Claims (10)
1. Fremgangsmåde til fremstilling af fytasevarianter med en forbedret termostabilitet, inklusive trinnene med indføring af seks proliner på site S80, S151, T161, N176, S187 og A380 af fytasen med aminosyresekvensen, der er anført i SEQ ID NO:2, hvor fytasevarian-terne er mindst 70 % homologe med aminosyresekvensen, der er anført i SEQ ID NO:2.
2. Fytasevarianter med en forbedret termostabilitet med følgende kendetegn: med mindst 70 % homologi med aminosyresekvensen, der er anført i SEQ ID NO:2.; og seks proliner er indført på site S80, S151, T161, N176, S187 og A380 af aminosyresekvensen, der er anført i SEQ ID NO:2.
3. Polynukleotid, der koder for fytasevarianterne med en forbedret termostabilitet ifølge krav 2.
4. Nukleotidkonstruktion omfattende polynukleotidet ifølge krav 3 og en regulerende sekvens, der eventuelt er forbundet til et eller flere af polynukleotiderne for at styre ekspressionen i ekspressionsværten.
5. Rekombinant ekspressionsvektor omfattende nukleotidkonstruktionen ifølge krav 4.
6. Rekombinant værtscelle omfattende den rekombinante ekspressionsvektor ifølge krav 5.
7. Rekombinant værtscelle ifølge krav 6, som er kendetegnet ved, at værtscellen er en bakteriecelle eller svampecelle.
8. Fremgangsmåde til fremstilling af fytasevarianter ifølge krav 2 med den forbedrede termostabilitet omfattende trinnene med (a) dyrkning af den rekombinante værtscelle ifølge krav 6 til 7 for at fremstille supernatan-ten indeholdende fytasevarianterne og (b) genfinding af fytasen.
9. Anvendelse af fytasevarianterne med en forbedret termostabilitet ifølge krav 2 som foderadditiv.
10. Anvendelse af den rekombinante værtscelle ifølge krav 6 i fremstilling af foderadditiver.
