Abstract
В настоящем изобретении предусмотрен способ непрерывного производства на этапе накопления для получения продуктов на основе биспецифических антител, содержащих по меньшей мере два связывающих домена. Способ включает, по меньшей мере, стадии (i) обеспечения в перфузионном биореакторе по меньшей мере одной культуры клеток млекопитающих, которая способна экспрессировать продукт на основе биспецифического антитела, (ii) выращивания культуры клеток млекопитающих при первой скорости перфузии до момента достижения заданного значения плотности жизнеспособных клеток и (iii) поддержания перфузионной культуры при второй скорости перфузии, где концентрацию продукта на основе биспецифического антитела в биореакторе поддерживают ниже порогового значения. Затем продукт на основе биспецифического антитела подвергают последующей обработке на этапе выделения и очистки. Более того, в настоящем изобретении предусмотрен продукт на основе биспецифического антитела, полученный с помощью способа непрерывного производства на этапе накопления.
