Disclosure of Invention
A first object of the present invention is to provide a moisturizing composition having excellent moisturizing effect.
A second object of the present invention is to provide a method for preparing a moisturizing composition which is simple and easy to handle.
The third object of the invention is to provide an application of the moisturizing composition, which has wide application prospect.
In order to achieve the above purpose, the technical scheme adopted by the invention is as follows:
A moisturizing composition comprises, by mass, 5-10% of Inonotus obliquus extract, 6-15% of purslane extract, 3-5% of aloe vera extract, 1-3% of radix sophorae flavescentis extract, 7-9% of herba menthae in field, 10-12% of chamomile extract, 15-20% of butanediol, 8-12% of glycerol, 0.5-1% of 1, 2-hexanediol, 0.3-0.5% of p-hydroxyacetophenone, and the balance of water;
the preparation process of the inonotus obliquus extract comprises the following steps:
(1) Inoculating the inonotus obliquus strain into a basic seed culture medium added with sodium caseinate and synephrine for culturing to obtain inonotus obliquus mycelium;
(2) Inoculating the inonotus obliquus mycelium obtained in the step (1) to a fermentation medium added with shikimic acid and sodium copper chlorophyllin for continuous culture to obtain a product;
(3) Crushing the product obtained in the step (2), adding water for reflux extraction to obtain an extracting solution, filtering and concentrating the extracting solution, adding ethanol for precipitation, collecting the precipitate, and drying the precipitate to obtain the inonotus obliquus extract.
Further, the concentration of the sodium caseinate and the synephrine in the basic seed culture medium in the step (1) is 0.08-0.15g/L of sodium caseinate and 1-5mg/L of synephrine, and the nutrition composition of the basic seed culture medium is 15-20g/L of glucose, 1-5g/L of peptone, 0.5.5-1.5 g/L of potassium dihydrogen phosphate and 0.5-1.5g/L of anhydrous magnesium sulfate.
Further, the inoculum size of the inonotus obliquus strain in the step (1) is 8-10% of the total mass of the basic seed culture medium, the culture temperature is 20-30 ℃, and the culture time is 7-15d.
Further, the concentration of shikimic acid and sodium copper chlorophyllin in the step (2) in the fermentation culture medium is 9-15mg/L of shikimic acid and 0.4-1mg/L of sodium copper chlorophyllin, and the nutrition composition of the fermentation culture medium is 15-20g/L of glucose, 2-6g/L of beef extract, -1.5g/L of dipotassium hydrogen phosphate and 0.5-1.5g/L of anhydrous magnesium sulfate and 0.05-0.15g/L of L, mnSO 4·H2 O.
Further, the inoculation amount of the inonotus obliquus mycelium in the step (2) is 8-12% of the total mass of the fermentation medium, the temperature of continuous culture is 20-28 ℃, and the time is 10-14d.
Further, the reflux extraction time in the step (3) is 2-3h.
The preparation method of the moisturizing composition comprises the following steps:

The application of the moisturizing composition in preparing skin care products.
Further, the moisture-keeping composition is 1-5% by mass based on the total mass of the skin care product.
Further, the skin care product comprises the following raw materials, by mass, 0.1-0.3% of carbomer, 4-6% of PEG-40 hydrogenated castor oil, 0.5-1% of behenyl alcohol and the balance of water.
Compared with the prior art, the invention has the following main beneficial effects:
1. In the preparation process of the inonotus obliquus extract, synephrine and sodium caseinate are innovatively added into a basic seed culture medium, and shikimic acid and sodium copper chlorophyllin are added into a fermentation culture medium. The polysaccharide content test result shows that the addition of synephrine and shikimic acid can effectively improve the polysaccharide content in the inonotus obliquus extract, and the polysaccharide content in the inonotus obliquus extract reaches more than 35.56 weight percent.
2. The moisturizing composition provided by the invention is mild and has no stimulation to skin, is safe to use, and has an excellent moisturizing effect. The skin moisture test results show that the skin hydration content change rate reaches more than 68.47% after the skin care product 28d added with the moisturizing composition of the present invention is used.

The technical scheme of the invention is further described below with reference to the specific embodiments. It will be understood by those skilled in the art that the following examples are illustrative of the present invention and are not to be construed as limiting the invention. The specific conditions not specified in the examples were carried out according to conventional conditions or conditions recommended by the manufacturer. The reagents or apparatus used, unless otherwise specified, are all conventional products obtained from commercial sources.
Examples
Example 1
A moisturizing composition comprises, by mass, 8% of inonotus obliquus extract, 12% of purslane extract, 4% of aloe vera extract, 2% of radix sophorae flavescentis extract, 8% of peppermint leaf extract, 11% of chamomile extract, 16% of butanediol, 9% of glycerol, 0.8% of 1, 2-hexanediol, 0.4% of p-hydroxyacetophenone and the balance of water.
The preparation method of the inonotus obliquus extract comprises the following steps:
(1) According to the inoculation amount of the inonotus obliquus strain as 9% of the total mass of a basic seed culture medium, inoculating the inonotus obliquus strain into the basic seed culture medium added with sodium caseinate and synephrine, wherein the concentration of the sodium caseinate and the synephrine in the basic seed culture medium is 0.1g/L of sodium caseinate and 4mg/L of synephrine, and culturing for 12d at 28 ℃ to obtain inonotus obliquus mycelium, and the nutrition of the basic seed culture medium comprises 18g/L of glucose, 4g/L of peptone, g/L of monopotassium phosphate and 1g/L of anhydrous magnesium sulfate.
(2) Inoculating Inonotus obliquus mycelium obtained in the step (1) into a fermentation medium added with shikimic acid and sodium copper chlorophyllin according to the inoculation amount of the Inonotus obliquus mycelium accounting for 10% of the total mass of the fermentation medium, wherein the concentration of the shikimic acid and the sodium copper chlorophyllin in the fermentation medium is 12mg/L shikimic acid and 0.5mg/L sodium copper chlorophyllin, and continuously culturing for 12d at 25 ℃ to obtain a product, and the nutrition composition of the fermentation medium is 18g/L glucose, 5g/L beef extract, g/L dipotassium phosphate and 1g/L, mnSO 4·H2 O0.1 g/L anhydrous magnesium sulfate.
(3) Pulverizing the product obtained in the step (2) to a granularity of 5mm, adding water, extracting under reflux for 2.5h to obtain an extracting solution, filtering and concentrating the extracting solution, adding ethanol to precipitate, collecting the precipitate, and drying the precipitate to obtain the inonotus obliquus extract.
The preparation method of the purslane extract comprises the following steps:
Squeezing fresh herba Portulacae at 45deg.C for 10min to obtain juice and herba Portulacae residue, adding 60% (v/v) ethanol water solution 8 times the weight of herba Portulacae residue to the residue, performing solid-liquid separation to obtain extractive solution A and residue, adding 8 times the weight of water to residue, performing solid-liquid separation to obtain extractive solution B, mixing the juice, extractive solution A and extractive solution B, filtering with 0.45 μm filter membrane to obtain filtrate, concentrating under reduced pressure to remove ethanol and water, and spray drying to obtain herba Portulacae extract.
The aloe vera extract is prepared by the following steps:
Taking mature aloe vera leaf with length not less than 40cm and widest part not less than 6cm, cleaning, removing leaf skin to obtain internal gelatinous mesophyll, stirring, crushing, squeezing juice, centrifuging at 5000rpm for 60min to obtain supernatant, and obtaining aloe vera extract.
The preparation method of the kuh-seng root extract comprises the following steps:
Pulverizing radix Sophorae Flavescentis into radix Sophorae Flavescentis powder, adding 5 times of water into radix Sophorae Flavescentis powder, extracting at 40deg.C for 15 hr, and filtering with 200 mesh filter cloth to obtain first residue and filtrate. Adding 8 times of water into the first filter residue, reflux extracting for 5h, filtering with 200 mesh filter cloth to obtain second filtrate. Mixing the first filtrate and the second filtrate, concentrating, and drying to obtain radix Sophorae Flavescentis extract.
The preparation method of the field mint leaf extract comprises the following steps:
Washing Tian Yebao folium Nelumbinis, pulverizing, adding 2 times of water into herba Menthae powder, reflux extracting for 3 hr, cooling, filtering, concentrating, and drying to obtain herba Menthae extract.
The preparation method of the golden chamomile extract comprises the following steps:
soaking flos Matricariae Chamomillae in 14 times of ethanol water solution for 1 hr, heating and keeping micro-boiling for 1 hr, filtering the extractive solution to obtain filtrate, concentrating the filtrate to relative density of 1.10, and spray drying to obtain flos Matricariae Chamomillae extract.

Example 2
A moisturizing composition comprises, by mass, 10% of inonotus obliquus extract, 15% of purslane extract, 5% of aloe vera extract, 3% of radix sophorae flavescentis extract, 9% of peppermint leaf extract, 12% of chamomile extract, 20% of butanediol, 12% of glycerol, 1, 2-hexanediol, 0.5% of p-hydroxyacetophenone and the balance of water.
The preparation method of the inonotus obliquus extract comprises the following steps:
(1) According to the inoculation amount of the inonotus obliquus strain as 10% of the total mass of a basic seed culture medium, inoculating the inonotus obliquus strain into the basic seed culture medium added with sodium caseinate and synephrine, wherein the concentration of the sodium caseinate and the synephrine in the basic seed culture medium is 0.15g/L of sodium caseinate and 5mg/L of synephrine, and culturing for 7d at 30 ℃ to obtain inonotus obliquus mycelium, and the nutrition composition of the basic seed culture medium is 20g/L of glucose, 5g/L of peptone, g/L of monopotassium phosphate and 1.5g/L of anhydrous magnesium sulfate.
(2) Inoculating Inonotus obliquus mycelium obtained in the step (1) into a fermentation medium added with shikimic acid and sodium copper chlorophyllin according to the inoculation amount of the Inonotus obliquus mycelium being 12% of the total mass of the fermentation medium, wherein the concentration of the shikimic acid and the sodium copper chlorophyllin in the fermentation medium is 15mg/L shikimic acid and 1mg/L sodium copper chlorophyllin, and continuously culturing for 10d at 28 ℃ to obtain a product, and the nutrition composition of the fermentation medium is 20g/L glucose, 6g/L beef extract, 1.5.5 g/L dipotassium phosphate and 1.5g/L, mnSO 4·H2 O0.15 g/L anhydrous magnesium sulfate.
(3) Crushing the product obtained in the step (2) to a granularity of 5mm, adding water, extracting under reflux for 3h to obtain an extracting solution, filtering and concentrating the extracting solution, adding ethanol to precipitate, collecting the precipitate, and drying the precipitate to obtain the inonotus obliquus extract.
The preparation process of purslane extract, aloe vera extract, kuh-seng root extract, peppermint leaf extract and golden chamomile extract is the same as in example 1.
The preparation method of the moisturizing composition comprises the following specific steps:

Example 3
A moisturizing composition comprises, by mass, 5% of inonotus obliquus extract, 6% of purslane extract, 3% of aloe vera extract, 1% of kuh-seng extract, 7% of Tian Yebao lotus leaf extract, 10% of golden chamomile extract, 15% of butanediol, 8% of glycerol, 0.5% of 1, 2-hexanediol, 0.3% of p-hydroxyacetophenone and the balance of water.
The preparation method of the inonotus obliquus extract comprises the following steps:
(1) According to the inoculation amount of the inonotus obliquus strain as 8% of the total mass of a basic seed culture medium, inoculating the inonotus obliquus strain into the basic seed culture medium added with sodium caseinate and synephrine, wherein the concentration of the sodium caseinate and the synephrine in the basic seed culture medium is 0.08g/L of sodium caseinate and 1mg/L of synephrine, and culturing for 15d at 20 ℃ to obtain inonotus obliquus mycelium, and the nutrition composition of the basic seed culture medium is 15g/L of glucose, 1g/L of peptone, g/L of monopotassium phosphate and 0.5g/L of anhydrous magnesium sulfate.
(2) Inoculating Inonotus obliquus mycelium obtained in the step (1) into a fermentation medium added with shikimic acid and sodium copper chlorophyllin according to the inoculation amount of the Inonotus obliquus mycelium being 8% of the total mass of the fermentation medium, wherein the concentration of the shikimic acid and the sodium copper chlorophyllin in the fermentation medium is 9mg/L shikimic acid and 0.4mg/L sodium copper chlorophyllin, and continuously culturing for 14d at 20 ℃ to obtain a product, and the nutrition composition of the fermentation medium is 15g/L glucose, 2g/L beef extract, g/L dipotassium phosphate, 0.5g/L, mnSO 4·H2 O0.05 g/L anhydrous magnesium sulfate.
(3) Crushing the product obtained in the step (2) to a granularity of 5mm, adding water, extracting for 2h under reflux to obtain an extracting solution, filtering and concentrating the extracting solution, adding ethanol to precipitate, collecting the precipitate, and drying the precipitate to obtain the inonotus obliquus extract.
The preparation process of purslane extract, aloe vera extract, kuh-seng root extract, peppermint leaf extract and golden chamomile extract is the same as in example 1.

Test examples
Test example 1
Test example 1 polysaccharide content of Inonotus obliquus extracts prepared in examples 1-3 and comparative examples 1-3 was detected by phenol-sulfuric acid method, and the specific procedure is as follows:
10mg of the Inonotus obliquus extract prepared in examples 1-3 and comparative examples 1-3 was weighed and placed in a 50mL volumetric flask, and then dissolved, diluted and fixed to volume with ultrapure water until the volume was calibrated, and shaken uniformly to obtain an Inonotus obliquus extract sample solution.
2ML of each of the above sample solutions was taken, 2mL of a 5% phenol solution and 10mL of concentrated sulfuric acid were immediately added to each of the sample solutions, and mixed uniformly, and the mixture was subjected to constant temperature water bath at 80℃for 30 minutes, cooled to room temperature, and then absorbance of the samples was measured at 486nm wavelength, and each of the samples was measured three times, and the average was taken, and the polysaccharide content results in the Inonotus obliquus extract were shown in Table 1.

As can be seen from the observation of Table 1, the polysaccharide content in the Inonotus obliquus extract prepared in examples 1-3 was higher, reaching more than 35.56 wt%.
The polysaccharide content of the Inonotus obliquus extract prepared in comparative examples 1-3 was reduced to a different extent than that of the Inonotus obliquus extract prepared in examples 1-3, wherein the polysaccharide content of the Inonotus obliquus extract prepared in comparative example 2 was the lowest, and was only 18.27wt%. In the process of the inonotus obliquus extract, synephrine is not added in the basic seed culture medium of the comparative example 1, shikimic acid is not added in the fermentation culture medium of the comparative example 2, and the culture medium of the comparative example 3 changes the adding time of the synephrine and the shikimic acid. From the above, in the process of the inonotus obliquus extract, the content of polysaccharide in the inonotus obliquus extract can be influenced by changing the usage amount and the adding time of synephrine and shikimic acid.
The effects of adding the moisturizing compositions of examples 1 to 3 and comparative examples 1 to 3 of the present invention will be described below.
The skin care product consists of the following raw materials in percentage by mass:
Carbomer 0.2%, PEG-40 hydrogenated castor oil 5%, behenyl alcohol 0.7%, each of the moisturizing compositions described above 3%, the balance being water.
Test example 2
Skin irritation of skin care products obtained by adding the moisturizing compositions of examples 1 to 3 and comparative examples 1 to 3 was detected by using 2015 "cosmetic safety technical Specification" as a reference standard, and the specific procedure is as follows:
Subjects were selected from 120 subjects aged 18-45 years, and randomized into 6 groups with a 1:1 ratio between men and women.
The detection method comprises the steps of putting 0.02g of skin care products obtained by adding the moisturizing compositions of examples 1-3 and comparative examples 1-3 into spot testers respectively, covering the spot testers added with the test substances on the curved sides of forearms of the testers by using a non-stimulated cloth-based adhesive tape, pressing the non-stimulated cloth-based adhesive tape by using palms to uniformly adhere the non-stimulated cloth-based adhesive tape to the surfaces of the forearms of the testers, continuing for 24 hours, removing the spot testers for 30 minutes, and observing skin reactions after the indentations disappear. If the result is negative, the observation is carried out again 48 hours after the patch test, and the skin irritation detection result after 48 hours is shown in Table 2.

From an examination of Table 2, it is clear that the moisturizing composition provided by the present invention is mild and non-irritating to the skin and safe to use.
Test example 3
The moisturizing effect of the skin care products obtained by adding the moisturizing compositions of examples 1 to 3 and comparative examples 1 to 3 was examined with reference to QB/T4256-2011, evaluation guidelines for moisturizing efficacy of cosmetics, and the specific procedure is as follows:
Subjects were selected from 60 healthy female subjects aged 18-45 years, randomized into 6 groups.
The temperature is 22 ℃ and the humidity is 50% in the tested environment, so that direct sunlight is avoided.
Test method all subjects began the test 30 minutes after entering the test environment. The subjects applied the moisturizing compositions of examples 1-3 and comparative examples 1-3 on their faces once a day, each morning and evening, and the skin moisture content was measured before use and after use of the skin care product 28d, respectively, and each data set is an average of the test results of the panel.
Wherein, skin hydration content change rate (%) = (skin hydration content after use of skin care product 28 d-skin hydration content before use)/skin hydration content before use×100%, and the results are shown in table 3.

The higher the value of the change rate of the skin hydration content, the better the moisturizing effect of the skin care product is indicated. From an examination of Table 3, it is apparent that the composition of the present invention has an excellent moisturizing effect. Specifically, after the skin care product 28d added with the moisturizing composition of the present invention is used, the skin hydration content variation rate reaches more than 68.47%.
The moisturizing effect of the skin care products added with the moisturizing compositions of comparative examples 1-3 was reduced to a different extent than those of examples 1-3. The improvement of the polysaccharide content can promote the moisturizing effect of the skin care product by combining the result of the polysaccharide content in the inonotus obliquus extract.
