Abstract
The invention belongs to the technical field of cosmetics, and particularly relates to a cosmetic composition capable of resisting wrinkle and compacting through mitochondria and application thereof. The cosmetic composition comprises sodium mannose phosphate, a huperzia serrata (GIGARTINA STELLATA) extract and a undaria pinnatifida (Undaria Pinnatifida) extract, wherein the weight ratio of the sodium mannose phosphate is as follows: extract of huperzia serrata (GIGARTINA STELLATA): undaria pinnatifida (Undaria Pinnatifida) extract= (1-50): (1-50): (1-50). Research results show that the cosmetic composition provided by the invention can remove free radicals for aged skin, promote metabolism, promote collagen synthesis, accelerate skin regeneration, improve skin toughness from a cellular energy level, and enable the skin to be tough, smooth, elastic, compact and full, and thin and fade.

Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a cosmetic composition capable of resisting wrinkle and compacting through mitochondria and application thereof. The invention particularly relates to a cosmetic composition with the functions of optimizing skin energy regulation, establishing an autonomous anti-aging barrier of skin, improving youthful texture of skin and improving overall skin state and application thereof.
Background
Skin cells require energy to cope with external environmental pressure and repair skin damage, such as collagen synthesis in skin, DNA repair, regulation of cell proliferation, activation of apoptosis, etc., all of which require energy consumption.
About 95% of the energy required for vital cell activity is produced in the mitochondria within the cell. Mitochondria are used as an energy factory of cells, and nutrient substances are converted into chemical energy Adenosine Triphosphate (ATP) required for driving cell metabolism mainly through three stages of glycolysis, pyruvic acid circulation, tricarboxylic acid circulation and oxidative phosphorylation by aerobic respiration, and meanwhile, the growth and apoptosis of the cells are regulated and controlled.

In a third aspect, the present invention provides the use of a cosmetic composition as described in the first aspect or a cosmetic preparation as described in the second aspect above for the preparation of a cosmetic having anti-wrinkle tightening efficacy.
It is understood that within the scope of the present invention, the above-described technical features of the present invention and technical features specifically described below (e.g., in the examples) may be combined with each other to constitute new or preferred technical solutions. Detailed Description
In order to better understand the present invention, the action mechanism of the cosmetic composition of the present invention by mitochondrial wrinkle resistance and the action of the main active ingredient in the cosmetic composition of the present invention are further explained below.
Mitochondrial production ATP aerobic respiratory link corresponds to component mechanism:
the main energy supply material in the human body is sugar, and the way in which ATP energy is generated by decomposing sugar by the human body: aerobic oxidation and anaerobic oxidation, wherein aerobic oxidation is the primary energy supply.

1. Glycolysis-sodium mannose phosphate
The production of cellular energy requires carbohydrates, and glucose is decomposed in the cytoplasm by glycolysis to produce pyruvic acid and NADH during glycolysis, producing energy ATP. The preparation method of the anti-wrinkle tightening essence cream is as follows:
(1) Sequentially adding cetostearyl alcohol, cetostearyl glucoside, lecithin hydrogenated polyisobutene, jojoba seed oil, squalane and polydimethylsiloxane into an oil phase pot, heating to 80-85 ℃, and stirring for 10min at a constant temperature;
(2) Sequentially adding water, EDTA-2NA, carbomer, glycerol and methyl propylene glycol into a water phase pot, heating to 80-85 ℃, and stirring for 10min under heat preservation;
(3) Then extracting the raw materials of the water phase pot and the oil phase pot into an emulsifying pot, starting homogenizing emulsification, stirring for 10min, then cooling to 60 ℃, adding arginine into the emulsifying pot, stirring for neutralization completely, and continuously cooling to 45 ℃;
(4) Sequentially adding panthenol, phenoxyethanol, abies astrospora (GIGARTINA STELLATA) extract, undaria pinnatifida (Undaria Pinnatifida) extract, sodium mannose phosphate, ethylhexyl glycerol and p-hydroxyacetophenone into an emulsifying pot, and stirring uniformly;

Example 2:

a face cream consists of the following components in percentage by weight: cetostearyl alcohol 2.5%, cetostearyl glucoside 2%, lecithin 1%, hydrogenated polyisobutene 3%, jojoba seed oil 1%, squalane 5%, polydimethylsiloxane 3%, EDTA-2NA 0.05%, sodium mannose phosphate 0.3%, carbomer 0.45%, glycerol 2%, methyl propylene glycol 4%, panthenol 1%, arginine 0.3%, phenoxyethanol 0.25%, ethylhexyl glycerol 0.05%, p-hydroxyacetophenone 0.2%, and the balance water.
The cream was prepared as in example 1, except that the extract of Huperzia serrata (GIGARTINA STELLATA) and the extract of undaria pinnatifida (Undaria Pinnatifida) were not added in the step (4).
Comparative example 2:
a face cream consists of the following components in percentage by weight: 2.5% of cetostearyl alcohol, 2% of cetostearyl glucoside, 1% of lecithin, 3% of hydrogenated polyisobutene, 1% of jojoba seed oil, 5% of squalane, 3% of polydimethylsiloxane, 0.05% of EDTA-2NA, 0.3% of Cochuria starchinense (GIGARTINA STELLATA) extract, 0.45% of carbomer, 2% of glycerol, 4% of methyl propylene glycol, 1% of panthenol, 0.3% of arginine, 0.25% of phenoxyethanol, 0.05% of ethylhexyl glycerol, 0.2% of p-hydroxyacetophenone and the balance of water.
The cream was prepared in the same manner as in example 1, except that sodium mannose phosphate and undaria pinnatifida (Undaria Pinnatifida) extract were not added in the step (4).
Comparative example 3:
a face cream consists of the following components in percentage by weight: 2.5% of cetostearyl alcohol, 2% of cetostearyl glucoside, 1% of lecithin, 3% of hydrogenated polyisobutene, 1% of jojoba seed oil, 5% of squalane, 3% of polydimethylsiloxane, 0.05% of EDTA-2NA, 0.3% of undaria pinnatifida (Undaria Pinnatifida) extract, 0.45% of carbomer, 2% of glycerol, 4% of methyl propylene glycol, 1% of panthenol, 0.3% of arginine, 0.25% of phenoxyethanol, 0.05% of ethylhexyl glycerol, 0.2% of p-hydroxyacetophenone and the balance of water.
The cream was prepared in the same manner as in example 1, except that sodium mannose phosphate and extract of huperzia starring (GIGARTINA STELLATA) were not added in step (4).
Comparative example 4:
a face cream consists of the following components in percentage by weight: 2.5% of cetostearyl alcohol, 2% of cetostearyl glucoside, 1% of lecithin, 3% of hydrogenated polyisobutene, 1% of jojoba seed oil, 5% of squalane, 3% of polydimethylsiloxane, 0.05% of EDTA-2NA, 0.1% of Abies starbuchneri (GIGARTINA STELLATA) extract, 0.1% of undaria pinnatifida (Undaria Pinnatifida) extract, 0.1% of vitronectin, 0.45% of carbomer, 2% of glycerol, 4% of methylpropanediol, 1% of panthenol, 0.3% of arginine, 0.25% of phenoxyethanol, 0.05% of ethylhexyl glycerol, 0.2% of p-hydroxyacetophenone and the balance of water.
The cream was prepared in the same manner as in example 1, except that sodium mannose phosphate in step (4) was replaced with a glassy agent.
Comparative example 5:
a face cream consists of the following components in percentage by weight: 2.5% of cetostearyl alcohol, 2% of cetostearyl glucoside, 1% of lecithin, 3% of hydrogenated polyisobutene, 1% of jojoba seed oil, 5% of squalane, 3% of polydimethylsiloxane, 0.05% of EDTA-2NA, 0.1% of Fucus vesiculosus extract, 0.1% of Undaria pinnatifida (Undaria Pinnatifida) extract, 0.1% of sodium mannose phosphate, 0.45% of carbomer, 2% of glycerol, 4% of methyl propylene glycol, 1% of panthenol, 0.3% of arginine, 0.25% of phenoxyethanol, 0.05% of ethylhexyl glycerol, 0.2% of p-hydroxyacetophenone and the balance of water.
The preparation of the cream was the same as in example 1, except that the extract of the huperzia serrata in step (4) was replaced with the extract of fucus.

As can be seen from the experimental results of Table 2, the creams prepared in examples 1-2 of the present invention were significantly superior to the creams prepared in comparative examples 1-5 and the blank cream base described in comparative example 6 in scavenging free radicals.
Also, as shown in the results of comparative examples 1 to 3 in table 2, when the cream contains only one of three active ingredients of the extract of huperzia starring (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) or sodium mannose phosphate, the effect in scavenging free radicals is remarkably inferior to the combined group of the three, although the amount thereof in the cosmetic composition is the same. The results indicate that the combination of the three active ingredients produces a significant synergistic effect in scavenging free radicals.
It can also be seen from the results of comparative examples 4 and 5 that three components, namely, the extract of astrospora sinensis (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) and sodium mannose phosphate, are special and irreplaceable, and the effect of scavenging free radicals of the corresponding face cream is remarkably reduced after being replaced with other components having similar effects in the cosmetic field.
In addition, as shown in comparative example 6, the conventional cosmetic base for preparing a face cream alone has substantially no effect of scavenging free radicals.
Effect example 3: antioxidant effect of the cosmetic composition of the present invention
1. Evaluation principle
Reactive Oxygen Species (ROS) are oxygen-containing chemically reactive chemicals. Including peroxides, superoxides, hydroxyl radicals, and the like. ROS are formed as natural byproducts of normal metabolism of oxygen and have important roles in cell signaling and homeostasis. However, ROS levels increase dramatically during uv exposure. This can cause serious damage to the cellular structure, which is known as oxidative stress. Oxidative stress occurs when the free radical production is greater than the body scavenging capacity. Menadione can generate active oxygen free radicals, and when the free radical generation amount is larger than the organism scavenging capacity, oxidation stress reaction occurs, and the antioxidation effect of the test sample is evaluated through the fluorescence intensity of the zebra fish yolk sac.
2. Experimental protocol
The tested zebra fish were divided into several groups, including normal control group, model control group, examples 1-2 and comparative examples 1-6. Wherein the normal control group was not treated with any treatment, the model control group and the groups of examples 1-2 and comparative examples 1-6 were all added with an equal amount of menaquinone. Examples 1-2 and comparative examples 1-6 samples prepared in examples 1-2 and comparative examples 1-6 were added at a concentration of menaquinone at the same time. After a period of time, the whole zebra fish is subjected to specific fluorescent staining, and the generation condition of active oxygen free radicals in the yolk sac of the zebra fish is observed.

The results show that the combination of the three active ingredients produces a significant synergistic effect in terms of antioxidation.

As can be seen from fig. 1, the yolk sac fluorescence intensity of example 1 is significantly reduced compared with the control group of the residual model, indicating that the sample has significant antioxidant effect.
Effect example 4: human efficacy testing
1. Purpose of test
Skin tightness test was performed by using an instrument by using 4-week samples continuously from adult female volunteers under normal conditions, and the skin roughness and skin fish tail area ratio were photographed and analyzed to evaluate the anti-wrinkle and tightening effects of the test subjects.
2. Test article
Example 1, method of use: after the face is cleaned and the skin is refreshed, a proper amount of the product is evenly smeared on the face skin, and the product is gently massaged until the product is absorbed. Parameter description: the area ratio of fish tail in the skin selected area; the smaller the ratio, the fewer wrinkles.
And (3) result judgment: the skin fish tail area of the sample side is obviously reduced (P is less than 0.05) before and after the test product is tested, or the skin fish tail area of the sample side is obviously improved (P is less than 0.05) compared with the control side, so that the test object has anti-wrinkle effect.
4.3 skin firmness F4 value:
the elastic tester was used to measure the overall amplitude log curve area of the skin during ten consecutive negative pressure cycles.
Parameter description: reflecting the degree of skin firmness; the smaller the measured value, the tighter the skin.
And (3) result judgment: after the product is tried out, the skin compactness F4 value of the sample side is obviously improved (P is less than 0.05), or the skin compactness F4 value difference of the sample side is obviously improved compared with the control side (P is less than 0.05), which shows that the test object has the compacting effect.
5. Test environment
Environmental requirements: temperature: (21+ -1) deg.C; relative humidity: (50.+ -. 10)%.
6. Test design
The test object is used on one side of the left side surface part and the right side surface part of the test object at random, the test object is not used on the other side, any product cannot be smeared on the test object on the same day, the clean water is uniformly used for cleaning the face, and the test is carried out after the test object is statically sitting in a constant temperature chamber for half an hour. The sample side and control side skin firmness F4 was tested before trial (D0) and after trial 28 days (D28); VISIA-CR photographs were taken and analyzed for sample side and control side skin roughness and fish tail area ratios using IPP image analysis software. Under the test condition, after the product is tested for 28 days, the F4 value of the skin compactness of the side face of the sample is obviously reduced (P is less than 0.05), and the difference value between the front and back is obviously improved compared with the comparison side (P is less than 0.05), so that the test object has the effect of improving the skin compactness.
In conclusion, under the test conditions, the test object has anti-wrinkle and tightening effects.

10. Use of the cosmetic composition according to claim 1 or claim 2 or the cosmetic preparation according to any one of claims 3 to 9 for the preparation of a cosmetic having anti-wrinkle tightening efficacy.
