIQUITIN-HIDROLASA.
