Description
The present invention relates to a method for extracting nutrient components from pollen by using enzyme.
There are two types and four methods for extracting nutrients from pollen so far. The first type is the physical extraction of nutrients from pollen. It features that the molecular structure of original nutrients of pollen is kept unchanged. This type contains three methods: namely a mechanical wall-breaking warm water extraction method, a temperature difference extraction method and a solvent extraction method. The nutrient components of the pollen extract obtained by the method are mainly pollen protein which can be absorbed by human body only through the action of enzyme in the human body. The pollen extract is difficult to filter and has low product yield. If the beverage is prepared by using the protein, the phenomenon of protein solidification can be generated in the process of heating sterilization, and the appearance and the quality of the product are seriously influenced. Therefore, only pastry, food and cosmetics can be made with it. The second type is that the protein in pollen is converted into various amino acids which are easily absorbed by human body directly by means of biochemical reaction. The prior art for this type of process is a fermentation process. In the method, the reproduction and growth of microorganisms need to consume part of nutrients of pollen in the fermentation process, so that the yield is low. Meanwhile, the production period is long (the solid fermentation period is 10-20 days, and the liquid fermentation period is 4-5 days).
The task of the invention is to research a new method for extracting nutrient components from pollen. The method has short production period and high product yield, and the nutrient components of the extracting solution are various amino acids easy to be absorbed by human body.
The present invention features that the catalytic action of enzyme is utilized to convert the protein in pollen into amino acids easy to be absorbed by human body. The enzymes selected may be: enzyme preparations such as papain, bromelain, bacterial protease or mixed protease. The invention is to put the pollen material into a container with stirring, soak the pollen material with warm drinking tap water, fully wet the pollen cells, and adjust the pH value and temperature of the material in the container to meet the condition requirements of enzyme catalytic reaction. Adding enzyme preparation, stirring, and allowing the materials in the container to start enzyme catalysis reaction, so that the protein in pollen is gradually converted into various amino acids. Heating to kill enzyme after reaction (for some food needing enzyme preservation, other disinfection methods can be adopted, and enzyme killing is not needed). Filtering to remove the residue. The pollen extract with high amino acid content is obtained, and the amino acid content is shown in the attached table.
The present invention utilizes the catalytic action of enzyme to convert the protein in pollen into several amino acids easy to be absorbed by human body. The nutrient components of pollen are not lost in the process of enzyme catalysis reaction, so that the extraction method has high yield. The enzyme reaction speed is high (for this fermentation method), so the production cycle is short, generally about eight hours. The pollen extract obtained by the invention is easy to filter, can effectively remove the green smell of pollen, does not have the phenomenon of protein coagulation caused by heating of the pollen extract, and can be directly used for preparing beverages. The beverage can retain the original fragrance and flavor of pollen and other nutrients.
Example one
1. Adding pollen material into a container with stirring, and soaking in 50 deg.C drinking tap water for 2 hr. The proportion of the added water is 1: 4-5. After fully stirring, adding 40 ℃ drinking tap water until the ratio of pollen to water is 1: 8-10.
2. And adjusting the pH value of the material in the container to be about 6 and the temperature to be 40-50 ℃.
3. Adding the cucurbitacin. The enzyme addition amount is 0.1-0.2% of the amount of the pollen (dry basis).
4. Stirred well for 5 hours.
5. Heating to kill enzyme, heating the container to 100 deg.C, and stopping heating.
6. Filtering with a plate-frame filter while hot.
The nutrient components of the obtained pollen extract are shown in the data of the column of papain in the attached table. Such an extract can be used as: directly preparing pollen beverage, pollen, royal jelly, etc., adsorbing the extracted liquid with adsorbent (powdered sugar or wheat flour, etc.), drying and granulating to obtain solid powdered product, such as pollen extract, etc.
The obtained residue can be used for preparing cake food, etc.
Example 2
The operation method and the process conditions are the same as those of example 1. Only the enzyme species is added, and the papain is changed into the bromelain. The nutrient components of the obtained pollen extract are shown in the data of bromelain column in the attached table.
Example 3
The operation method and the process conditions are the same as those of example 1. Only the enzyme species added was changed from papain to bacterial protease. The nutrient contents of the obtained pollen extract are shown in the data of bacterial protease column in the attached table.
Example 4
The operation method and the process conditions are the same as those of example 1, and only the enzyme type is changed from papain to mixed protease (i.e., mixed enzyme of bromelain and bacterial protease). The nutrient contents of the obtained pollen extract are shown in the column of mixed protease in the attached table.
