Claims (30)
- PATENTKRAY1. Anvendelse af en ikke-integratsv HIV-ientiviral vektorpartikel omfattende et defekt integraseprotein og yderligere omfattende et RNA-genom som er transkriptionsproduktet af et vektorgenom omfattende et polynukleotid kodende for mindst et antigent polypeptid, to LTR'er og en DNA-f!ap omfattende en cRPT og et GTS-domæner, ved fremstillingen af en immunogen sammensætning ti! fremkaldelse af et beskyttende immunrespons mod mindst en af nævnte antigene polypeptid for at forhindre infektion med et patogen eller maligne tilstande i et menneske der administreres med nævnte immunogene sammensætning omfattende doser af nævnte HIV ientiviral vektorpartikler i området fra 1 pg ti! 100 pg af p24-antigen, hvor nævnte defekte integraseprotein er en integrase som har en klasse i-mufation til specifikt at ændre sin integrase-aktivitet.
- 2. Anvendelse af en HIV-lentivirai vekforpartikel ifølge krav 1, hvor nævnte lentivirusvektor Ikke er repiikativ,
- 3. Anvendelse af en HIV-lentivirai vektorpartikel ifølge et hvilket som helst af kravene 1 til 2, hvor nævnte immunrespons er et beskyttende humoralt immunrespons.
- 4. Anvendelse af en HIV-lentivirai vektorpartikel ifølge et hvilket som helst af kravene 1 til 2, hvor nævnte immunrespons er et cellulært immunrespons.
- 5. Anvendelse af en HIV-ientivirai vektorpart!kel ifølge krav 4, hvor nævnte immunrespons er et GD8-medieref cellulært immunrespons.
- 6. Anvendelse af en HIV-ientiviral vektorpartikel ifølge krav 4, hvor nævnte immunrespons er et CD4-medieret cellulært immunrespons.
- 7. Anvendelse af en HIV-lentivirai vektorpartikel ifølge krav 1, hvor nævnte mutation er D64V.
- 8. Anvendelse af en HlV-lentiviral vektorpartikei ifølge et hvilket som helst af kravene 1 til 7, hvor det lentivirale genom er blottet for gag, pol og/eiler env lentiviral gener,
- 9. Anvendelse af en Hl V-lentiviral vektorpartikei ifølge et hvilket som helst af kravene 1 til 7, hvor det lentivirale genom er blottet for funktionelle gag, pol og/eiler env lentivirale gener.
- 10. Anvendelse af en HIV-ientivira! vektorpartikei ifølge et hvilket som helst af kravene 1 til 8, hvor det lentivirale genom er blottet for alle endogene kodende lentivirale sekvenser.
- 11. Anvendelse af en HIV-ientiviral vektorpartikei ifølge et hvilket som helst af kravene 1 til 10, hvor det lentivirale genom omfatter (a) en LTR5! og en LTR3!, (b) en psi-sekvens, (c) en DNA-flap omfattende en cRPT og et GTS-domæner og (d) en sekvens kodende for mindst en af nævnte polypeptid.
- 12. Anvendelse af en Η i V-lentivi ral vektorpartikei ifølge krav 11, hvor nævnte lentivirale genom yderligere omfatter et splejsningsdonorsted (SD), et spiejs-ningsacceptorsted (SA) og et Rev-responsivt element (RRE).

- 17. Anvendelse af en HIV-lentiviral vektorpartikel ifølge et hvilket som helst af kravene 1 til 16, hvor mindst en af nævnte polypeptid er kodet af en sekvens der stammer fra genomet af et patogen.
- 18. Anvendelse af en HiV-lentiviral vektorparti kel ifølge krav 17, hvor nævnte patogen er en virus, især retrovirus, ientivirus, fiavivirus eller coronavirus, en bakterie eller en parasit.
- 19. Anvendelse af en HiV-lentiviral vektorpartikel ifølge krav 18, hvor mindst en af nævnte polypeptid er afledt fra kappen af AiDS-virus, herunder HIV-1 eller HIV-2, fra kapsidet af HIV eller fra kappen af gul feber-virussen, vestnilvi-russen, denguevirussen (DV), japanske hjernebetændelsevirussen (JEV) eller den SARS-associerede coronavirus.
- 20. Anvendelse af en HIV-lentiviral vektorpartikel ifølge et hvilket som helst af kravene 1 fil 16, hvor mindst en af nævnte polypeptid omfatter en tumorepitop eller antigen.
