Abstract
The invention discloses an oral care product for improving the remineralization efficiency of enamel, and a preparation method and application thereof, and particularly relates to the field of oral care. The oral care implement comprises a substrate comprising a soluble calcium salt, a substrate comprising a soluble phosphate salt, and a container separating the two substrates. When the toothpaste prepared by the invention is used, the matrix containing the soluble calcium salt and the matrix containing the soluble phosphate are simultaneously extruded from the mouth of the toothpaste and are mixed in the oral cavity, so that higher-concentration free calcium ions and free phosphate ions can be provided, the remineralization efficiency of enamel is improved, and the problem that one toothpaste simultaneously contains high-concentration calcium ions and phosphate ions and is easy to react is solved.
Description
Technical Field
The invention relates to the technical field of daily chemical and oral hard tissue medical care, nursing and cleaning products, in particular to a medical and oral cleaning nursing product, and more particularly relates to an oral care product for improving the remineralization efficiency of enamel, and a preparation method and application thereof.
Background
Enamel is the hardest tissue of the human body. Mature enamel contains approximately 96% hydroxyapatite, 3% water, and 1% protein and lipids by weight. As the outermost layer of the tooth, enamel is subject to various physical and chemical attacks such as occlusion, abrasion, and acid erosion from food and bacteria. In general, dissolution of hydroxyapatite at pH < 5.5 leads to demineralization problems of tooth enamel.
Saliva contains calcium ions and phosphate ions with certain concentrations, and the change of the pH value of the saliva can be buffered to a certain extent, so that the dissolution of hydroxyapatite is inhibited. At pH > 5.5 and at higher concentrations of calcium and phosphorus ions, calcium phosphate is formed in the saliva, which remineralizes the demineralized teeth. In general, saliva contains low concentrations of calcium and phosphate ions, and it is difficult to form sufficient amounts of hydroxyapatite to repair demineralized enamel.
The toothpaste added with calcium salt and phosphate can improve the concentration of calcium ions and phosphate ions in the oral cavity during tooth brushing, and is beneficial to the tooth remineralization process. However, when less soluble calcium salts and phosphate salts are added to toothpaste, the calcium ion concentration and phosphate ion concentration can be provided at a lower level. If calcium salt and phosphate with higher solubility are added into the toothpaste, the calcium ions and phosphate ions in the toothpaste rapidly react to form precipitates because the solubility product of calcium phosphate, calcium hydrophosphate, hydroxyapatite and the like is smaller, so that the concentration of free calcium ions and phosphate ions contained in the toothpaste is still lower. Thus, by adding calcium salts and phosphates directly to toothpaste, toothpaste can provide lower concentrations of free calcium and phosphate ions, with limited remineralization rates on enamel for limited brushing times and brushing frequencies.
Disclosure of Invention
Therefore, the invention provides an oral care product for improving the remineralization efficiency of enamel as well as a preparation method and application thereof, and aims to solve the problem that the concentration of free calcium ions and phosphate ions which can be utilized in the existing oral care product is low.
In order to achieve the above purpose, the invention provides the following technical scheme:
according to a first aspect of the present invention there is provided an oral care implement for increasing the efficiency of remineralisation of dental enamel, comprising:
a matrix comprising a soluble calcium salt, a matrix comprising a soluble phosphate salt, and a container separating the two matrices.
The matrix containing soluble calcium salt and the matrix containing soluble phosphate are separately arranged in the unconnected cavities and are extruded simultaneously when the dental enamel remineralization solution is used, so that the two ions do not react before use, and the concentrations of calcium ions and phosphate ions are ensured to the maximum extent, so that better dental enamel remineralization efficiency is achieved.
Further, the soluble calcium salt base comprises a soluble calcium salt and common auxiliary materials for preparing oral care products;
and/or, the matrix containing the soluble phosphate comprises the soluble phosphate and common auxiliary materials for preparing the oral care products.
Further, the soluble calcium salt is a substance which can be dissolved in water and alcohols and can generate free calcium ions. By way of example, the soluble calcium salts include, but are not limited to, one or more of calcium chloride, calcium hydroxide, calcium oxide, calcium nitrate, calcium gluconate, calcium bicarbonate, calcium lactate;
and/or the soluble phosphate is a compound that produces free phosphate, hydrogen phosphate, or dihydrogen phosphate ions, including, by way of example and not limitation, one or more of potassium phosphate, sodium phosphate, ammonium phosphate, potassium monohydrogen phosphate, sodium monohydrogen phosphate, ammonium monohydrogen phosphate, potassium dihydrogen phosphate, sodium dihydrogen phosphate, and ammonium dihydrogen phosphate.
Further, the concentration range of calcium ions in the soluble calcium salt is 0.0001-10 mol/L;
and/or the concentration range of phosphate ions in the soluble phosphate is 0.0001-10 mol/L.
Further, the common auxiliary materials for preparing the oral care product comprise at least one of an abrasive, a humectant, a surfactant, a thickening agent, a sweetening agent, a preservative, an active additive, a spice and water.
The abrasive is one or more of calcium carbonate, calcium hydrogen phosphate, calcium pyrophosphate, silicon dioxide and aluminum oxide which are commonly used in oral care preparation.
The humectant is one or more of glycerol, sorbitol, propylene glycol and polyethylene glycol.
The surfactant is a surfactant commonly used in preparation of oral care products, and comprises one or more of sodium dodecyl sulfate, sodium 2-acyloxy sulfonate and sodium lauroyl sarcosinate.
The thickening agent is one or more of carboxymethyl cellulose, antler pectin, hydroxyethyl cellulose, xanthan gum, guar gum and carrageenan which are commonly used for preparing oral care products.
The sweetening agent is one or more of common sweetening agents for preparing oral care products, including but not limited to glycerin, sodium cyclamate and saccharin sodium.
Preservatives commonly used preservatives for oral care products are prepared, including but not limited to potassium sorbate and/or sodium benzoate.
The active additive is one or more of bisoxyphenyldiguanidinohexane, cypress alcohol, copper chlorophyllin, stannous fluoride, sodium monofluorophosphate, sodium fluoride and ammonium fluoride which are commonly used for preparing oral care products.
The spice is one or more of menthol, peppermint oil and mint.
Further, the container for separating two substrates is internally divided into two unconnected cavities, and the two cavities share one container nozzle.
Further, the oral care product is a toothpaste or gel.
According to a second aspect of the present invention, there is provided a method of preparing an oral care implement for improving enamel remineralization efficiency as described above, comprising separately canning a substrate comprising a soluble calcium salt and a substrate comprising a soluble phosphate salt in a container having two cavities which are not connected to each other.
Further, the matrix containing the soluble calcium salt is obtained by uniformly mixing the soluble calcium salt with common auxiliary materials for preparing oral care products;
and/or the matrix containing the soluble phosphate is obtained by uniformly mixing the soluble phosphate and common auxiliary materials for preparing the oral care products.
According to a third aspect of the present invention there is provided a method of improving the efficiency of remineralisation of dental enamel by daily brushing using an oral care implement as described above or an oral care implement prepared as described above, or by simulating daily brushing in artificial saliva.
The invention has the following advantages:
the toothpaste tube is filled with the matrix containing soluble calcium salt and the matrix containing soluble phosphate in two separated cavities in the toothpaste tube, and the two cavities are communicated with the tube opening of the toothpaste tube. The soluble calcium salt-containing matrix and the soluble phosphate-containing matrix are not mixed until the toothpaste is used. When the toothpaste is used, the matrix containing the soluble calcium salt and the matrix containing the soluble phosphate are simultaneously extruded from the mouth of the toothpaste and are mixed in the oral cavity, so that high-concentration free calcium ions and free phosphate ions can be provided, the remineralization efficiency of enamel is improved, and the problem that one toothpaste simultaneously contains high-concentration calcium ions and phosphate ions and is easy to react is solved.
The preparation method is simple and easy to implement, and has simple process and strong practicability.

The present invention is described in terms of particular embodiments, other advantages and features of the invention will become apparent to those skilled in the art from the following disclosure, and it is to be understood that the described embodiments are merely exemplary of the invention and that it is not intended to limit the invention to the particular embodiments disclosed. All other embodiments, which can be obtained by a person skilled in the art without making any creative effort based on the embodiments in the present invention, belong to the protection scope of the present invention.
Example 1
The preparation of matrix a containing soluble calcium salts was as follows:
mixing 20ml of 0.1mol/l calcium chloride solution with 65g of 70 percent sorbitol uniformly and marking the solution 1 for later use; 2g of ethylene glycol is used for sequentially dispersing 0.5g of xanthan gum and 0.6g of sodium carboxymethyl cellulose, the xanthan gum and the sodium carboxymethyl cellulose are uniformly dispersed and poured into the solution 1 at the stirring speed of 50 revolutions per minute to prepare a gel phase;
adding 2g of sodium dodecyl sulfate and 12g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 300 revolutions per minute, degassing for 40 minutes, adding essence, then continuously stirring and degassing for 5 minutes to obtain a matrix A containing soluble calcium salt;
the preparation of matrix B containing soluble phosphate was as follows:
20ml of dipotassium phosphate solution containing 0.06mol/l is mixed with 65g of 70 percent sorbitol evenly to mark the solution 1 for standby; 2g of ethylene glycol is used for sequentially dispersing 0.5g of xanthan gum and 0.6g of sodium carboxymethyl cellulose, the xanthan gum and the sodium carboxymethyl cellulose are uniformly dispersed and poured into the solution 1 at the stirring speed of 50 revolutions per minute to prepare a gel phase;
adding 2g of sodium dodecyl sulfate and 12g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 300 revolutions per minute, degassing for 40 minutes, adding essence, continuing stirring, and degassing for 5 minutes to obtain a matrix B containing soluble phosphate;
the preparation method of the toothpaste containing the matrix A containing soluble calcium salt and the matrix B containing soluble phosphate at the same time comprises the following steps:
matrix a containing soluble calcium salt and matrix B containing soluble phosphate salt were filled into the same toothpaste tube. The toothpaste tube is provided with two separated cavities which respectively contain a substrate A containing soluble calcium salt and a substrate B containing soluble phosphate, but the two cavities share one toothpaste tube opening.
As shown in figure 1, the toothpaste comprises a substrate A cavity 1, a substrate B cavity 3 and a shell cap 2, wherein the shell is cylindrical, a double-tube structure is arranged in the shell, namely two cavities are arranged in the toothpaste, the two cavities are separated and extend upwards to a tube opening 4, and the substrate A containing soluble calcium salt is respectively arranged in the substrate A cavity 1; the matrix B containing soluble phosphate is filled into the cavity 3 of the matrix B, and the matrix A containing soluble calcium salt and the matrix B containing soluble phosphate are extruded at the same time when in use.
Example 2
The preparation of matrix a containing soluble calcium salts was as follows:
20ml of 0.5mol/l calcium lactate solution is mixed with 70g of 70 percent sorbitol to be uniformly mixed and marked with solution 1 for standby; dispersing 0.5g of xanthan gum and 0.7g of sodium carboxymethyl cellulose by 3g of ethylene glycol in sequence, uniformly dispersing, and pouring into the solution 1 at a stirring speed of 3000 r/min to prepare a gel phase;
adding 1.5g of sodium dodecyl sulfate and 17g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 300 revolutions per minute, degassing for 5 minutes, adding essence, continuing stirring, and degassing for 10 minutes to obtain a matrix A containing soluble calcium salt;
the preparation of matrix B containing soluble phosphate was as follows:
20ml of disodium hydrogen phosphate solution containing 0.3mol/l is mixed with 60g of 70 percent sorbitol to be uniformly mixed, and the solution 1 is marked for standby; dispersing 0.4g of xanthan gum and 0.8g of sodium carboxymethylcellulose in 5g of ethylene glycol in sequence, uniformly dispersing, and pouring into the solution 1 at a stirring speed of 300 revolutions per minute to prepare a gel phase;
adding 1.7g of sodium dodecyl sulfate and 20g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 300 revolutions per minute, degassing for 15 minutes, adding essence, continuing stirring, and degassing for 10 minutes to obtain a matrix B containing soluble phosphate;
the preparation method of the toothpaste containing the matrix A containing soluble calcium salt and the matrix B containing soluble phosphate at the same time comprises the following steps:
matrix a containing soluble calcium salt and matrix B containing soluble phosphate salt were filled into the same toothpaste tube. The toothpaste tube is provided with two separated cavities which respectively contain a substrate A containing soluble calcium salt and a substrate B containing soluble phosphate, but the two cavities share one toothpaste tube opening.
Preferably, as shown in fig. 2, the toothpaste comprises a substrate A cavity 1, a substrate B cavity 3 and a shell cap 2, wherein the shell is cylindrical, and a double-tube structure is arranged inside the shell, namely two cavities are arranged in the toothpaste, the two cavities are separated and extend upwards to a tube opening 4, and the substrate A containing soluble calcium salt is respectively arranged in the substrate A cavity 1; matrix B containing soluble phosphate is filled in the cavity 3 of the matrix B; the matrix A containing soluble calcium salt and matrix B containing soluble phosphate can be directly extruded for use.
Example 3
The preparation of matrix a containing soluble calcium salts was as follows:
mixing 25ml of 0.03mol/l calcium gluconate solution with 55g of 70% sorbitol to uniformly mark the solution 1 for later use; sequentially dispersing 0.9g of xanthan gum and 0.4g of sodium carboxymethylcellulose by using 4g of ethylene glycol, uniformly dispersing, and pouring into the solution 1 at a stirring speed of 1000 rpm to obtain a gel phase;
adding 2.2g of sodium dodecyl sulfate and 20g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 300 revolutions per minute, degassing for 10 minutes, adding essence, continuing stirring, and degassing for 5 minutes to obtain a matrix A containing soluble calcium salt;
the preparation of matrix B containing soluble phosphate was as follows:
mixing 15ml of sodium phosphate solution containing 0.03mol/l with 65g of 70 percent sorbitol to uniformly mark the solution 1 for later use; dispersing 0.7g of xanthan gum and 0.5g of sodium carboxymethyl cellulose by 5g of ethylene glycol in sequence, uniformly dispersing, and pouring into the solution 1 at a stirring speed of 300 revolutions per minute to prepare a gel phase;
adding 1.8g of sodium dodecyl sulfate and 20g of silicon dioxide into the gel phase, placing the gel phase into a paste making pot, stirring at the speed of 200 revolutions per minute, degassing for 15 minutes, adding essence, continuing stirring, and degassing for 10 minutes to obtain a matrix B containing soluble phosphate;
the preparation method of the toothpaste containing the matrix A containing soluble calcium salt and the matrix B containing soluble phosphate at the same time comprises the following steps:
the matrix a containing soluble calcium salt and the matrix B containing soluble phosphate salt are filled in the same toothpaste tube. The toothpaste tube is provided with two separated cavities which respectively contain a substrate A containing soluble calcium salt and a substrate B containing soluble phosphate, but the two cavities share one toothpaste tube opening.

Comparative example 1
The present comparative example is different from example 1 in that it is calcium phosphate-containing toothpaste obtained by directly mixing the soluble calcium salt-containing substrate a and the soluble phosphate-containing substrate B obtained in example 1.
Comparative example 2
The present comparative example is different from example 2 in that it is a hydroxyapatite-containing toothpaste obtained by directly and uniformly mixing the soluble calcium salt-containing substrate a and the soluble phosphate salt-containing substrate B obtained in example 2.
Test example
Enamel remineralization experiments:
the tooth enamel modules were brushed 3 times a day for 3 minutes after 15 days using the toothpastes prepared in example 1, example 2, comparative example 1 and comparative example 2, and the tooth enamel modules were cut from the side and the thickness of the growth of the tooth enamel modules was measured under a scanning electron microscope, and the results are shown in table 1.

As can be seen from table 1, the enamel remineralization rate of example 1 is more than 10 times that of the ordinary calcium phosphate toothpaste of comparative example 1; the enamel remineralization rate of example 2 was more than 12 times that of the ordinary calcium phosphate-containing toothpaste of comparative example 2.
Although the invention has been described in detail above with reference to a general description and specific examples, it will be apparent to one skilled in the art that modifications or improvements may be made thereto based on the invention. Accordingly, such modifications and improvements are intended to be within the scope of the invention as claimed.
Claims (10)
1. An oral care implement for increasing the efficiency of remineralization of enamel, comprising:
a substrate comprising a soluble calcium salt, a substrate comprising a soluble phosphate salt, and a container separating the two substrates.
2. The method of claim 1, wherein the dental composition further comprises a second ingredient selected from the group consisting of a mineral,
the soluble calcium salt base comprises soluble calcium salt and common auxiliary materials for preparing oral care products;
and/or, the matrix containing the soluble phosphate comprises the soluble phosphate and common auxiliary materials for preparing the oral care products.
3. The oral care implement for enhancing the remineralization efficiency of enamel according to claim 2,
the soluble calcium salt is selected from one or more of calcium chloride, calcium hydroxide, calcium oxide, calcium nitrate, calcium gluconate, calcium bicarbonate and calcium lactate;
and/or the soluble phosphate is selected from one or more of potassium phosphate, sodium phosphate, ammonium phosphate, potassium monohydrogen phosphate, sodium monohydrogen phosphate, ammonium monohydrogen phosphate, potassium dihydrogen phosphate, sodium dihydrogen phosphate and ammonium dihydrogen phosphate.
4. The oral care implement for enhancing the remineralization efficiency of enamel according to claim 3,
the concentration range of calcium ions in the soluble calcium salt is 0.0001-10 mol/L;
and/or the concentration range of phosphate ions in the soluble phosphate is 0.0001-10 mol/L.
5. The oral care implement for enhancing the remineralization efficiency of enamel according to claim 2,
the common auxiliary materials for preparing the oral care product comprise at least one of an abrasive agent, a humectant, a surfactant, a thickening agent, a sweetening agent, a preservative, an active additive, a spice and water.
6. The oral care implement for enhancing the remineralization efficiency of enamel according to claim 1 wherein said container separates two substrates, the interior of the container being divided into two discrete cavities, the two cavities sharing a container nozzle.
7. The oral care implement for increasing the efficiency of remineralization of enamel according to claim 1 wherein said oral care implement is a toothpaste or gel.
8. A method of making an oral care implement for enhancing the remineralization efficiency of enamel according to any one of claims 1 to 7,
respectively canning the substrate containing soluble calcium salt and the substrate containing soluble phosphate in a container with two unconnected cavities.
9. The method of claim 8, wherein the oral care composition is formulated to enhance the remineralization efficiency of enamel,

10. A method of increasing the remineralization efficiency of enamel by daily brushing using an oral care implement according to any one of claims 1 to 7 or an oral care implement prepared by the method according to claim 8 or 9, or by simulating daily brushing in artificial saliva.
