Abstract
本发明公开了一种以胡芦巴种子为原料提取富含4-羟基-异亮氨酸的天然氨基酸混合物的方法。该方法为取经粉碎后的胡芦巴种子粉,以甲醇为提取剂,在室温下,采用超声波提取,滤渣另行处理;合并提取液,减压浓缩回收甲醇。浓缩液添加适量无离子水,用溶剂汽油萃取三次,以脱除提取液中胡芦巴种子的油脂。水层用酸调pH=1-6,使蛋白变性,滤液上阳离子交换树脂柱,并用无离子水洗涤,流出液和洗涤液合并另行处理;树脂柱用0.05N~2N氨水解吸,脱氨、浓缩至粘稠膏体状,冷冻干燥,得以4-羟基-异亮氨酸为主的天然氨基酸混合物;该提取方法具有明显的经济效益和社会效益。

技术领域
本发明涉及一种植物提取方法,特别是涉及一种从胡芦巴种子中提取4-羟基-异亮氨酸及胡芦巴胶等多种副产品的方法。
背景技术
胡芦巴原产地中海地区,自汉代传入我国,自南到北均有栽培,资源丰富。其嫩茎、叶可作香料佐料,种子药食两用。胡芦巴成熟种子入药,是我国传统中药之一,载入药典。《本草纲目》记载:“胡芦巴苦温纯阳入肾补命门……”,具有补肾阳,祛寒湿作用。主治寒疝、腹胁胀满、寒湿脚气、肾虚腰痛、阳痿遗精、腹泻,无毒副作用。国外亦用其作为治疗糖尿病、高血压病、肾结石、糙皮病、肺病等。
胡芦巴种子中化学组成分主要有脂肪类、聚多糖类、三萜类、甾类、黄酮类、生物碱类、蛋白质和游离氨基酸。这些化合物因其结构特征不同而呈现出不同的生物活性和药理功效。
近年来发现并公认了胡芦巴种子粉有较好的降血糖效果,引起国际上医药界许多专家的兴趣和关注,但对其降糖活性组分的报导较为混乱,其中有胡芦巴肽酯(Fenugreekine)、水溶性食用纤维(即胡芦巴胶)、胡芦巴皂苷、烟酸、香豆素(在地上部分)等等。1992年Yres Sauvaire等报导了从胡芦巴种子中提取、分离出来特有天然的4-羟基-异亮氨酸,经动物实验证明其具有明显的刺激胰腺分泌胰岛素的功能,因此,呈现出显著的降低血糖水平和提高耐葡萄糖量的效果。国内对胡芦巴种子的应用,除了药用之外,已报道了从胡芦巴种子中分离出胚乳并用以生产工业级或食品级增稠剂和从胡芦巴种子(或脱除胚乳后种子粉)中提取薯蓣皂甙的二种综合利用产物。至今,在国内尚未见到从胡芦巴种子中提取天然游离氨基酸及其特有的4-羟基-异亮氨酸的研究报道。同时,无论在国内或国外,均未见到从胡芦巴种子中提取天然的4-羟基-异亮氨酸工业化生产的报道,也未见到在一条生产线上同时提取、制造以4-羟基-异亮氨酸为主的胡芦巴籽游离氨基酸、胡芦巴胶、胡芦巴籽毛油、胡芦巴黄酮及皂素五种产品的综合利用技术与工艺的报导。
胡芦巴胶(Fenugreek Gum),又称胡芦巴可溶性食用纤维、香豆胶,即胡芦巴种子的胚乳部分,其中60%为半乳甘露聚糖(Galactomannan),总糖含量占80%以上,蛋白质占6%左右,其余为水、粗脂肪、粗纤维、灰份等等。曾有人推猜它是降血糖的主要组分,Ali L.在1995年报导了他的研究结果,证明了这种可溶性食用纤维对正常大鼠和非胰岛素依赖型糖尿病大鼠无降糖活性,但同时喂饲葡萄糖时血糖明显降低,仅能说明它具有提高耐葡萄糖量的效果。胡芦巴胶水溶液可抑制旋涡的生成与发展,显示出减阻作用,这些特性决定了它的用途。
当前胡芦巴胶的主要产品与用途有以下二种:工业级胡芦巴胶,可用于石油开采、日用化工、印染浆纱、配制炸药等用途;食品药用级胡芦巴胶:可制得食品、医药级胡芦巴胶增稠剂、赋形剂。
胡芦巴毛油(Oil of Common Fenugreek seed),胡芦巴种子含油量仅为7%左右,属于低油种子,若作为油料作物处理,没有经济效益,但通过综合利用途径提取胡芦巴籽油,可大幅度降低成本,增加经济效益。有关文献报导胡芦巴毛油有含催乳成分,但无任何性激素样作用。毛油及其非皂苷成分具有抗菌作用,其中含有羽扇豆醇具有显著的降血糖功能。毛油经精制,可从油脚、皂脚中分离、提取出磷酯、VE、β-谷甾醇等高附加值的营养保健品。精制的胡芦巴油富含不饱和脂肪酸(高于80%),其中亚油酸和亚麻酸总量达50%以上。不饱和脂肪酸具有降血脂、抑制血小板聚集、防止动脉硬化、抗炎、抗癌、增强免疫功能等生理作用,故可作为保健油。
以天然4-羟基-异亮氨酸(4-hydroxyisoleucine)为主的天然游离氨基酸,4-羟基-异亮氨酸是一种非蛋白组分,极性非电荷(即中性)的新型氨基酸,学名是2-氨基-3-甲基-4-羟基-戊酸,在哺乳动物细胞中没有发现,仅存在于某些(胡芦巴种属)植物中。1973年L Fowdent等首先从胡芦巴种子中分离并鉴定出4-羟基-异亮氨酸,并提出它有三个手性碳原子,有4种异构体:(2S,3R,4R)-,(2R,3R,4R)-,及它们所形成的二种内酯盐酸盐,这种开链和内酯构型可以互相转化。4-羟基-异亮氨酸占干燥种子的游离氨基酸总量的30-50%。而在1984年Y.Sauvaire等的研究,也证明了这个4-羟基-异亮氨酸分子中有三个手性中心,存在于胡芦巴种子中这种氨基酸有二个非对映的异构体形式,主要的一种以(2S,3R,4S)-构型占这种氨基酸含量的90%左右。而次要的一种是以(2R,3R,4S)-构型,占10%左右,这个结果被Hasan(1986),Alcock等(1989)所确证,其它的4-羟基-异亮氨酸立体构型在天然植物中似乎是难以得到的。1992年Y.Sauvaore等研究发现了从胡芦巴种子中提取,分离出的天然4-羟基-异亮氨酸有明显的刺激胰腺分泌胰岛素的功能,呈现出显著的降血糖和提高耐糖量的效果。Masiell等(1998)、C.Broca等(1999,2000)都验证了这个结果,并提出了在4-羟基-异亮氨酸的三个异构体中(2S,3R,4S)-构型刺激胰腺分泌胰岛素效果显著,而(2R,3R,4S)-构型及其内脂亦有明显的效果。但分泌量低于前者。因此,可以认为4-羟基-异亮氨酸是一种具有潜在的,有益于II型糖尿病患者的抗糖尿病新制剂(药剂)。
由于在某种环境条件作用下,(2S,3R,4S)-构型会转化为(2R,3R,4S)-构型或内酯构型,导致相应地削弱了该产品的降血糖和提高耐糖量的功效,在产品的制备过程避免和减少上述转化的发生是关键。
胡芦巴总黄酮(Total Flavones of Fenugreek seed),有文献报导胡芦巴总皂苷、黄酮苷能促进核酸及蛋白质合成及胆固醇酯质代谢,抗动脉硬化,降低胆固醇等功效,;胡芦巴中黄酮类化合物成分母核多为4’,5,7-三羟基黄酮,如牡荆素,牡荆素-7-葡萄糖苷,异牡荆素,荭草素,异荭草素,荭草素苷,异荭草素苷,芹菜素-6-木糖苷-8-葡萄糖苷,芹菜素-6,8-二碳-葡萄糖苷,牡荆素-2”-O-对羟基苯基本柚皮素,木樨草素,小麦黄素,小麦黄素-7-O-葡萄糖苷,6,8-二碳-β-D-呋喃葡萄糖苷,金合欢素等十五种黄酮,及二种黄酮醇:槲皮素,莰非醇。上述17种黄酮类化合物中十三种是黄酮苷类化合物,其中碳-苷十二种,氧-苷一种,这是胡芦巴黄酮的特点,但至今尚未见到胡芦巴总黄酮、黄酮苷的提取和应用的报导。
胡芦巴皂苷(Fenugreek Saponin,Trigosaponin),从胡芦巴种子中已提取鉴定出34种皂苷,其中属于甾体皂苷有30种,分别为薯芋皂苷元(Diosgenin)、雅莫皂苷元(Yamogenin)、支脱皂苷元(Gitogenin)、新支脱皂苷元(Neogiogenin)、替告皂苷元(Tigogenin)、新替告皂苷元(Neotigogenin)、丝兰皂苷元(Yuccagenin)等的不同糖苷化合物,三萜皂苷有4种,分别为胡芦巴皂苷I(Trigosaponin I)、大豆皂苷I(SayasaponinI)、大豆皂苷I甲酯(Methylsayasaponin I)、β-谷甾醇吡喃葡萄糖苷。其中具有薯芋皂苷元母体结构的皂苷,除薯芋皂苷外,胡芦巴皂苷(Gzaecunin)H、I、J、K、L、M、N和胡芦巴素B(Fenugrin B)其苷元都是薯芋皂苷元。这类皂苷是胡芦巴皂苷中主要组分。已有文献报导胡芦巴皂苷有降血糖、降血脂、降低胆固醇、利尿、抗炎等活性,其中薯芋皂苷、雅莫皂苷元有抗真菌、杀虫作用,薯芋皂苷元有雌激素作用和降胆固醇作用,大豆皂苷有抗氧化、抗癌、抗肝损伤、抗血栓及增强免疫力作用,但大多皂苷化合物都有溶血性缺陷。由于胡芦巴皂苷水解成苷元后,其中主要是薯芋皂苷元,可以作为甾体激素,医药工业的原料,目前还没有全合成的原料能与其竞争或代用。国际市场上甾体药物总销售额以每年6-10%的速度递增,进入九十年代以来年产值已突破100亿美元,年皂苷元用量(不包括中国)已超过2000吨,市场呈现原料紧张,求过于供的现状。原来薯芋皂苷元生产的主要原料是盾叶薯芋、穿龙薯芋,而薯芋生长周期要4-7年才挖掘,造成资源短缺,而我国年需薯芋皂苷元(包括出口)1500吨左右,现有50余家工厂生产总量不足1000吨,缺口甚大,因此从胡芦巴籽中提取皂苷,经水解精制,制造薯芋皂苷元,可以弥补市场供应不足之问题。
技术内容
本发明目的在于提供一种以中药胡芦巴(TrigonellaFoenum-graecum.L.,Common Fenugreek)的种子为原料,提取天然4-羟基-异亮氨酸为主体的游离氨基酸产品的方法;
本发明目的还在于提供一种提取天然4-羟基-异亮氨酸的同时可获得胡芦巴胶、胡芦巴籽毛油、胡芦巴黄酮与皂苷等多种副产品的生物化工技术。
本发明目的是通过如下技术方案实现的:
1、胡芦巴种子中五种提取物的提取:
制备胡芦巴籽毛油:取经粉碎机粉碎后的胡芦巴种子粉,60-80℃恒温恒压下石油醚提取1-3小时,抽滤上述提取物,滤渣分别用石油醚再提取二次,合并三次提取液,滤渣A另行处理;提取液浓缩回收溶剂,得到胡芦巴籽毛油;
制备胡芦巴粗黄酮:滤渣A在60-80℃恒温恒压下用10-90%乙醇提取2-3次,每次1-3小时,合并提取液,滤渣B另行处理;提取液通过超滤去除大分子蛋白,收集透过2万分子量膜的滤液;回收乙醇,浓缩,浓缩液加无离子水稀释,即残液A;残液A用液—液萃取法分别用乙酸乙酯萃取三次,水层经减压蒸馏至无乙酸乙酯残留止,作为残留液B;合并乙酸乙酯层,浓缩,回收乙酸乙酯,釜残加纯水至溶解,浓缩,干燥,得到黄色粉末即为胡芦巴粗黄酮;
制备胡芦巴粗皂苷:用液—液萃取法,用正丁醇作为提取剂从残液B中提取三次,水层作为残留液c备用,合并醇层,减压浓缩至干,回收正丁醇得到褐色粗皂苷固体粉末;
制备4-羟基-异亮氨酸:残液c调节PH为1~6,上阳离子交换树脂柱,进行离子交换,并用无离子水洗涤,树脂柱用0.05N~2N氨水解吸,脱氨,浓缩至干后,真空干燥4-8hr,得到黄色或黄褐色固体,产物中4-羟基-异亮氨酸的含量为10-60%;
制备胡芦巴胶:取滤渣B,加10-18倍水,加热,搅拌提取1-3h,抽滤,滤渣弃去,滤液浓缩至1/3体积后,加80-95%乙醇沉淀、洗涤、脱水、干燥得到淡黄至白色的药用或食用胡芦巴胶粉末。
本发明上述方法中石油醚可以用正己烷、环己烷或溶剂汽油代替。
本发明上述方法胡芦巴黄酮制备中滤渣A恒温恒压下乙醇提取可用甲醇提取代替。
本发明上述方法乙醇提取液用超滤方法除去蛋白质还可用矿物酸(如硫酸、盐酸)、有机酸〔如鞣酸、三氯醋酸、乳酸〕或氯仿、聚乙二醇等其他有机物沉降蛋白质来代替。
本发明上述方法胡芦巴黄酮制备中残液A,可用聚酰胺树脂吸附,无离子水洗涤,合并流出液、洗涤液为残液B,另行处理;树脂柱用10~90%乙醇解吸,收集解析液,浓缩回收乙醇,浓缩至干,真空干燥得到黄褐色粗黄酮;
本发明上述方法胡芦巴粗皂苷制备还可以是残留液B用大孔径树脂吸附,无离子水洗涤,树脂柱用10~90%乙醇解吸,收集解吸液浓缩至干,得到黄褐色粗皂苷固体粉末。
本发明上述方法胡芦巴粗皂苷制备还可以是残液B加硫酸或盐酸直接加热回流5~8hr水解糖苷,使水不溶性的皂苷元呈晶态析出,过滤,滤饼烘干后,加适量活性碳混匀,用石油醚连续回流提取8~10hr,提取液经浓缩回收溶剂,浓缩至适当的浓度后,室温放置使结晶完全析出后,过滤,滤液回收使用,滤饼抽干后呈黄色晶体,经乙醇或丙酮溶液重结晶,呈白色皂苷元晶状物;
本发明上述方法工业胡芦巴胶的制备也可以是胡芦巴种子经浸水软化后,破碎、筛分出来的胚乳,经去杂、磨粉、过筛可制得工业胡芦巴胶;
本发明上述方法原料可以是筛分出胚乳后的胡芦巴种子粉。
2、本发明4-羟基-异亮氨酸的提取方法,该方法为:取经粉碎机粉碎后的胡芦巴种子粉,60-80℃恒温恒压下石油醚提取1-3小时,抽滤上述提取物,滤渣分别用石油醚再提取二次,合并三次提取液,滤渣A在60-80℃恒温恒压下用10-90%乙醇提取2-3次,每次1-3小时,合并提取液,滤渣B另行处理;提取液通过超滤,收集透过2万分子量膜的滤液;,回收乙醇,浓缩,浓缩液加无离子水稀释,即残液A;残液A调节PH为1~6,上阳离子交换树脂柱,进行离子交换,并用无离子水洗涤,流出液和洗涤液合并为残液B另行处理。树脂柱用0.05N~2N氨水解吸,脱氨,浓缩至干后,真空干燥4-8hr,得到黄色或黄褐色固体,产物中4-羟基-异亮氨酸的含量为10-60%;
本发明上述方法还可以是:残液A调节PH为1~6,上阳离子交换树脂柱,进行离子交换,并用无离子水洗涤后,以0.05~2N氨水解吸,用茚三酮试剂检测解吸液,当出现氨基酸呈色反应时,即开始收集解吸液至流出解吸液呈微黄色止,合并解析液,蒸发浓缩,脱氨,浓缩至干后,真空干燥4-8hr,得到黄色或黄褐色固体,产物中4-羟基-异亮氨酸的含量为10-60%。
本发明上述方法还可以是:残液A分别用乙酸乙酯萃取三次,水层分别用正丁醇提取三次,在正丁醇提取后的水层B,用盐酸调PH=5,上阳离子树脂柱上进行离子交换,加3-4倍柱体积的无离子水洗涤,弃去流出液及洗涤液,用1N氨水解吸,用茚三酮试剂检测,解吸液呈黄褐色后开始收集,至解吸液呈淡黄色止,合并解吸液,蒸发浓缩、脱氨,浓缩至干后干燥4-8h,得到黄色粉末状物质,其中4-羟基-异亮氨酸的含量为14.47%。
本发明所述阳离子交换树脂还可以是强酸性、中酸性或弱酸性阳离子树脂;干燥可用喷雾干燥、真空干燥、冷冻干燥。
3、本发明还提供了一种胡芦巴种子中四种提取物的提取方法,该方法为:
制备胡芦巴毛油:取经粉碎脱胚乳后的胡芦巴粉在60~80℃恒温恒压下,用10~90%乙醇提取三次,每次1-3hr,合并滤液;滤液经浓缩回收乙醇,浓缩液用无离子水稀释,用用正己烷进行液—液萃取,萃取三次,合并油层,水层另行处理;油层经浓缩回收溶剂,得到胡芦巴毛油;
制备4-羟基-异亮氨酸:水层用稀醋酸调PH4-6,使蛋白变性析出,经沉降过滤,滤液用阳离子交换树脂吸附氨基酸,并用无离子水洗涤,合并流出液、洗涤液为残液d,另行处理;阳离子制树脂柱用0.05~2N氨水解吸,解析液经减压浓缩、脱氨、蒸干,并在80℃下真空干燥4-8hr,得到黄色固体粉末产品,产物中4-羟基-异亮氨酸含量为10-60%;
制备胡芦巴黄酮:残液d通过聚酰胺树脂吸附,并用无离子水洗涤,合并流出液和洗涤液为残液e,另行处理,聚酰胺柱用10-90%乙醇解吸,收集解析液,经浓缩回收乙醇,蒸干,在80℃真空干燥4-8hr,得到黄色固体粉末胡芦巴黄酮;
制备胡芦巴皂苷:残液e通过大孔径树脂柱吸附,并用无离子水洗涤,流出液、洗涤液排入污水处理池,大孔径树脂柱用10-90%乙醇解吸,收集解析液,经浓缩,回收乙醇,蒸干,在80℃真空干燥4-8hr,得到黄褐色固体粉末胡芦巴皂苷。
本发明上述方法中制备胡芦巴毛油还可以是取经粉碎脱胚乳后的胡芦巴粉以乙醇为提取剂用20KHz超声波提取2-3次,每次30-50min,合并滤液;滤液经浓缩回收乙醇,浓缩液用无离子水稀释,用正己烷进行液—液萃取,萃取三次,合并油层,水层另行处理;油层经浓缩回收溶剂,得到胡芦巴毛油。
本发明上述方法中为制备胡芦巴毛油还可以是其中乙醇提取液的浓缩液加无离子水稀释后,还可用矿物酸(如硫酸、盐酸)、有机酸〔如鞣酸、三氯醋酸、乳酸〕或氯仿、聚乙二醇等其他有机物沉降蛋白质,过滤,滤液再用非极性溶剂(石油醚,正己烷,环己烷,溶剂汽油等)萃取胡芦巴籽油。
本发明上述方法中制备胡芦巴黄酮还可以是残液d用乙酸乙酯萃取,合并酯层,经浓缩,干燥,得到黄色固体粉末——胡芦巴黄酮。
本发明所述方法中,阳离子交换树脂为强酸性、中酸性、弱酸性阳离子树脂均可;
本发明所述方法中干燥方法,可用喷雾干燥、真空干燥、冷冻干燥等方法;
本发明所指的4-羟基-异亮氨酸包括以下几个异构体:
(2S,3R,4S)-4-Hydroxylisoleucine,(2R,3R,4S)-4-Hydroxylisoleucine,(2S,3R,4S)-4-Hydroxylisoleucine lactone Hcl,(2R,3R,4S)-4-Hydroxylisoleucine lactone Hcl。
本发明所指的4-羟基-异亮氨酸可加入常规辅料是碳酸氢钙、硫酸钠、甘露糖、聚半乳糖、微粉硅胶、淀粉及其衍生物、羟乙基纤维素、羟丙基纤维素等物质。
本发明方法优点在于在一条生产线上可同时获得天然4-羟基-异亮氨酸、胡芦巴胶、胡芦巴毛油、粗黄酮、粗皂苷等五类产品,这一综合利用生物化工技术填补了国内空白,具有很好的应用前景。目前胡芦巴籽主要利用以提取胡芦巴胶和薯蓣皂苷元为主的胡芦巴皂苷元。以100kg胡芦巴籽进行综合利用为例,可提取约20kg工业胡芦巴胶(20元/公斤),约1kg皂苷元(500元/公斤),6kg胡芦巴种子油(5元/公斤),合计930元产值,按本专利工艺进行综合利用,可制得约3kg含10%4-羟基-异亮氨酸为主的天然混合氨基酸产品,以300元/公斤计,3kg产值为900元,及制得1.5kg胡芦巴黄酮,为胡芦巴黄酮的药效研究和利用提供一定的基础。从上可知本专利技术可增加附加值约一倍左右,其经济效益十分显著。
1992年Y.Sauvaore等研究发现了从胡芦巴种子中提取,分离出的天然4-羟基-异亮氨酸有明显的刺激胰腺分泌胰岛素的功能,呈现出显著的降血糖和提高耐糖量的效果。Masiell等(1998)、C.Broca等(1999,2000)都验证了这个结果。因此,4-羟基-异亮氨酸是一种具有潜在的有益于II型糖尿病患者的抗糖尿的新的辅助药品和预防II型糖尿病患者的发生与扩大的良好保健食品,其社会效益是明显的。
实施例1.
取200g经粉碎机粉碎后的胡芦巴种子粉,温度在75±2℃,800ml石油醚溶媒提取2小时后,抽滤上述提取物及物料,抽毕,将固体物料在75±2℃分别用600ml石油醚再提取二次,合并三次提取液,滤渣A另行处理;提取液进行蒸发浓缩回收溶剂,最后减压浓缩脱除残留溶剂,得到12.53g胡芦巴籽毛油,得率为6.265%;
制备胡芦巴粗黄酮:将滤渣A在75±2℃下,用800ml、600ml、600ml 80%乙醇提取三次,每次2h,滤渣B另行处理,合并提取液,通过微滤、10万分子量膜、5万分子量膜、2万分子量膜,收集透过2万分子量膜的滤液,浓缩,回收乙醇,浓缩液加400ml无离子水稀释,即残液A;残液A分别用300ml乙酸乙酯萃取三次,水层经减压蒸馏至无乙酸乙酯残留止,作为残留液B;合并乙酸乙酯层,进行蒸馏浓缩,回收乙酸乙酯,釜残加适量纯水溶解,浓缩,脱除残留乙酸乙酯,而后蒸干,80℃下干燥,得到3.02g黄色粉末,得率1.51%;该物质即为胡芦巴黄酮;
制备胡芦巴粗皂苷:残液B分别用300ml正丁醇提取三次,水层作为残留液c备用;合并正丁醇层,浓缩回收正丁醇溶剂,釜残即为黄褐色粗皂苷,重量10.26g,得率为5.13%;
制备4-羟基-异亮氨酸:残留液c用盐酸调PH=5,然后用装有300ml已再生处理好并交换成H+型的阳离子树脂柱上进行离子交换,而后加3-4倍柱体积的无离子水进行洗涤,流出液及洗涤液弃之,洗涤后用1N氨水进行解吸,用茚三酮试剂检测,当解吸液呈黄褐色后开始收集,至解吸液呈淡黄色止,合并解吸液,蒸发浓缩、脱氨,浓缩至干后,在80℃下干燥6h,得到黄色粉末状物质4.32g,得率为2.16%,其中4-羟基-异亮氨酸的含量为14.47%;
制备胡芦巴胶:取滤渣B,加15倍的水,加热,搅拌提取3h,抽滤,滤渣弃去,滤液浓缩至1/3体积后,加95%乙醇进行沉淀、过滤、洗涤、风干后,80℃下干燥6h,得到胡芦巴胶淡黄色粉末27.64g,得率为13.32%。
实施例2:
取200g经粉碎筛分去胚乳的胡芦巴种子粉,在75±2℃恒温恒压下,用800ml,600ml,600ml的80%乙醇提取三次,每次2hr,合并提取液另行处理,提取液进行浓缩,回收至浓缩液无醇味止,浓缩液加无离子水600ml稀释,稀释液各用200ml正己烷提取三次,水层另行处理,合并油层进行浓缩,回收正己烷,最后用减压蒸馏脱除残留正己烷,得12.02g粗胡芦巴毛油,得率为6.01%;
制备4-羟基-异亮氨酸:水层用稀醋酸调PH=5,则有大量蛋白质沉淀析出,沉降,过滤,滤液用300gH+型阳离子交换树脂柱进行离子交换,并用600ml无离子水洗涤,合并流出液,洗涤液为残液d另行处理;阳离子树脂柱用0.6N氨水解吸,解吸液经减压浓缩,脱氨,蒸干,并在80℃下真空干燥6hr,得到黄色固体粉末5.44g,得率为1.61%,产品中4-羟基-异亮氨酸含量为30.4%;
制备胡芦巴黄酮:残液d用450g聚酰胺树脂吸附,用600ml无离子水洗涤,合并流出液、洗涤液为残液e,另行处理,聚酰胺树脂柱用80%乙醇进行解吸至流出液为淡黄色为止,合并解析液,经浓缩至干,回收乙醇,釜残为黄色固体,80℃真空干燥4hr,经研磨,得2.92g黄色的粗黄酮粉末,得率为1.46%;
制备胡芦巴皂苷:残液e用500gD-X型大孔径树脂柱吸附,并用800ml无离子水洗涤,大孔径树脂柱用85%乙醇进行解吸,至流出液为淡黄色为止,合并解析液,经浓缩至干,回收乙醇,釜残为褐色固体,80℃真空干燥4hr,经研磨得11.72g粗皂苷,得率为5.68%
Claims (4)
1、一种富含4-羟基-异亮氨酸的天然氨基酸混合物提取方法,其特征在于该方法为:取经粉碎机粉碎后的胡芦巴种子粉,60-80℃恒温恒压下石油醚提取1-3小时,抽滤上述提取物,滤渣分别用石油醚再提取二次,合并三次提取液,滤渣A在60-80℃恒温恒压下用10-90%乙醇提取2-3次,每次1-3小时,合并提取液,滤渣B另行处理;提取液通过超滤,收集透过2万分子量膜的滤液;回收乙醇,浓缩,浓缩液加无离子水稀释,即残液A;残液A调节PH为1~6,上阳离子交换树脂柱,进行离子交换,并用无离子水洗涤,流出液和洗涤液合并为残液B另行处理;树脂柱用0.05N~2N氨水解吸,脱氨,浓缩至干后,真空干燥4-8hr,得到黄色或黄褐色固体,产物中4-羟基-异亮氨酸的含量为10-60%w/w。
2、如权利要求1所述的富含4-羟基-异亮氨酸的天然氨基酸混合物提取方法,其特征在于该方法是:残液A调节PH为1~6,上阳离子交换树脂柱进行离子交换吸附,用无离子水洗涤后,用0.05~2N解吸并用茚三酮试剂检测解吸液,当出现氨基酸呈色反应时,即开始收集解吸液至流出解吸液呈微黄色止,合并解析液,蒸发浓缩,脱氨,浓缩至干后,真空干燥4-8hr,得到黄色或黄褐色固体,产物中4-羟基-异亮氨酸的含量为10-60%w/w。
3、如权利要求1所述的富含4-羟基-异亮氨酸的天然氨基酸混合物提取方法,其特征在于该方法是:残液A分别用乙酸乙酯萃取三次,水层分别用正丁醇提取三次,在正丁醇提取后的水层B,用盐酸调PH=5,上阳离子树脂柱上进行离子交换,加3-4倍柱体积的无离子水洗涤,弃去流出液及洗涤液,用0.05~2N氨水解吸,用茚三酮试剂检测,解吸液呈黄褐色后开始收集,至解吸液呈淡黄色止,合并解吸液,蒸发浓缩、脱氨,浓缩至干后干燥4-8h,得到黄色粉末状物质,其中4-羟基-异亮氨酸的含量为10~60%w/w。
4、如权利要求1所述的富含4-羟基一异亮氨酸的天然氨基酸混合物提取方法,其特征在于该方法中阳离子交换树脂是指强酸性、中酸性或弱酸性阳离子树脂;干燥是指喷雾干燥、真空干燥或冷冻干燥。
