Performance testing
1. Skin micro-ecological balance test
The experimental principle is as follows: experiments are carried out to compare and test the absorption and utilization rates of the skin flora to the specific plant protoplasm and the common glucose, thereby researching the consumption process of the specific probiotic nutrients by the skin flora.
The experimental method comprises the following steps: a part of the flora on the skin surface was taken and transplanted into each of the groups of examples 1-6 (using the compositions of examples 1-6 as experimental media), comparative examples 1-14 (using the compositions of comparative examples 1-14 as comparative media), and a control (using a control medium of commercially available glucose), and the amount of the glucose/plant puree nutrient remaining in each group, i.e., the consumption level, was measured after 48 hours.
The consumed water balance criteria are shown in table 5.
TABLE 5 Water consumption balance Standard
Consumption level (%) Consumption state
0-20 Without consumption
20-40 Poor degree of consumption
40-60 Moderate degree of consumption
60-100 Up to total consumption
The results of the experiment are shown in Table 6.
Table 6 skin micro-ecological balance test results
Figure BDA0003524212450000161
Figure BDA0003524212450000171
According to tables 5-6, the probiotics and the pathogenic bacteria in the control group have higher absorption and utilization rate to the common glucose; by single factor outcome analysis: the synbiotics have the greatest effect on skin micro-ecology, the absorption and utilization rate of the synbiotics by probiotics is higher than that of pathogenic bacteria, wherein the lactobacillus fermentation product has the strongest inhibition effect on the pathogenic bacteria, the schizosaccharomyces fermentation product lysate has a certain inhibition effect, and the trehalose and beta-glucan have stronger promotion effect on the probiotics; the damaged repairing component has a certain effect on the skin micro-ecology; the absorption and utilization rate of the pathogenic bacteria on the sebum bionic components is slightly higher than that of the probiotics; the irritation inhibiting component has no effect on skin microecology. The embodiment group has higher absorption and utilization rate of nutrient components in the obtained composition through reasonable compounding of the components, has a synergistic effect, and is superior to a single component or a combination mode of any two or three components. The composition with the soothing and repairing effects provided by the invention can be utilized by probiotics on the surface of skin, can selectively protect the probiotics without being obviously metabolized by pathogenic bacteria, and is used as a biological inhibitor relative to the pathogenic bacteria.
2. Microbial competitive growth assay
The experimental principle is as follows: the effect and effect of specific compositions on the production of the flora in the presence of both probiotic and pathogenic bacteria was tested.
The experimental method comprises the following steps: the compositions of examples 1 to 4, the compositions of comparative examples 1 to 14 and the test medium (tryptone USP) without the added composition (control) were prepared at a concentration of 0.5%, respectively. The bacterial groups (mixed bacterial groups of Micrococcus clarkii and Staphylococcus aureus) are respectively cultured in each experimental medium, the number of Micrococcus clarkii after 0h, 6h and 24h culture (T0, T6 and T24) and the change rate of Micrococcus clarkii before and after 24h culture (T24/T0), the number of Staphylococcus aureus after 0h, 6h and 24h culture (T0, T6 and T24) and the change rate of Staphylococcus aureus before and after 24h culture (T24/T0) of each group are calculated. The test results are shown in table 7.
TABLE 7 microbial Competition test results
Figure BDA0003524212450000181
Figure BDA0003524212450000191
According to table 7, the compositions of examples 1 to 6 and comparative examples 1 to 14 all exhibited different degrees of promotion or inhibition of probiotics or pathogenic bacteria after 24h of cultivation. According to the single-factor experimental comparative examples 11-14, the composition has the most remarkable effects of promoting probiotics and inhibiting pathogenic bacteria, and shows that the nutrients in the composition can promote the growth of the probiotics, the growth of the pathogenic bacteria can not be promoted, but the number of the pathogenic bacteria is remarkably reduced due to lack of food and nutrition, so that the probiotics are selectively utilized by the probiotics, and the pathogenic bacteria are inhibited, wherein the lactobacillus fermentation product has the strongest effect of inhibiting the pathogenic bacteria; the stimulus-inhibiting ingredient has no effect on both probiotics and pathogenic bacteria; the damaged repairing components have certain promotion on probiotics and inhibition on pathogenic bacteria, but the effect is weak; the sebum bionic component has certain function of promoting the growth of pathogenic bacteria. Compared with comparative examples 1-14, the probiotic bacteria of example 1 are increased in number and the pathogenic bacteria are reduced in number, which shows that the components in the example are matched with each other, have synergistic effect and are better than a single component or a combination mode of any two or three components.
3. Test of moisturizing Effect
The experimental method comprises the following steps: 25 skin sensitive subjects aged 19-36 years were selected, the cream products of examples 1-6 and comparative examples 1-14 were used once a day at night, and the moisture content of the face was tested 20min after the subjects cleaned their face on days 0, 7 and 14 at 20 ± 2 ℃ and humidity (50 ± 10)% to evaluate the skin rejuvenation efficacy of the product. Each parameter was measured 3 times in succession and averaged to reduce measurement error.
Judging indexes: as shown in table 8.

TABLE 9 rate of change of moisture content
Figure BDA0003524212450000192
Figure BDA0003524212450000201
As can be seen from the data in Table 9, the creams of examples 1-6 and comparative examples 1-14 all had different degrees of effects on the moisture content of the skin compared to the control group; according to the single-factor experimental comparative examples 11-14, the bionic sebum component has the largest influence on the moisture content of the skin, and the moisture retention effect is the best; synbiotic and damaged repair ingredients have a certain effect on skin moisture; the stimulus suppressing component has no effect. In the bionic sebum component, the higher the ceramide and C10-30 acid cholesterol/lanosterol mixed ester is, the stronger the water holding capacity of the product is. Compared with comparative examples 1-14, example 1 has a better moisturizing effect, which shows that the components in the examples are matched with each other, have a synergistic effect, and are better than a single component or a combination mode of any two or three components.
4. Test of fading effect
The purpose of the experiment is as follows: an acute skin barrier injury model is established by a tape continuous pasting method, and the Erythema Index (EI) of the area is tested to judge whether the product has the function of repairing the skin barrier.
The test method comprises the following steps: selecting 25 skin sensitive subjects with age of 19-36 years, respectively using the cream products of examples 1-6 and comparative examples 1-14, selecting and marking a piece of skin with the size of 2cm multiplied by 2cm at the equivalent position of the two forearms of the subject, randomly dividing the two test areas into a natural repair area and a sample intervention area after establishing an acute skin barrier damage model, and selecting an undamaged area with the equivalent size at the adjacent position of the natural repair area as a blank control area. The natural repair area and the blank control area do not use products, and the sample intervention area uses corresponding products. And (3) testing the red pigment conditions of the natural restoration area, the sample intervention area and the blank control area within 30min by using a melanin index and erythema index measuring instrument.
And (3) judging standard: and judging the state of the barrier repair process for recovering the damaged skin according to the erythema index, wherein the higher the repair rate is, the better the red fading effect of the product is.
The results of the red fading test are shown in table 10.
TABLE 10 results of the discoloration test
Figure BDA0003524212450000211
Figure BDA0003524212450000221
According to table 10, the creams of examples 1 to 6 have a remarkable immediate repairing effect on damaged skin, showing red fading and soothing in a short time, compared to comparative examples 1 to 14 and natural repair zones. From the test results of the single-factor experimental comparative examples 11-14, the irritation-inhibiting component has obvious effects of fading red and relieving, the sebum-simulating component has certain effects of fading red and relieving, the synbiotic and the damaged repairing component have no instant repairing effect, and part of the comparative example cream has the effect of relieving skin fading instead, so that adverse effects appear. The components in the embodiment are matched with each other, have synergistic effect, are superior to a single component or a combination mode of any two or three components, and the red fading effect of the hydroxyphenyl propionamide benzoic acid is superior to that of the bisabolol.
5. Evaluation test for lactic acid stimulation efficacy on face
This test is used to evaluate the skin barrier repair efficacy of the product.
The experimental method comprises the following steps: selecting 40 skin sensitive subjects, age 19-36 years, performing test at nasolabial sulcus of the subjects, dripping 50uL of 10% lactic acid solution on a single-layer filter paper with diameter of 0.8cm, respectively inquiring the subjects for stabbing pain at 3min, 4min and 5min, grading according to 4-point method (0 is no stabbing pain, 1 is mild stabbing pain, 2 is moderate stabbing pain and 3 is severe stabbing pain), and recording the grading of the volunteers as the lactic acid stimulation grading (D)0). The product application was carried out as required 2 times a day (examples 1-6 and comparative examples 1-14 creams were used, respectively) for 28 consecutive days and for the 14 th day of the test (D)14) And day 28 (D)28) And (6) performing return visit.
The results of the experiment are shown in Table 11.
TABLE 11 lactic acid test results
Figure BDA0003524212450000222
Figure BDA0003524212450000231
According to table 11, the creams of examples 1 to 6 and comparative examples 1 to 14 have certain repairing effect on skin barrier, can strengthen skin, enhance skin resistance and relieve response to external stimulation, and the effect of damaged repairing components on enhancing skin barrier is most obvious from single-factor experimental comparative examples 11 to 14; the synbiotic and sebum bionic components have a certain repairing effect in 14 days; although the irritation-suppressing component has immediate effects of fading red and soothing, the long-acting repairing effect is not good. The components in the embodiment are mutually matched, have synergistic action and are superior to a single component or a combination mode of any two or three components, and the combination of the inonotus obliquus extract and the madecassoside is superior to the repairing effect of the tetrahydro-methylpyrimidine carboxylic acid.
Therefore, the reasonable combination of the synbiotic, the stimulation and inhibition component, the sebum bionic component and the damaged repair component has good relieving and repairing effects, can improve the skin problem from multiple dimensions, can balance the skin micro-ecology, does not use a bactericide but uses a microorganism competition mode to kill harmful bacteria, and can effectively relieve acne and inflammation; can increase the water content of the horny layer, reduce the loss of external water of the horny layer of the skin and solve the skin discomfort caused by dryness; skin problems such as redness, swelling, itching, fever and the like caused by external stimulation can be quickly relieved without using hormone components; has effects of strengthening skin barrier and enhancing skin tolerance after long-term use. The combination can be reasonably adjusted, and the problem of various sensitive muscles can be solved in a targeted manner.
Finally, it should be noted that the above embodiment modes are only used for illustrating the technical solutions of the present application and are not limited, and although the present application is described in detail with reference to the above preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made on the technical solutions of the present application without departing from the spirit and scope of the technical solutions of the present application.
