Abstract
本发明提供了一种面膜,其包括乳双歧杆菌的发酵产物,所述的乳双歧杆菌的保藏号为CGMCC NO.24307,于2022年1月14日保藏于中国普通微生物菌种保藏管理中心。该面膜中还包括银耳提取物、水解胶原、小麦培养提取物等。本发明提供的面膜具有抗衰、锁水、修护、舒缓的功效。
Description
技术领域
本发明属于化妆品领域,具体涉及一种包含乳双歧杆菌发酵产物的面膜。
背景技术
益生菌可以定殖在人体内,改变宿主某一部位菌群,可以通过调节宿主黏膜与系统免疫功能或通过调节肠道内菌群平衡,促进营养吸收保持肠道健康的作用,可以说益生菌与人类息息相关。实际上益生菌不仅可以在维持肠道菌群平衡中发挥作用,其发酵产物中富含的各种活性成分,当前也被广泛地应用于化妆品领域。
面膜作为日常护肤不可或缺的步骤,在化妆品中占据了很大的市场,其产品的种类也是丰富多样,市面上各种功效如:补水保湿、美白祛斑、抗衰老的面膜产品层出不迭。根据面膜的成分,一般可以划分为两种,一种是化学面膜,另一种是天然面膜。化学面膜多采用化学原料进行复配以达到面膜护肤的功效,天然面膜多采用天然提取物进行复配,其中植物提取物的应用范围尤其广泛。但以上两个种类的面膜尚都存在一些问题:化学面膜中采用的化工原料容易含有刺激性成分如酒精、防腐剂等,天然面膜中的提取物由于具体成分过于复杂也多容易致敏,而利用活性益生菌制备面膜可以作为改善以上情况的发展方向之一。
中国专利201910715219.2中公开了一种益生菌萃取物的生物纤维面膜及其制备方法,膜液具体包括以下组成成分:芦荟、珍珠粉、脱脂乳粉、葡萄糖、可溶性淀粉、乙基麦芽酚、海藻酸钠、食品级苯基酸钠、蛋白酶、脂肪酶、菌粉、蛋清、蜂蜜。该发明中将芦荟提取液和面膜基质液加入乳酸菌培养皿中,且在制作过程中加入甘草粉、土豆提取粉制得的生物纤维面膜的亲肤能力强,温和性高、抗菌、抗炎性高、且使用时,精华吸收率高。但其成分较为复杂,对于敏感肌肤容易造成负担。
中国专利201711297075.0中公开了一种含益生菌发酵液的美白面膜液及其制备方法。以酶解和生物发酵的方法对蛤蜊进行处理制备出了具有高抗氧活性的抗氧化肽,然后采用2-氨基丁醇作为稳定剂与化妆品辅料制备出高抗氧化的美白面膜液。本发明制备的美白面膜液理化性质稳定,自由基清除率高,产品绿色安全。但其制备方法复杂且发酵过程可控性不强。
挖掘适宜的菌种并优化其发酵方法,从而使其发酵产物更好地应用于面膜领域,是本领域亟待解决的问题。
发明内容
为了解决上述问题,本发明提供了一种包含乳双歧杆菌发酵产物的面膜。
本发明中,所述的“乳双歧杆菌”的发酵产物在化妆品领域也称为二裂酵母发酵产物。
一方面,本发明提供了一种面膜。
所述的面膜中包括乳双歧杆菌的发酵产物,所述的乳双歧杆菌的保藏号为CGMCCNO.24307,于2022年1月14日保藏于中国普通微生物菌种保藏管理中心。
所述的乳双歧杆菌发酵产物的制备方法为:将乳双歧杆菌发酵后采用碟式离心机去除菌体、悬浮颗粒等固态杂质,继而微滤去除可能导致发酵液变性的可溶性杂质,得发酵上清液。
所述的乳双歧杆菌的发酵条件为37±1℃,厌氧,培养72±3h。

当所述的面膜形态为涂抹面膜时,所述的面膜中还可能包括其他赋形剂等成分。

所述的制备方法用于制备前述的面膜
所述的制备方法中包括乳双歧杆菌发酵产物的制备,所述的乳双歧杆菌保藏号为CGMCC NO.24307。
所述的乳双歧杆菌发酵产物的制备方法中包括:将乳双歧杆菌发酵后采用碟式离心机去除菌体、悬浮颗粒等固态杂质,继而微滤去除可能导致发酵液变性的可溶性杂质,得发酵上清液。
所述的制备方法中还包括按照前述的配方进行复配。
本发明中的面膜中,富含海藻糖,海藻糖属于小分子保湿剂,与膜蛋白有很好的亲和性,可增加皮肤对营养成分的吸收,增加细胞的水化功能,有良好的保湿性能;
本发明中,水解胶原至少具有以下功效:
1、拥有多个RGD(由精氨酸、甘氨酸、天门冬氨酸组成)的重复序列,可与至少8种不同的整合素(整联蛋白)结合。通过与整合素的识别点位结合,从而参与纤维细胞的信号传导,大大促进纤维母细胞的分裂与增殖,提升细胞分化的速度与数量,最终实现新生细胞取代老细胞这一更替过程;
2、拥有大量的氨基酸片段序列,可特别识别胶原蛋白的分子伴侣Hsp47(热休克蛋白)中的三肽序列,通过靶向结合,有效牢固并稳定三螺旋结构,胶原蛋白只有在三螺旋结构下才可以承托肌肤,令肌肤持久水嫩、有光泽;
3、拥有的GER系列片段对纤维母细胞具有更强的粘附性,可有效修复纤维母细胞的损伤,从而提升其活性。
本发明中,乳双歧杆菌发酵产物至少具有以下功效:
1、产物中包含氨基酸、小肽、多糖、B族维生素等有益护肤的成分,可加强角质层代谢,去除细胞自由基,抑制脂质的过氧化,激活机体修复性能,抑制细胞酪氨酸酶活性,具有修复、美白、抗氧化的功能。
2、乳双歧杆菌发酵产物使各种介质的比例恢复平衡,利于免疫系统的调控,缓解皮肤炎症,具有抗菌、抗炎的作用。
3、产物中富含乳酸和乳酸钠,是人体天然保湿因子(NMF)中主要的水溶性酸类,吸湿性强,有效增强角质层的水合作用,防止皮肤水分流失;乳酸和乳酸钠组成缓冲溶液,对皮肤具有良好的亲和性,可调节皮肤酸碱值。
本发明的有益效果:
本发明提供的面膜具有抗衰、锁水、修护、舒缓的功效。经检测,本发明的面膜中具有显著免疫学活性的生物活性物质,采用生物发酵技术,高效提取核心功能活性物质,与多种佐剂科学复配而成,可全面增强皮肤耐受性,补水锁水等,具有透皮吸收快、生物利用度高等优势。

具体实施方式
下面结合具体实施例,对本发明作进一步详细的阐述,下述实施例不用于限制本发明,仅用于说明本发明。以下实施例中所使用的实验方法如无特殊说明,实施例中未注明具体条件的实验方法,通常按照常规条件,下述实施例中所使用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1一种乳双歧杆菌发酵产物的制备方法
乳双歧杆菌CGMCC NO.24307进行发酵,发酵培养基选择莫匹罗星锂盐和半胱氨酸盐酸盐改良MRS琼脂培养基(青岛海博生物),发酵条件为37℃,厌氧,培养72h,并对发酵产物进行检测。
结果表明:产物中包含氨基酸(14.26mg/g)、小肽(3.29mg/g)、多糖(13.24mg/g)、B族维生素(0.15mg/g)等有益护肤的成分,可加强角质层代谢,去除细胞自由基,抑制脂质的过氧化,激活机体修复性能,抑制细胞酪氨酸酶活性,具有修复、美白、抗氧化的功能。
在化妆品领域,乳双歧杆菌发酵产物也可以归为乳酸杆菌发酵产物大类。
实施例2一种护肤品活性成分的制备方法
对实施例1中的发酵产物进行处理制备乳双歧杆菌发酵产物活性成分,具体步骤如下:发酵结束后,采用碟式离心机去除菌体、悬浮颗粒等固态杂质,继而微滤去除可能导致发酵液变性的可溶性杂质等,得发酵上清液。
对产物进行检测结果如下:氨基酸(13.83mg/g)、小肽(3.15mg/g)、多糖(12.38mg/g)、B族维生素(0.15mg/g)、乳酸(11.84mg/g)和乳酸钠(9.23mg/g)。
各类活性成分检测分析方法如下:
(1)氨基酸
氨基酸标准曲线的绘制:氨基酸称取0.5g,用水溶解后,定容至100mL配成5g/L的氨基酸标准溶液,HPLC测定不同浓度的氨基酸标准溶液对应的峰面积,绘制标准曲线。发酵上清液中氨基酸含量测定:吸取发酵上清液2μL,使用特定衍生缓冲液稀释及定容,采用HPLC测定方法,根据氨基酸的保留时间和紫外吸收谱进行定性分析;外标峰面积法进行氨基酸定量测定,计算氨基酸的含量。
(2)小肽
定量取样,采用三氯乙酸TCA沉淀法,离心分离高分子蛋白,得到小肽和氨基酸的混合液体,再用凯氏定氮法测定总氮含量,得到小肽和氨基酸混合液中的氮含量,折算成绝对量,除以取样样品量,乘以6.25,即可得到发酵上清液中的小肽和氨基酸总含量,扣除氨基酸的含量,即为小肽的含量。
(3)多糖
蒽酮-硫酸比色法,葡萄糖标准曲线的绘制:定量称取葡萄糖标准品,配制浓度为0.5mg/mL的对照品溶液,并稀释成不同浓度,按照葡萄糖:0.1%蒽酮-硫酸溶液体积比1:4的比例,混匀,煮沸10min,迅速放冷,黑暗中房放置10min,显色后采用紫外分光光度法测定其在特定波下的吸光度,绘制标准曲线。定量量取发酵上清液上清液,经过浓缩干燥等后处理,得粗多糖。定量称取粗多糖,配制成特定浓度的溶液,照标准曲线制备项下的方法,显色后采用紫外分光光度法测定其吸光度,计算含量。
(4)B族维生素
B族维生素标准曲线的绘制:定量称取并溶解定容,然后稀释至不同的浓度,采用HPLC测定不同浓度的维生素标准溶液对应的峰面积,绘制标准曲线。发酵上清液中B族维生素含量测定:定量吸取发酵上清液,并溶解定容,采用HPLC测定方法,根据不同维生素的保留时间和紫外吸收谱进行定性分析;外标峰面积法进行维生素定量测定,计算B族维生素的含量。
(5)乳酸
L-乳酸标准曲线的绘制:定量量取L-乳酸标准品,配制成一定浓度对照品母液,并稀释成不同浓度,采用HPLC测定在210nm下的峰面积,绘制标准曲线。发酵上清液中乳酸含量测定:定量吸取发酵上清液,稀释并定容,采用HPLC测定方法,根据乳酸的保留时间和紫外吸收谱进行定性分析;外标峰面积法进行定量测定,计算乳酸的含量。
(6)乳酸钠
乳酸钠标准曲线的绘制:定量称取乳酸钠标准品100mg,稀释定容至100mL,摇匀,配制对照品母液,并稀释成不同浓度,采用HPLC测定峰面积,绘制标准曲线。发酵上清液中乳酸钠含量测定:定量吸取发酵上清液,稀释并定容,采用HPLC测定方法,根据乳酸钠的保留时间和紫外吸收谱进行定性分析;外标峰面积法进行定量测定,计算乳酸钠的含量。
实施例3乳双歧杆菌发酵产物抗氧化能力检测
称取DPPH 4mg溶于100mL无水乙醇。
取不同稀释倍数(蒸馏水稀释)的乳双歧杆菌发酵产物各1mL,加入3mL DPPH溶液,室温避光反应,6h后测定OD517(三组平行计算平均值,记为A值)。对照组1:1mL样品+3mL无水乙醇(三组平行计算平均值,测OD517记为B值);对照组2:1mL水+3mL DPPH溶液(三组平行计算平均值测OD517记为C值);DPPH自由基清除率计算公式如下:
C-(A-B)/C×100%;
最终测得本申请乳双歧杆菌发酵产物DPPH自由基清除率为42.7%。
实施例4一种包含乳双歧杆菌发酵产物的面膜
包括以下成分:

水解胶原购自武汉百瑞森生物医药有限公司;
二裂酵母发酵产物溶胞物购自上海比莱化工。
芽孢杆菌/谷氨酸发酵产物滤液为以谷氨酸为原料,通过芽孢杆菌常规条件发酵,然后过滤,得到发酵产物过滤液,本实施例中选用的芽孢杆菌为枯草芽孢杆菌CGMCC1.3358。
实施例5一种包含乳双歧杆菌发酵产物的面膜
包括以下成分:
乳双歧杆菌发酵产物参照实施例2的方法制备。
水解胶原购自武汉百瑞森生物医药有限公司;
二裂酵母发酵产物溶胞物购自上海比莱化工。
芽孢杆菌/谷氨酸发酵产物滤液为以谷氨酸为原料,通过芽孢杆菌常规条件发酵,然后过滤,得到发酵产物过滤液,本实施例中选用的芽孢杆菌为枯草芽孢杆菌CGMCC1.3358。
实施例6包含乳双歧杆菌发酵产物的面膜生物活性物质检测
将实施例4中提供的面膜委托第三方进行活性成分检测,检测步骤如下:
收集公司委托样品,提取、浓缩,BCA定量,准备进行监测分析。
选择Anti-EGF antibody[EPR19899](ab206423)为主要抗体,以WesternBlotting检测样品中活性成分,结果显示:样品含有具有显著免疫学活性的生物活性物质。如下图所示,参见第6道,1道为对照品,2、3道为市售其他产品。
结论,面膜富含活性成分。
实施例7包含乳双歧杆菌发酵产物的面膜活性成分透皮能力检测
将实施例4中提供的面膜送至深圳大学医学部药学院进行活性物质透皮能力检测,检测以《化学品皮肤吸收体外试验方法》(GB/T 27818-2011)为参考,选用Franz扩散池试验方案对样品活性物质透皮能力进行检测分析。结果显示:样品0.5h的渗透水平达到25%,1h渗透水平为39%,2h渗透水平为47%,接近峰值水平,4、12h渗透水平稳定48%。
结论:包含乳双歧杆菌发酵产物的面膜活性成分具有显著透皮能力。
实施例8面膜人群实验
征集80名志愿者(女性,年龄25-35),分为2组,每组40人,使用不包括乳双歧杆菌发酵产物的面膜的为对照组,其他为实验组,实验季节冬季,初次检测时间早上8:00,检测部位脸部,分别在使用面部精华前和使用面部精华后0h、6h、12h、24h进行皮肤含水量测试,对测试结果进行平均数计算,结果如下:
