Abstract
Pharmazeutische Zusammensetzung, umfassend kristalline Macitentan freie Base, gekennzeichnet durch ein Röntgenpulverbeugungsmuster mit Peakmaxima bei 2 Theta/°-Werten von 11,4 ± 0,2, 13,0 ± 0,2, 16,1 ± 0,2 und 25,4 ± 0,2, wenn eine Strahlungswellenlänge von 1,5419 Å verwendet wird, und mindestens einen Hilfsstoff.
Claims (13)
- Pharmazeutische Zusammensetzung, umfassend kristalline Macitentan freie Base, gekennzeichnet durch ein Röntgenpulverbeugungsmuster mit Peakmaxima bei 2 Theta/°-Werten von 11,4 ± 0,2, 13,0 ± 0,2, 16,1 ± 0,2 und 25,4 ± 0,2, wenn eine Strahlungswellenlänge von 1,5419 Å verwendet wird, und mindestens einen Hilfsstoff.
- Pharmazeutische Zusammensetzung nach Anspruch 1, welche eine orale feste Darreichungsform ist.
- Pharmazeutische Zusammensetzung nach Anspruch 1 und/oder feste orale Darreichungsform nach Anspruch 2, umfassend höchstens 5 Gewichts-%, bevorzugt höchstens 1 Gewichts-%, amorphe Macitentan freie Base.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 3 als eine komprimierte Darreichungsform, bevorzugt eine Tablette, besonders bevorzugt eine Tablette mit sofortiger Freisetzung.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 4, umfassend mindestens einen Füllstoff, mindestens ein Sprengmittel, mindestens ein Bindemittel, mindestens ein Schmiermittel und mindestens ein Tensid.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 5, wobei die kristalline Macitentan freie Base eine Teilchengrößenverteilung D98% von höchstens 680 μm und D5% von mindestens 0,5 μm aufweist, gegebenenfalls auch eine D50% von 3 μm bis 250 μm, insbesondere von 15 μm bis 150 μm.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 6, wobei die feste orale Darreichungsform an Patienten in einem Land zu verabreichen ist, welches nach der Köppen-Geiger Klimaklassifikation eine Region mit einem Af oder einem Am Klima aufweist, bevorzugt einem Af Klima.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 7, wobei die pharmazeutische Zusammensetzung in einem Verpackungsmaterial verpackt ist, welche eine Wasserdampftransmissionsrate von mindestens 0,4 g m–2 d–1 aufweist, gemessen nach der Norm DIN 53122-1, wobei das Verpackungsmaterial bevorzugt aus Polypropylen, Polyvinylidenchlorid und/oder Polyvinylchlorid hergestellt ist.
- Orale feste Darreichungsform nach irgendeinem der Ansprüche 2 bis 8 zur Verwendung bei der Behandlung von pulmonaler arterieller Hypertonie in Patienten in einem Land, welches nach der Köppen-Geiger Klimaklassifikation eine Region mit einem Af oder einem Am Klima aufweist.
- Verfahren zur Herstellung einer pharmazeutischen Zusammensetzung, bevorzugt der oralen festen Darreichungsform nach irgendeinem der Ansprüche 2 bis 8, wobei das Verfahren umfasst: a) Bereitstellen von kristalliner Macitentan freier Base nach Anspruch 1, b) Mischen der in a) bereitgestellten kristallinen Macitentan freien Base mit mindestens einem Hilfsstoff, c) Herstellen der pharmazeutischen Zusammensetzung, bevorzugt der festen oralen Darreichungsform, aus dem in b) erhaltenen Gemisch.
- Verfahren nach Anspruch 10, wobei die kristalline Macitentan freie Base aus Schritt a) eine Zusammensetzung von Kristallen aus kristallinem Macitentan freier Base nach Anspruch 1 ist, welche eine Partikelgrößenverteilung D98% von höchstens 680 μm und D5% von mindestens 0,5 μm aufweist.
- Verfahren nach Ansprüchen 10 und 11, wobei die kristalline Macitentan freie Base aus Schritt a) eine Zusammensetzung aus Macitentan freier Base von Form I Kristallen ist, welche eine Partikelgrößenverteilung D50% von 3 μm bis 250 μm, bevorzugt von 15 μm bis 150 μm, aufweist.
- Verwendung von kristalliner Macitentan freier Base nach Anspruch 1 zur Herstellung einer oralen festen Darreichungsform, umfassend kristalline Macitentan freie Base nach Anspruch 1, und mit, nach dem Verpacken in eine Polypropylenfolie und Lagerung in der Dunkelheit bei 40°C bei einer relativen Luftfeuchtigkeit von 75% für einen Zeitraum von mindestens 14 Tagen, erhöhter chemischer Stabilität, verglichen mit einer identisch verpackten und gelagerten festen oralen Darreichungsform, umfassend, anstelle der kristallinen Macitentan freien Base, amorphe Macitentan freie Base.
