Abstract

Process for the production of a specified protein, characterised in that the following operations are carried out starting with a strain of the genus Bacillus in the chromosome of which the following have been incorporated into a nonessential gene of the said strain. a) an amplifiable unit comprising a specified DNA sequence M framed by two identical DNA sequences D oriented in direct direction, the sequence M comprising the gene for the structure of the said protein placed under the control of expression and secretion elements capable of being induced into Bacillus of the said protein and optionally a selectable gene, and b) a replication system or replicon capable of being induced or conditional also incorporated into the said nonessential gene of the chromosome of the strain upstream of the amplifiable unit; 1. the amplified clones obtained by culture on a selection medium corresponding to the selectable gene are optionally selected and the strains whose genome corresponds to the presence of a number of copies of the said gene which is enhanced in relation to the bacterial population before selection are abstracted; and 2. a gene amplification is induced or conditioned in an appropriate culture medium by the activation of the replicon and the expression and the secretion of the protein and the activation of the said expression elements are induced, 3. the said expressed protein is recovered. The invention also relates to a process for the preparation of a Bacillus strain, amplified or otherwise, employed especially in the production process according to the invention.
