Description
PATENTKRAV 1. Fremgangsmåde til fremstilling af et immunoglobulinmolekyle med tunge og lette kæder eller fragmenter deraf, som er forbundet således, at det overordnede molekyle udviser ønskede bindings- og genkendelsesegenskaber, idet fremgangsmåden omfatter (a) dyrkning af en transformeret eller transfekteret prokaryot vært under passende betingelser, sådan at tungt kædeimmunoglobulin og let kædeimmunoglobulin eller fragmenter deraf udtrykkes som et resultat af transskription af polynucleotid-sekvenser i værten, hvor polynucleotidsekvenseme koder for mindst én funktionelt virkende region af en variabel region af et antistof, og et prokaryot sekretionssignalpeptid, hvor den variable region under ekspression operabelt bindes ved dens N-ende til et prokaryot sekretionssignalpeptid til muliggørelse af sekretion fra værten; og (b) opnåelse af immunoglobulinkæden eller fragmentet af en immunoglobulin-kæde, som forud for sekretion fra værten oprindeligt var således bundet til signal-peptidet i form af en immunoglobulinmolekyle med tunge og lette kæder eller fragmenter deraf, at det overordnede molekyle udviser de ønskede bindings- og genkendelsesegenskaber. 2. The method of claim 1, wherein the secretion signal peptide is a pectal lyase secretion signal. The method of claim 1 or claim 2, wherein the immunoglobulin molecule is isolated from the periplasmic compartment. The method of claim 1 or claim 2, wherein the immunoglobulin chain or fragment of an immunoglobulin chain is isolated from the culture growth medium. A method according to any one of the preceding claims, wherein the immunoglobulin molecule is an Fd fragment. The method of any of the preceding claims, wherein the immunoglobulin molecule comprises a chimeric immunoglobulin chain. The method of any of claims 1-4, wherein the variable region of antibody is operably linked to a sequence encoding a polypeptide other than an immunoglobulin chain. The method of claim 7, wherein the polypeptide is an enzyme.
