Abstract
一种属于中药制备技术领域的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,本发明包括以下步骤:将买麻藤的藤晾干后,干燥,再粉碎成40目颗粒;用10倍于买麻藤藤粉碎物重量的混合溶剂,混合溶剂中的乙醇∶去离子水的体积比为7∶3;加热回流提取,真空浓缩,将混合溶剂提取物浓缩成浸膏;利用超声波,用去离子水溶解膏状物后,等体积乙酸乙酯萃取;将乙酸乙酯萃取物真空浓缩至干后,聚酰胺为固定相,去离子水―乙醇为流动相梯度洗脱;洗脱液真空浓缩,冷冻干燥,得到深褐色粉末状物;用胎牛血清的RPMI1640培养液配成悬液。本发明提取工艺简单,无需特别仪器;用人肿瘤细胞株的体外试验表明:0.063mg/ml的浓度对人肝癌细胞株BEL-7402具有显著的抑制效果。
Description
技术领域
本发明涉及的是一种中药制备技术领域的方法,具体地说,是一种对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法。
背景技术
买麻藤主要分布于我国云南南部,广东等省及东南亚地区,在民间与小叶买麻藤混用,治疗风湿性关节炎,腰肌老损,筋骨酸软,跌打损伤,溃疡出血,支气管炎等疾病。经药理动物实验证实买麻藤乙醇提取物对大鼠蛋清性及酵母性关节炎有明显的抗炎作用,但在抗肿瘤方面并没有文献报道。
经对现有技术文献的检索发现,陈浩、林茂在1999年《中草药》第8期“大叶买麻藤化学成分研究”一文中提到,对大叶买麻藤粗提液采用聚酰胺、硅胶为固定相,水-乙醇、氯仿-甲醇、甲醇-水为流动相进行多次柱层析,得到15个化合物,但用该方法所制备的物质是否具有抗肿瘤效果尚未明确。
发明内容
本发明的目的在于克服现有技术中存在的不足,提供一种对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,使其经过加热回流提取以及柱层析淋洗后,提高其洗脱液对人肝癌细胞株BEL-7402的抑制效果。
本发明是通过以下技术方案实现的,本发明以买麻藤藤为原料,采用溶剂提取法、真空浓缩、溶剂萃取法、冷冻干燥和柱层析工艺方法制备而成。
本发明具体步骤如下:
(1)将买麻藤的藤晾干后,在20-30℃下干燥,再粉碎成40目颗粒;
(2)用10倍于买麻藤藤粉碎物重量的混合溶剂,混合溶剂中的乙醇∶去离子水的体积比为7∶3;
(3)50-90℃下加热回流提取四次,1次2小时,45℃下真空浓缩,将混合溶剂提取物浓缩成浸膏;
(4)利用频率为100KHZ的超声波,用去离子水溶解膏状物后,等体积乙酸乙酯萃取6次;
(5)将乙酸乙酯萃取物45℃下真空浓缩至干后,聚酰胺(14-100目)为固定相,去离子水—乙醇为流动相梯度洗脱;
(6)洗脱液45℃下真空浓缩,-40℃下冷冻干燥,得到深褐色粉末状物;
(7)用5%胎牛血清的RPMI1640培养液配成悬液。
从对买麻藤藤茎进行化学成分研究发现,其中含有异丹叶大黄素、白藜芦醇、胡萝卜苷、硬脂酸、买麻藤戊素、买麻藤醇、β-谷甾醇、买麻藤丙素和异丹叶大黄素-3-O-β-D-葡萄糖苷等成分。其中异丹叶大黄素和白藜芦醇等属于二苯乙烯单体化合物,众所周知,二苯乙烯单体化合物有抗菌、降血脂、降血压、抗氧化、抗自由基以及蛋白激酶抑制作用等活性,此外尚有不同程度的抗炎活性,特别是异丹叶大黄素和白藜芦醇具有很强的白三烯生成抑制和受体作用等活性,这可能是此洗脱液具有抑制人肝癌细胞株BEL-7402活性效果的原因。
本发明以多年生的野生植物买麻藤的藤为原料,利用其再生性优势,可以提供丰富的植物资源;提取工艺简单,无需特别仪器;用人肿瘤细胞株的体外试验表明:0.063mg/ml的浓度对人肝癌细胞株BEL-7402具有显著的抑制效果。
具体实施方式
实施例1
称取20℃干燥后粉碎的买麻藤藤粉200g(40目粒度的细粉),选取无水乙醇∶去离子水=7∶3混合溶剂2000ml,50℃下加热回流提取三次,每次2小时,45℃下真空浓缩回收溶剂,得膏状物80.9g,用去离子水溶解,等体积乙酸乙酯萃取6次,浓缩得10.8g浸膏。取其中的1g物质,以去离子水-乙醇为淋洗液,聚酰胺(14目)进行柱层析得有效组分,浓缩,-40℃冷冻干燥得81.4mg。
体外该组分0.063mg/ml采用磺酰罗丹明B蛋白染色法,对人肝癌细胞株BEL-7402的抑制率为87.0%。
实施例2
称取25℃下干燥后粉碎的买麻藤藤粉200g(40目粒度的细粉),选取无水乙醇∶去离子水=7∶3混合溶剂2000ml,70℃下加热回流提取三次,每次2小时,45℃下真空浓缩回收溶剂,得膏状物80.9g,用去离子水溶解,等体积乙酸乙酯萃取6次,浓缩得10.8g浸膏。取其中的1g物质,以去离子水-乙醇为淋洗液,聚酰胺(100目)进行柱层析得有效组分,浓缩,-40℃冷冻干燥得62.3mg。
体外该组分0.25mg/ml采用磺酰罗丹明B蛋白染色法,对人肝癌细胞株BEL-7402的抑制率为63.7%。
实施例3
称取30℃干燥后粉碎的买麻藤藤粉200g(40目粒度的细粉),选取无水乙醇∶去离子水=7∶3混合溶剂2000ml,90℃下加热回流提取三次,每次2小时,45℃下真空浓缩回收溶剂,得膏状物80.9g,用去离子水溶解,等体积乙酸乙酯萃取6次,浓缩得10.8g浸膏。取其中的1g物质,以去离子水-乙醇为淋洗液,聚酰胺(40目)进行柱层析得有效组分,浓缩,-40℃冷冻干燥得71.5mg。
体外该组分0.016mg/ml采用磺酰罗丹明B蛋白染色法,对人肝癌细胞株BEL-7402的抑制率为63.1%。
Claims (10)
1.一种对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征在于,包括以下步骤:
(1)将买麻藤的藤晾干后,干燥,再粉碎成40目颗粒;
(2)用10倍于买麻藤藤粉碎物重量的混合溶剂,混合溶剂中的乙醇∶去离子水的体积比为7∶3;
(3)加热回流提取,真空浓缩,将混合溶剂提取物浓缩成浸膏;
(4)利用超声波,用去离子水溶解膏状物后,等体积乙酸乙酯萃取;
(5)将乙酸乙酯萃取物真空浓缩至干后,聚酰胺为固定相,去离子水—乙醇为流动相梯度洗脱;
(6)洗脱液真空浓缩,冷冻干燥,得到深褐色粉末状物;
(7)用胎牛血清的RPMI1640培养液配成悬液。
2.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的步骤(1),其干燥温度为20-30℃。
3.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的加热回流提取,其温度为50-90℃。
4.根据权利要求3所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的加热回流提取,为四次,1次2小时。
5.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的真空浓缩,其温度为45℃。
6.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的聚酰胺为14-100目。
7.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的超声波,其频率为100KHZ。
8.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的萃取,为6次。
9.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的步骤(6),其冷冻干燥温度为-40℃。
10.根据权利要求1所述的对人肝癌细胞株具有抑制效果的买麻藤提取液的制备方法,其特征是,所述的胎牛血清的RPMI1640培养液,其浓度为5%。
