Description

Milk cow nipple medicated bath liquid and preparation method thereof
Technical Field
The invention relates to the technical field of bath lotion for veterinary use, in particular to a cow nipple bath lotion and a preparation method thereof.
Background
Cow teats are susceptible to inflammation due to long-term milking, and a cow teat medicated bath is an effective method for preventing inflammation of cow mammary gland. The medicinal bath liquid can prevent mastitis of the milk cow, and can quickly act on the milk head of the milk cow when being used as a bactericide before milk is extruded, so that various bacteria can be killed more quickly, and the quality of the milk extruded after the medicinal bath liquid is cleaned can be better.
The liquid medicine for dairy cows generally comprises a front medicine bath liquid and a rear medicine bath liquid. Before milking, the cow teats need to be soaked and sterilized by using the prodrug bath solution, and the prodrug bath solution is rinsed with clean water after the teats stay for 2-5s and then is milked. In the milking process, sterilization of the inside and outside of the nipple is an important link for reducing milk bacteria, so that the pro-drug bath lotion is required to quickly kill various pathogenic bacteria without damaging the skin, and the soaked drug bath lotion is required to be quickly condensed on the nipple of the milk, so that the pro-drug bath lotion is more concentrated on the nipple of the milk, and the concentration of the pro-drug bath lotion is favorable for improving the action effect of the bactericide.
The existing medicated bath liquid generally comprises a sterilization component and a film-forming agent, wherein the sterilization component adopts iodine preparations, quaternary ammonium salts, guanidines and the like. The traditional iodine medicated bath liquid is mostly prepared by mixing other surfactants with film-forming substances such as povidone, polyethylene glycol, saccharide derivatives, hydroxymethyl cellulose, arabic gum and the like. The medicinal bath liquid is generally used after milk is squeezed out, forms a film on the surface of a nipple of a cow, acts on the nipple of the cow for a long time, and protects the nipple for a long time, but the medicinal bath liquid is mainly used as a post-medicinal bath liquid. The prodrug bath solution is generally drained after the cow breasts are soaked for 2 to 5 seconds before milking, the prodrug bath solution forms a wall-hanging state on the surfaces of the breasts, a formed film is uniformly spread on the surfaces of the breasts and nipples and cannot be rapidly condensed on the nipples, and the content of the prodrug bath solution on the nipples is low. The pro-drug bath lotion is generally kept for 1-2min in the breasts, and the breasts are washed with clean water and then milked. Because the prodrug bath solution has short residence time at the nipple and low content at the nipple, the prodrug bath solution can not thoroughly sterilize the inside and outside of the nipple, and the bacterial content of the dairy product is still easy to increase during milking, and the risk of nipple infection is increased.

The cow teat dip solutions of comparative examples 1 to 9 were prepared according to method example 3.
Iodine complexation and stability test
1. Iodine complexation and stability testing
The iodine content of the bath liquids obtained in application examples 1 to 15 and comparative examples 1 to 10 was measured according to the starch identification and iodine content measurement methods of povidone iodine in pharmacopoeia, and the results are shown in table 1:
TABLE 1 iodine content and stability in each bath liquid
Figure SMS_2
As can be seen from the application example 15 and the comparative example 1, the initial content of the comparative example 1 is basically the same as that of the application example 15, and the fact that the isotridecanol polyoxyethylene ether is compounded with the auxiliary surfactant can improve the iodine complexation is demonstrated. By applying example 15 and comparative example 2, it can be seen that the initial content of iodine is low in comparative example 2 without adding an auxiliary surfactant, and the isomeric tridecanol polyoxyethylene ether is relatively weak in stability and is easy to precipitate and separate out due to the fact that no auxiliary surfactant is compounded in comparative example 2. As can be seen from application example 15, comparative example 1, comparative example 2 and comparative example 8, the isotridecanol polyoxyethylene ether in application example 15 of the invention can improve the complexation of iodine by compounding with an auxiliary surfactant.
In comparative examples 3, 4, 5 and 6, the isotridecanol polyoxyethylene ether is free from the problems of precipitation and precipitation, and shows that the isotridecanol polyoxyethylene ether is a main component for iodine complexation and plays a main role in iodine complexation and stability. Due to the fact that the auxiliary surfactant is not compounded, the relative stability of the isomeric tridecanol polyoxyethylene ether is weak. By comparing the application example 15 with the comparative examples 7 and 9, the compound complexing system of the isomeric tridecanol polyoxyethylene ether has obvious complexing stability advantage compared with the compound complexing system of the polyoxyethylene ether; in contrast, application example 15 and comparative example 10 show that application example 15 also has a distinct complexing stability advantage compared to the commercial bath products.
As can be seen from comparative examples 5 and 9, the fatty alcohol-polyoxyethylene ether added in comparative example 5 is less, and therefore, precipitation occurs; the fatty alcohol-polyoxyethylene ether added in the comparative example 9 is more, and the complexation of iodine has certain advantages, but compared with the application example 15, the stability of the fatty alcohol-polyoxyethylene ether to iodine is obviously reduced in a long-term storage process, and the complexation and long-term stability of isomeric tridecanol polyoxyethylene ether to iodine have good effects.
2. Temperature resistance test of medicated bath solution
The bath solutions of application examples 1 to 15 were subjected to high temperature resistance and low temperature resistance stability tests, respectively, wherein the high temperature resistance (120 ℃) was 4 hours and the low temperature resistance (-20 ℃) was 4 hours, and the obtained results are shown in table 2;
TABLE 2 results of temperature resistance test
Figure SMS_3
As can be seen from Table 2, the medicated bath solutions of application examples 1-15 all reached their standards. Moreover, as can be seen from the results of comparative example 2, the medicated bath liquid of the present invention could not be stabilized in a high-temperature or low-temperature state when formulated without the addition of a cosurfactant. As can be seen from comparative examples 3-6, the drug bath solution has no stability at high temperature and low temperature when no isomeric tridecanol polyoxyethylene ether is added, so that the addition of isomeric tridecanol polyoxyethylene ether can obviously improve the temperature resistance of the drug bath solution as can be seen from application examples 1-15 and comparative examples 3-6.
3. Cohesion test
3.1, respectively taking the medicated bath liquids of the application examples 1 to 15 and the comparative examples 1 to 10 as samples, respectively placing the samples in beakers, respectively, vertically inserting the skin simulation finger models into the samples by using the skin simulation finger models, keeping the height of the samples in the beakers to be not less than 50mm, then vertically taking out the skin simulation finger models, observing the dropping condition of the medicated bath liquids, and finding that the medicated bath liquids can be gathered at the tail ends of the fingers within 1min and do not drop. The medicinal bath liquid has the effects of quick coagulation and no dripping when being applied to the cow nipple, can achieve quick and efficient sterilization when being applied to the cow nipple, and can achieve the effect of long-acting protection on the cow nipple.
3.2, taking the medical bath solution in the application examples 1-15 and the comparative examples 1-10 as samples, respectively, taking 2ml of each sample as a sample to be tested, uniformly spreading the sample to be tested on an adhesive glass slide (25 mm in width and 75mm in height) which is horizontally placed, and observing the test phenomenon to obtain the results shown in Table 3;
TABLE 3 cohesion property
Figure SMS_4
As can be seen from the data in table 3 and fig. 1-3, the systems do not have an agglomeration effect in the absence of gemini silicon surfactants in comparative example 1 and comparative example 9. As can be seen from application example 15 and comparative example 7, when the copolymer is compounded with fatty alcohol-polyoxyethylene ether, the gathering effect of comparative example 7 is also poor. The comparative example 10 is a conventional commercial product, and a nonionic surfactant is generally added into the commercial product, but a polyether modified heptamethyltrisiloxane dimeric silicon surfactant is not added, so that the gathering effect is poor.
It can be seen from application example 15 and comparative example 8 that the gemini silicon surfactant does not produce a coagulation effect in a system obtained by compounding with all surfactants, and a good coagulation effect can be achieved only by compounding with the isomeric tridecanol polyoxyethylene ether and the auxiliary surfactant of the present invention.
4. Determination of Breast bacteria killing Effect before and after milk cow infiltration
The method comprises the steps of taking the medicinal bath liquid obtained in the application example 15, the comparative example 1, the comparative example 7, the comparative example 8, the comparative example 9, the comparative example 10 and a blank control group (distilled water) as samples to carry out a breast sterilization experiment, dividing the cows into 7 groups, selecting 30 cows in the lactation period for each group, respectively infiltrating the nipples by the samples, taking the medicinal bath liquid away after the infiltration time is 5s and 5s, staying the medicinal bath liquid on the surfaces of the breasts of the cows for 3min, and then cleaning the medicinal bath liquid by distilled water. Before infiltration, respectively use the aseptic cotton swab to sample at same nipple after the washing, cultivate and the colony count statistics according to disinfection technical specification mode, later calculate the average value of the bactericidal effect of dipping liquid to the nipple.
The calculation formula is as follows:
Figure SMS_5
the results obtained are shown in table 4:
TABLE 4 cow udder sterilization test results
Figure SMS_6
As can be seen from table 4, the bath liquid of application example 15 of the present invention has better bactericidal effect than that of comparative examples 1, 8 and 9; compared with the comparative example 7, the isomeric tridecanol polyoxyethylene ether system in the application example 15 of the invention has better bactericidal effect compared with other systems. Application example 15 of the present invention compared with comparative commercially available bath solution example 10, the bath solution of application example 15 of the present invention has a more excellent bactericidal effect as a bath solution before milking.
5, the protective effect of the medicated bath liquid on the nipples of the dairy cows
The protection of the mastitis of the cows was observed by conducting a teat protection test using application example 15, comparative example 1, comparative example 8, comparative example 9, comparative example 10 and a blank group (distilled water), dividing the cows into 6 groups, selecting 60 cows in the lactation period for each group, and performing an experimental period of 30 days, with the results shown in table 5:
TABLE 5 cow breast protection results
Figure SMS_7
As can be seen from the application example 15 and the comparative examples 1, 8 and 9, the medicated bath liquid with good coagulation effect of the application example 15 of the invention has better sterilization effect and better protection effect; as can be seen from comparison of application example 15 with comparative example 10, the application example 15 of the present invention has more excellent effect of preventing occult mastitis than the commercially available medicated bath solution.
