Abstract
本发明提供一种具有抑菌作用的蚂蚁外用组合物,所述外用组合物的原料由以下重量份数的组分组成:蚂蚁40‑60份、干姜40‑60份、艾叶40‑60份、苦参40‑60份、黄柏20‑40份、蛇床子20‑40份、白鲜皮20‑40份、白芷20‑40份、桂枝20‑40份、蒲公英10‑30份、冰片5‑15份。针对足部主要的致病菌大肠杆菌、金黄色葡萄球菌、白色念珠菌等均有抑制作用,对传统中药需要水煎,本发明提前将所需的中药进行提取浓缩成泡脚液,使用时用热水进行勾兑即可,具有使用方便安全、温和无刺激、疗效稳定,并且具有促进患者创口快速愈合的功效,可有效减小重复感染的概率等特色。

技术领域
本发明涉及中药制剂,尤其涉及一种具有抑菌作用的蚂蚁外用组合物。
背景技术
足浴的历史有数千年,最早的文献记载是晋代《肘后备急方》,至今已有千余年历史。泡脚对很多疾病的治疗,有很好的辅助作用。人们常说一句话:"富人吃补药,穷人泡泡脚"。可见泡脚的作用很大。人们在紧张劳动、运动、工作后,或在旅游出差时,或是天气炎热时,脚部在出汗潮热的情况下,易滋生细菌,导致脚部出现发臭、发痒,甚至脱皮、溃烂的症状,有各种细菌藏于皮肤中导致脚部感染,加之脚部角质层老化硬化需要对脚进行保健和特殊杀菌,但目前一般洗浴用品难以满足上述各项要求。
鉴于此,有必要研发一款既具有清洁脚部皮肤等常规功能,又具有杀菌消毒、防治病菌感染功效的外用组合物,尤其是可以用于脚部的外用组合物。
发明内容
本发明克服现有技术的不足,提供一种具有抑菌作用的蚂蚁外用组合物,针对足部主要的致病菌大肠杆菌、金黄色葡萄球菌、白色念珠菌均有抑制作用,对传统中药需要水煎,本发明提前将所需的中药进行提取浓缩成外用组合物,使用时用热水进行勾兑即可,具有使用方便安全、无副作用、疗效稳定等特色。
本发明的第一个目的是提供一种具有抑菌作用的蚂蚁外用组合物,所述外用组合物的原料由以下重量份数的组分组成:蚂蚁40-60份、干姜40-60份、艾叶40-60份、苦参40-60份、黄柏20-40份、蛇床子20-40份、白鲜皮20-40份、白芷20-40份、桂枝20-40份、蒲公英10-30份、冰片5-15份。
进一步的,所述外用组合物的原料由以下重量份数的组分组成:蚂蚁50份、干姜50份、艾叶50份、苦参50份、黄柏30份、蛇床子30份、白鲜皮30份、白芷30份、桂枝30份、蒲公英20份、冰片10份;或,
蚂蚁40g、干姜50g、艾叶40g、苦参40g、黄柏40g、蛇床子40g、白鲜皮40g、白芷30g、桂枝30g、蒲公英20g、冰片10g;或,
蚂蚁60g、干姜60g、艾叶60g、苦参60g、黄柏30g、蛇床子40g、白鲜皮40g、白芷30g、桂枝30g、蒲公英20g、冰片10g;或,
蚂蚁40g、干姜60g、艾叶50g、苦参60g、黄柏20g、蛇床子20g、白鲜皮30g、白芷30g、桂枝20g、蒲公英30g、冰片10g;或,
蚂蚁50g、干姜40g、艾叶50g、苦参50g、黄柏20g、蛇床子30g、白鲜皮30g、白芷20g、桂枝40g、蒲公英30g、冰片10g。
进一步的,所述外用组合物采用以下方法制备得到:
(1)用60%乙醇浸泡除冰片之外所有药材;
(2)保持微沸,回流提取,过滤;
(3)合并两次滤液,浓缩至密度1.1-1.2g.cm-3(60℃)的稠状液体,加入溶解后的冰片,即为所述外用组合物。
进一步的,步骤(1)所述60%乙醇的用量为除冰片之外所有药材8倍量,60%乙醇浸泡时间为1小时。
进一步的,步骤(2)所述回流提取的次数为2次,每次2h;所述过滤为350目。
本发明的第二个目的是提供一种具有抑菌作用的蚂蚁外用组合物的制备方法,所述方法包括以下步骤:
(1)用60%乙醇浸泡除冰片之外所有药材;
(2)保持微沸,回流提取,过滤;
(3)合并两次滤液,浓缩至密度1.1-1.2g.cm-3(60℃)的稠状液体,加入溶解后的冰片,即为所述外用组合物。
进一步的,步骤(1)所述60%乙醇的用量为除冰片之外所有药材8倍量,60%乙醇浸泡时间为1小时。
进一步的,步骤(2)所述回流提取的次数为2次,每次2h;所述过滤为350目。
本发明的第三个目的是提供前述的蚂蚁外用组合物在制备抑菌泡脚液中的应用。
进一步的,所述抑菌为能够抑制大肠杆菌、金黄色葡萄球菌、白色念珠菌中的一种或多种。
有益效果:
1、本发明所制得蚂蚁外用组合物,通过提取蚂蚁中的有效物质抗菌肽成分,干姜中的有效物质姜辣素、黄酮、多酚,艾叶中的有效成分挥发油、总三萜等,苦参中的有效物质苦参碱,黄柏中的有效物质生物碱,蛇床子中的有效物质总黄酮,白鲜皮中的有效物质白鲜碱等生物碱类成分、β-谷甾醇等甾体类化合物,白芷中的有效物质总黄酮,桂枝中的有效物质桂皮醛等挥发油类成分,蒲公英中的有效物质绿原酸、总黄酮等,使制得的外用组合物含这些活性物质,对脚部细菌真菌感染具有很好的治疗作用,尤其是对常见的金黄色葡萄球菌、大肠杆菌、白色念珠菌等致病菌均具有很好的抑菌效果,且使用的是纯植物的提取物,安全可控。
2、本发明所制得外用组合物抑菌效果好且稳定,抑菌率>90%。组方解析如下:蚂蚁中具有的抗菌肽成分对大肠杆菌、金黄色葡萄球菌等的生长均表现出一定的抑制作用,艾叶性味辛、苦、温,具有温经止血,散寒止痛,外用祛湿止痒,现代药理学研究表明具有止血、抗菌、抗病毒等药理作用。苦参有清热燥湿,杀虫,利尿之功,常与黄柏、蛇床子连用,煎水外洗,治疗皮肤瘙痒达到抑菌止痒的功效。白芷解表散寒,燥湿止带,现代药理学表明其具抑菌等作用。白鲜皮苦燥泄,善清热解毒、燥湿利湿、止痒,治疮疹、疥藓常用。蒲公英清热解毒、利尿通淋、强壮骨骼,现代药理学作用表明其具有良好的抗菌作用。冰片止痛防腐、开窍醒神、清热生肌,外用具有清热、止痒、生肌的作用。桂枝及干姜温通经脉,助阳化气可以增强外用组合物的热感,使外用组合物使用起来更加舒适。
3、从加工工艺角度来说,蚂蚁外用组合物所用的药材是经过60%的乙醇加热回流提取的,较水提取而言可增强药材中的有效成分的溶出,增强疗效,通过多次实验对比,发现用此提取方法,生产效率高,产品质量稳定,适合大规模生产。
4、本发明与现有技术相比,其显著的优点是:通过采用中草药配方,使制得的外用组合物具有疗效显著,不易耐药等多种优势,方便携带,按配比加入热水即可,且具有抑制足部细菌真菌的效果,可有效缓解脚臭脚痒等脚部疾病。并且具有促进患者创口快速愈合的功效,可有效减小重复感染的概率。

试验一组方的用量
蚂蚁泡脚液,包括如下质量份数的组分:
蚂蚁40-60份、干姜40-60份、艾叶40-60份、苦参40-60份、黄柏20-40份、蛇床子20-40份、白鲜皮20-40份、白芷20-40份、桂枝20-40份、蒲公英10-30份、冰片5-15份。
作为优先的,各组分的质量份数的配比如下:
蚂蚁50份、干姜50份、艾叶50份、苦参50份、黄柏30份、蛇床子30份、白鲜皮30份、白芷30份、桂枝30份、蒲公英20份、冰片10份。
上述蚂蚁泡脚液的制备方法,以蚂蚁40-60份、干姜40-60份、艾叶40-60份、苦参40-60份、黄柏20-40份、蛇床子20-40份、白鲜皮20-40份、白芷20-40份、桂枝20-40份、蒲公英10-30份、冰片5-15份的质量分数配比各组分,并对其进行加工,包括:
步骤一、将除冰片之外药材采用8倍量乙醇为提取溶剂,所属乙醇的酒精度为60%,浸泡1小时后,加热回流提取2次,每次2h。
步骤二、将两次加热回流后的药液合并,过350目滤布。
步骤三、将过滤后的药液减压浓缩至密度为1.1-1.2g.cm-3(60℃)的浓缩液。
步骤四、将冰片5-15份加适量的乙醇溶解后,与步骤三中的浓缩液搅拌均匀即可。
实施例1
取蚂蚁50g、干姜50g、艾叶50g、苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、桂枝30g、蒲公英20g、冰片10g。
制备方法:加入除冰片之外药材采用8倍量的60%的乙醇,浸泡一小时后,加热回流提取2次,每次2h,收集两次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
实施例2
蚂蚁40g、干姜50g、艾叶40g、苦参40g、黄柏40g、蛇床子40g、白鲜皮40g、白芷30g、桂枝30g、蒲公英20g、冰片10g。

抑菌检测试验
GB 15979-2002《一次性使用卫生用品卫生标准》附录C规定的方法检,产品杀菌性能、抑菌性能与稳定性测试方法。
1.样品采集
取自制泡脚液实施例1-5中的样品各3份。
2.试验菌与菌液制备
2.1细菌:金黄色葡萄球菌(ATCC 6538),大肠杆菌(ATCC 25922)。
2.2酵母菌:白色念珠菌(ATCC 10231)。
菌液制备:取菌株第3-14代的营养琼脂培养基斜面新鲜培养物(18-24h),用5mL0.03mol/L磷酸盐缓冲液(以下简称PBS)洗下菌苔,使菌悬浮均匀后用上述PBS稀释至所需浓度。
3.操作步骤
将试验菌24h斜面培养物用PBS洗下,制成菌悬液(要求的浓度为:用100μL滴于对照样片上或5mL样液内,回收菌数为1*104-9*104cfu/片或mL)。
取被试样片(2.0cm*3.0cm)或样液(5mL)和对照样片或样液(与试样同质材料,同等大小,但不含抗菌材料,且经灭菌处理)各4片(置于灭菌平皿内)或4管。
取上述菌悬液,分别在每个被试样片或样液和对照样片或样液上或内滴加100μL,均匀涂布/混合。开始计时,作用2、5、10、20min,用无菌镊分别将样片或样液(0.5mL)投入含5mL PBS的试管内,充分混匀,作适当稀释,然后取其中2-3个稀释度,分别吸取0.5mL,置于两个平皿,用凉至40-45℃的营养琼脂培养基(细菌)或沙氏琼脂培养基(酵母菌)15mL作倾注,转动平皿,使其充分均匀,琼脂凝固后翻转平板35℃±2℃培养48h(细菌)或72h(酵母菌),作活菌菌落计数。
按式计算抑菌率:
X=(A-B)/A*100%
式中:X—抑菌率(%);
A—对照样品平均菌落数;
B—被试样品平均菌落数。
评价标准抑菌率≥50%-90%,产品有抑菌作用,抑菌率≥90%,产品有较强抑菌作用。
实验结果

根据实验结果,可以看出,此配方在实施例1~5的各配比时都具有较强的抑菌作用,作用2min既对大肠杆菌、金黄色葡萄球菌、白色念珠菌有明显抑菌效果;作用时间5min后,实施例1~5的配方对大肠杆菌、金黄色葡萄球菌的杀菌率均已大于99%;作用时间10min后,对大肠杆菌、金黄色葡萄球菌、白色念珠菌杀菌率均大于99%。在此5个实施例中,实施例1抑菌效果优于其他实施例,起效最迅速。
试验二组方的选择
除试验一组方的用量中的实施例1至5以外,申请人还筛选了不同组方,具体如对比例1至3:
对比例1
蚂蚁50g、干姜50g、艾叶50g、苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、桂枝30g、蒲公英20g用8倍量的60%乙醇,泡一小时后,加热回流提取2次,每次2h,收集两次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),即得。
对比例2
苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、蒲公英20g用8倍量的60%乙醇,泡一小时后,加热回流提取2次,每次2h,收集两次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将10g冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
对比例3
干姜50g、艾叶50g、桂枝30g用8倍量的60%乙醇,泡一小时后,加热回流提取2次,每次2h,收集两次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将10g冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
一、抑菌检测试验
GB 15979-2002《一次性使用卫生用品卫生标准》附录C规定的方法检,产品杀菌性能、抑菌性能与稳定性测试方法。
取自制泡脚液对比例1至3中的样品各3份,与实验一抑菌检测试验一并操作。
实验结果

根据表1和表2的实验结果,可以看出,实施例1至5的抑菌效果、抑菌率均优于其他实施例,尤其对于白色念珠菌,对比例1无法在在10min以内实现较强抑制,对比例2、3在全过程均无法实现较强抑菌效果。
二、皮肤刺激性试验
根据《消毒技术规范》(2002年版)中皮肤刺激试验的方法,考察实施例1和对比例1至3的皮肤刺激性。
1.样品采集
取自制泡脚液实施例1和对比例1至3中的样品各3份。
2.操作步骤
采用皮肤完好的家兔18只(各实施例以3只家兔进行实验),雌性,体重2.2~2.4kg,以普通级家兔维持饲料喂养,温度20~25℃,相对湿度40%~70%。实验前约24h,将白兔背部脊柱两侧毛剔除,不得损伤皮肤,去毛范围左、右各约2.5cm×2.5cm,取受试物原样0.5ml直接滴于皮肤上,然后用一层玻璃纸覆盖,并用胶布固定,另一侧皮肤涂抹0.5ml蒸馏水做空白对照,敷贴时间为4h,用温水冲洗1min以去除受试物,分别于去除受试物后1h、24h、48h观察皮肤局部反应,并根据表3进行皮肤刺激反应评分。结果见表4。

由表4可知,实施例1对家兔皮肤均无刺激性,对比例1-3对家兔皮肤有刺激性。
试验三提取工艺筛选
除试验一组方的用量中的实施例1至5以外,申请人还筛选了不同组方,具体如对比例4至6:
对比例4
蚂蚁50g、干姜50g、艾叶50g、苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、桂枝30g、蒲公英20g用8倍量的75%乙醇,泡一小时后,加热回流两次,收集2次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将10g冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
对比例5
蚂蚁50g、干姜50g、艾叶50g、苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、桂枝30g、蒲公英20g用8倍量的95%乙醇,泡一小时后,加热回流两次,收集2次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将10g冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
对比例6
蚂蚁50g、干姜50g、艾叶50g、苦参50g、黄柏30g、蛇床子30g、白鲜皮30g、白芷30g、桂枝30g、蒲公英20g用8倍量的水,泡一小时后,加热回流两次,收集2次药液,合并后过滤(350目滤布),滤液浓缩至密度1.1-1.2g.cm-3(60℃),将10g冰片加入适量乙醇使其溶解后与浓缩液搅拌均匀,即得。
一、抑菌检测试验
取自制泡脚液对比例4至6中的样品各3份,与实验一抑菌检测试验一并操作。

根据表1和表5实验结果,可以看出,实施例1至5的抑菌效果、抑菌率均优于其他实施例,尤其对于白色念珠菌,对比例1无法在在20min以内实现较强抑制,对比例2、3在全过程均无法实现较强抑菌效果。
二、创面愈合试验
采用健康的小鼠15只(各实施例以3只小鼠进行实验),雌性,体重为25g左右,实验前适应性饲养一周,在做创伤模型的前一天进行脱毛处理。用动物麻醉机将小鼠麻醉后固定于操作台上,先用推毛剪去除小鼠背部大部分毛发,随后用棉签浸湿并蘸上脱毛膏擦拭剪毛区域,放置几分钟,而后用棉球擦去多余的脱毛膏,并用干净的棉球蘸取0.9%的生理盐水擦拭脱毛处皮肤,擦洗净皮肤上残留的脱毛膏,防止对皮肤产生灼伤。擦拭完后用洁净棉球擦干脱毛处皮肤。用酒精消毒皮肤后,用无菌手术器械造面积为1×1cm的圆形背部全皮层切除伤口,随机分为5组。空白组是在皮肤创面滴加100μL的无菌PBS;4个实施例组是将100μL受试物原样缓慢滴加在伤口上,再用医用无菌透明固定胶带固定伤口边缘。在第1,4,7,14天用异氟烷将小鼠麻醉,计算伤口面积,按如下公式计算创面愈合率。结果见表6。
创面愈合率=(原始创面面积-当前测量面积)/原始创面面积*100%。

根据实验结果,可以看出,本发明泡脚液可促进伤口愈合,且实施例1促愈合效果显著。
三、有效性实验
申请人邀请了患有足部真菌感染的患者,连续使用泡脚液,以10天一个疗程,持续观察患者治愈情况,以无瘙痒感、气味症状,红斑消退无鳞屑,糜烂处软化的角质脱去且皮损处伤口愈合,观察一个月不复发即为愈合;使用泡脚液前后症状无明显好转或症状复发即为无效。
安慰剂组的制备:1克焦糖色色素加入到10mL的水中溶解,兑入2L的热水即得。

实施例1较对比例1-6对丘疹鳞屑型、浸渍糜烂型的足部真菌患者的愈合有较好的作用,一个月内没有复发,基于以上研究发现,该泡脚液可消除患处红斑、瘙痒、糜烂等症状,能够治愈足部真菌感染,治愈率较高,制作简单,疗效显著。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (8)
1.一种具有抑菌作用的蚂蚁外用组合物,其特征在于,所述外用组合物的原料由以下重量份数的组分组成:蚂蚁40-60份、干姜40-60份、艾叶40-60份、苦参40-60份、黄柏20-40份、蛇床子20-40份、白鲜皮20-40份、白芷20-40份、桂枝20-40份、蒲公英10-30份、冰片5-15份。
2.根据权利要求1所述具有抑菌作用的蚂蚁外用组合物,其特征在于,所述外用组合物的原料由以下重量份数的组分组成:
蚂蚁50份、干姜50份、艾叶50份、苦参50份、黄柏30份、蛇床子30份、白鲜皮30份、白芷30份、桂枝30份、蒲公英20份、冰片10份;或,
蚂蚁40g、干姜50g、艾叶40g、苦参40g、黄柏40g、蛇床子40g、白鲜皮40g、白芷30g、桂枝30g、蒲公英20g、冰片10g;或,
蚂蚁60g、干姜60g、艾叶60g、苦参60g、黄柏30g、蛇床子40g、白鲜皮40g、白芷30g、桂枝30g、蒲公英20g、冰片10g;或,
蚂蚁40g、干姜60g、艾叶50g、苦参60g、黄柏20g、蛇床子20g、白鲜皮30g、白芷30g、桂枝20g、蒲公英30g、冰片10g;或,
蚂蚁50g、干姜40g、艾叶50g、苦参50g、黄柏20g、蛇床子30g、白鲜皮30g、白芷20g、桂枝40g、蒲公英30g、冰片10g。
3.根据权利要求1所述具有抑菌作用的蚂蚁外用组合物,其特征在于,所述外用组合物采用以下方法制备得到:
(1)用60%乙醇浸泡除冰片之外所有药材;
(2)保持微沸,回流提取,过滤;
(3)合并两次滤液,浓缩至密度1.1-1.2g.cm-3(60℃)的稠状液体,加入溶解后的冰片,即为所述外用组合物。
4.一种具有抑菌作用的蚂蚁外用组合物的制备方法,其特征在于,所述方法包括以下步骤:
(1)用除冰片之外所有药材8倍量的60%乙醇浸泡1小时;
(2)保持微沸,回流提取2次,每次2h,350目过滤;
(3)合并两次滤液,浓缩至密度1.1-1.2g.cm-3(60℃)的稠状液体,加入溶解后的冰片,即为所述外用组合物。
5.根据权利要求3或4所述的方法,其特征在于,步骤(1)所述60%乙醇的用量为除冰片之外所有药材8倍量,60%乙醇浸泡时间为1小时。
6.根据权利要求3或4所述的方法,其特征在于,步骤(2)所述回流提取的次数为2次,每次2h;所述过滤为350目。
7.权利要求1所述的蚂蚁外用组合物在制备抑菌泡脚液中的应用。
8.根据权利要求7所述的应用,其特征在于,所述抑菌为能够抑制大肠杆菌、金黄色葡萄球菌、白色念珠菌中的一种或多种。
