Abstract
本发明公开了一种婴幼儿手口湿巾抗微生物组合物及其制备方法,该抗微生物组合物包括如下组份及质量百分比:防腐剂8%~15%;快速杀菌剂3%~8%;助防腐剂20%~30%;多功能助剂40%~60%;溶剂12‑25%;上述各组份质量百分比之和为100%;所述防腐剂为苯甲酸;所述快速杀菌剂为月桂酰精氨酸乙酯盐酸盐(LAE);所述助防腐剂为山梨坦辛酸酯;所述多功能助剂为1,2‑己二醇;所述溶剂为1,3丁二醇。本发明采用食品级及准食品级防腐杀菌剂,配合绿色助防腐剂,得到了毒理安全,防腐效果可靠,无刺激性的婴幼儿手口湿巾抗微生物组合物,应用于婴幼儿手口湿巾中,达到了理想的预期目标。

技术领域
本发明属于组合物技术领域,涉及一种婴幼儿手口湿巾抗微生物组合物及其制备方法。
背景技术
湿巾由浸有湿巾液的多孔或吸收性片材制成,将它们存储在气密的容器或包装纸中,以防止片材变干。湿巾具有多种用途,主要用途有两类:第一类是用于一般家庭清洁目的,例如用于清洁硬表面,如地板或厨房表面;第二类是用于个人清洁的,例如用于卸妆或婴儿清洁等。湿巾还可以用于女性健康和成人失禁产品以及公共卫生用品。
用于清洁人体皮肤的湿巾中有很大一部分是用于婴幼儿,由于3岁以下婴童皮肤会变得非常敏感,其生殖器和肛门周围的皮肤尤其重要。因此,用于婴幼儿的手口湿巾要求很严,材质用全棉或木浆纸基质,清洁剂及防腐剂毒理安全性高,特别是易引起过敏的防腐剂以天然或低刺激材料为主。由于材质特殊,水及营养物存在,除了细菌感染外,真菌感染会导致发霉变质。为了防止这些微生物的繁殖和生长,一般都需要加入一定量的防腐剂。
CN201810001163.提供了一种手口湿巾及其制备方法。手口湿巾包括湿巾液和湿巾载体;所述湿巾液包括如下组分且各组分的重量比例为润肤剂:0.2%~0.5%;杀菌剂:4.1%~7.1%;溶菌酶:0.1%~0.5%;pH调节剂:0.3%~0.6%;余量为去离子水。本发明采用纯天然的、具有消炎、抗菌、抗病毒的溶菌酶,搭配杀菌剂构建防腐杀菌体系,不但防腐效果好,而且对皮肤温和无刺激,安全无毒。所述杀菌剂为聚六亚甲基双胍、无味L抗菌剂、三氯生、三环唑或者双十烷基二甲基氯化铵中的至少三种的混合物。遗憾的是三氯生早在2016年已经被美国FDA禁用于手用杀菌剂,三环唑不是《2015化妆品安全技术规范》中规定限制使用的防腐剂。
CN201310014515涉及一种婴儿手口湿巾及其制备方法,由湿巾复配液和承附复配液的无纺布组成,所述湿巾复配液按每100重量份计含有:0.01-7.5%的甜味剂;0.01-2.0%的防腐剂;0.01-2.0%的清洁剂;0.05-5.0%的增溶剂;0-10.0%的保湿剂;0-5.0%的护肤剂;0-0.5%的酸度调节剂;0-5.0%的精油;0-0.5%的稳定剂;0-1.0%的天然香精;余量为纯水。防腐剂为Geogard Ultra防腐剂、苯扎氯铵、对羟基苯甲酸甲酯钠及其钠盐、对羟基苯甲酸乙酯及其钠盐、聚六亚甲基单胍、聚六亚甲基双胍盐酸盐、苯氧乙醇、甲基异噻唑啉酮、山梨酸钾、脱氢醋酸、苯甲醇、二氯苯甲醇、乙基己基甘油、辛二醇、辛甘醇中任意一种或几种的混合。防腐剂为对羟基苯甲酸甲酯、对羟基苯甲酸乙酯、苯扎氯铵、GeogardUltra防腐剂中任意一种或几种的混合(其中Geogard Ultra防腐剂为D-萄糖酸-1,5-内酯和苯甲酸钠混合物)。事实上,异噻唑啉酮,尼泊金酯(对羟基苯甲酸酯)因生殖毒性和高刺激性在化妆品中逐渐弃用,在婴幼儿手口湿巾中更是无法使用。
CN201410647761.6提供了一种婴幼儿手口湿巾,通过对黄瓜汁进行闪式提取,将黄瓜提取液的润肤特性与三氯生的广谱抗菌性,以及氨基酸型表面活性剂温和亲肤性有效配伍,制备出接近人肌肤的弱酸性清洗液,并以无纺布为载体制备的湿巾,具有安全无刺激,杀菌效果显著的特点。事实上,三氯生早在2016年已经被美国FDA禁用于手用杀菌剂,其毒理安全性遭到普遍质疑。
CN201510226199.4提供一种婴儿手口湿巾复配液及湿巾。该复配液包括防腐剂,所述防腐剂在所述复配液中的质量百分含量为0.01%-1%,所述防腐剂为聚六亚甲基双胍、聚六亚甲基单胍、葡萄糖酸氯己定、聚季铵盐-15、尼泊金酯及钠盐、苯甲醇、甲基异噻唑啉酮、苯甲酸及钠盐、乳酸链球菌素、纳他霉素、乙二胺四乙酸二钠、葡萄糖酸内酯、脱氢醋酸中任意一种或两种以上的混合物,其中,所述防腐剂中的两种以上的混合物的组合中不包含同时含有所述葡萄酸内酯与所述苯甲酸钠盐的混合物。事实上,异噻唑啉酮,尼泊金酯(对羟基苯甲酸酯)因生殖毒性和高刺激性在化妆品中逐渐弃用,在手口湿巾中早已不用,苯甲醇因极易氧化为苯甲醛而有苦杏仁味,不适合用在手口湿巾中防腐。至于聚季铵盐-15因是甲醛缓释剂更是湿巾之大忌。
CN201710466627.X公开的婴儿手口湿巾由以下质量百分比的物质组成:西吡氯铵0.03~0.05%;氯己定二葡萄糖酸盐0.5~0.65%;乙基已基甘油0.05~0.08%;余量为纯化水。虽然该申请中的防腐剂选用属于低刺激体系,但防霉性能显然不足,且整个配方保湿、润肤功效远远不足(尽管乙基己基甘油有保湿作用,但量太小。而且整个体系没有足够增溶乙基己基甘油的方案)。
传统的防霉剂主要有尼泊金酯系列和碘丙炔基丁基氨甲酸酯(以下简称IPBC)等,在近年的使用过程中,这些品种也颇受争议,出现一些与安全性相关的热点问题。如IPBC虽然是效果最好的一种防霉剂,通常只需要几个ppm就能够起到非常好的防霉效果,但是由于其含碘量较高,法规对其使用的限制也越来越严格,特别是在婴幼儿产品的应用方面,欧盟之前的化妆品规程(Directive76/768/EEC)及最新施行的化妆品法规[Regulation
(ECNo1223/2009]中都规定,IPBC不能用于三岁以下的婴幼儿配方(洗浴用品和洗发水除外),在用量方面也受到了严格的限制。由于具有一定的雌激素效应,尼泊金酯系列品的安全性也受到了不少质疑。
CN CN201310511646.1提供一种含有植物提取物、协同增效剂和防腐杀菌剂的抗微生物组合物(含IPBC),通过协同增效作用,同时复配多种防腐剂,以达到对细菌和真菌的广谱抑菌效果,但IPBC在婴幼儿湿巾中属于法规禁用。
USP 8,377,459用于湿巾的组合物,增强清洁效果,同时对皮肤温和。防腐体系为尼泊金酯和苯氧乙醇。尽管该体系刺进性较小,温和,但苯氧乙醇的使用易于引起发热,尼泊金酯因生殖毒性备受争议。
因此,市场上急需要温和无刺激,毒理安全性良好,适合婴幼儿皮肤特点,护肤清洁性良好的新型手口湿巾。而这一切,重点是建立科学的防腐体系,特别是对于婴幼儿湿巾用的全棉或木浆纸基材湿巾的防腐剂的选择十分重要,因为传统的防腐剂已不适应法规或市场需要。
发明内容
本发明的目的是提供一种配方合理、毒理安全、防腐防霉效果好的婴幼儿手口湿巾抗微生物组合物及其制备方法。
为实现上述目的,本发明采用的技术方案为:
该抗微生物组合物包括如下组份及质量百分比:
防腐剂8%~15%;快速杀菌剂3%~8%;助防腐剂20%~30%;多功能助剂40%~60%;溶剂12-25%;上述各组份质量百分比之和为100%;所述防腐剂为苯甲酸;所述快速杀菌剂为月桂酰精氨酸乙酯盐酸盐(LAE);所述助防腐剂为山梨坦辛酸酯;所述多功能助剂为1,2-己二醇;所述溶剂为1,3丁二醇。
上述所述抗微生物组合物优选的组份及质量百分比为:防腐剂:8%~10%;快速杀菌剂:6%~8%;助防腐剂:25%~30%;多功能助剂:40%~50%:溶剂:15-20%;上述各组份质量百分比之和为100%。
该抗微生物组合物的制备方法包括如下步骤:
①将快速杀菌剂搅拌溶解在多功能助剂和溶剂中,继续搅拌加入防腐剂溶解;
②将助防腐剂溶解在步骤①混合物中,过滤,即得到浅黄色透明婴幼儿手口湿巾抗微生物组合物,其pH值为3-3.5;
所述防腐剂为苯甲酸;所述快速杀菌剂为月桂酰精氨酸乙酯盐酸盐(LAE);所述助防腐剂为山梨坦辛酸酯;所述多功能助剂为1,2-己二醇;所述溶剂为1,3丁二醇。
本发明具有如下优点:
1、充分利用原料的优异性能:
月桂酰精氨酸乙酯盐酸盐(LAE),作为高效低度杀菌剂已在美国和澳大利亚及部分欧洲国家作为食品防腐剂使用。中国已在2020年12月作为化妆品防腐剂使用,使得《2015化妆品安全技术规范》中规定的限用防腐剂由51种增加到52种,有关部门正在以欧盟食品标注起草我国的LAE在食品中的使用标准。作为一种新型的阳离子杀菌剂,安全无刺激是其亮点。月桂酰精氨酸乙酯盐酸盐(LAE)有着极好的安全性,它在人体内通过化学和代谢途径水解为人体日常饮食中摄入的化合物。本申请主要利用其快速高效的杀菌作用,使得湿巾在生产的过程中就杀灭原料中的大多数微生物,为后续保存建立基础。
山梨坦辛酸酯本身就是一种安全的防腐剂,由于其两亲分子结构,具有一定的表面活性,是一种防腐增效剂,可极大提高防腐剂的功效和产品配方的融合性。山梨坦辛酸酯破坏有害微生物的细胞膜,这使防腐剂更容易渗透到细胞中从内部攻击细菌和真菌并杀死它们。山梨坦辛酸酯有效减少了达到同等功效所需的多种防腐剂混合成分数量。由于不受“防腐剂准许列表”限制,所以该成分能以任何用量与其他防腐剂配合使用,为各类防腐困难的产品提供防腐保障。山梨坦辛酸酯是真正绿色防腐剂的代表,其由天然淀粉衍生物山梨醇与来自椰子油的辛酸酯化得到,极易在环境中降解。它的主要功能涵盖了乳化剂、润肤剂及增粘剂的特征。分子中多羟基的存在,具有优异的保湿效果。酯类产品的本质又使其具有润滑护肤作用。该种多功能化妆品成分没有致敏性,所以适用于各种驻留型、冲洗型配方以及面膜、湿纸巾类产品。权威检测表明,在0.5%的使用浓度下对皮肤无刺激。因其复合绿色概念,获得国际著名组织颁发的Ecocert认证。
苯甲酸及其盐是最常用的食品防腐剂,以其安全性著名,特别适合用于婴幼儿护理产品中。虽然其防腐作用有限,但在山梨坦辛酸酯及己二醇的协同下,其防腐性能大大提升,抑菌的pH上限可以拓展到6.5。因其防腐范围在酸性条件下,而湿巾正好在人体pH的弱酸性范围,因此,苯甲酸可以发挥较好的防腐作用。
1,2-己二醇是著名的无刺激防腐剂促经剂,自身也具备一定的对细菌的抑制效果,也是有益的保湿剂。在本申请中利用其对细菌的抑制作用及良好的溶解作用,同时可以增效苯甲酸的防腐功效。
选用1,3-丁二醇作为溶剂的目的,主要是整个组合物可能在低温时会析出,造成使用困难,失去防腐保湿功效。同时1,3-丁二醇也是一种保湿剂。低温时,低冰点的1,3-丁二醇将使得组合物处于溶液状态。
2、充分利用组合物中防腐组分的协同增效作用:
防腐剂的协同作用,使得可以用最少的防腐剂,最大的微生物耐受性,进而产生对婴幼儿良好的皮肤保护作用。
下表1是测试的山梨坦辛酸酯与苯甲酸之间(3:1)的协同增效情况。

各组分之间的增效性:表1是相关各组分的最小抑菌浓度。
根据Kull E.C.等1961年在《应用微生物》中所述的协同增效系数SI计算方法:
SI=Qa/QA+Qb/QB
QA—防腐剂A单独作用时的最小抑菌浓度
QB---防腐剂B单独作用时的最小抑菌浓度
Qa—混合防腐剂最小抑菌浓度时A的浓度(即A+B物质组合时最小抑菌浓度×A所占比例)
Qb—混合防腐剂最小抑菌浓度时B的浓度(即A+B物质组合时最小抑菌浓度×B所占比例)
如果SI大于1时,表明两种防腐剂有拮抗作用;SI=1时具有相加作用;SI小于1时具有协同增效作用。
山梨坦辛酸酯与苯甲酸(比例3:1)在pH=5.5时,对金黄色葡萄球菌增效系数为1,具有加和作用;对绿脓杆菌和大肠杆菌的增效系数为0.82,对黑曲霉的增效系数为0.75。因此,二者合理比例的复配具有强的增效作用。
具体实施方式
实施例1:
按照如下质量百分比称取组份:苯甲酸8%、LAE 6%、山梨坦辛酸酯25%、1.2-己二醇42%、1.3-丁二醇19%。将快速杀菌剂LAE搅拌溶解在1,2己二醇和1,3丁二醇的混合物中溶解后,继续搅拌加入苯甲酸钠溶解;最后将山梨坦辛酸酯助防腐剂溶解在上述混合物中,过滤后得到浅黄色透明的婴幼儿手口湿巾抗微生物组合物。
实施例2:
按照如下质量百分比称取组份:苯甲酸12%、LAE 3%、山梨坦辛酸酯30%、1.2-己二醇40%、1.3-丁二醇15%,将快速杀菌剂LAE搅拌溶解在1,2己二醇和1,3丁二醇中,继续搅拌加入苯甲酸钠溶解;将山梨坦辛酸酯助防腐剂溶解在上述混合物中,过滤后得到浅黄色透明的婴幼儿手口湿巾抗微生物组合物。
实施例3:
按照如下质量百分比称取组份:苯甲酸10%、LAE 8%、山梨坦辛酸酯20%、1.2-己二醇50%、1.3-丁二醇12%。制备方法同实施例1,得到浅黄色透明的婴幼儿手口湿巾抗微生物组合物。
对比例1:
按照如下质量百分比称取组份:苯甲酸8%、山梨坦辛酸酯25%、1.2-己二醇42%、1.3-丁二醇25%,制备方法同实施例1,得到浅黄色透明的婴幼儿手口湿巾抗微生物组合物。同实施例1不同的是配方中没有加入快速杀菌剂LAE,其LAE所占重量百分数由几乎无杀菌功效的1.3-丁二醇代替。
实验结果:
杀菌率测试:在相同条件下,分别取相同细菌培养液,接种相同的铜绿假单胞菌、金黄色葡萄球菌和大肠杆菌,再加入实施例1和对比例1的防腐杀菌剂,加入量均为1%(重量百分数)。将上述各实验组培养液放至恒温微生物培养箱中,保持温度在25~28℃进行培养,在不同的时间段测定不同组别的菌落数,计算杀菌率。
应用试验结果测试:本发明中婴幼儿手口湿巾液的示例性配方如下:
调整湿巾液的最终pH为5.5.
给湿巾液中接种规定量的经过活化的绿脓杆菌、金黄色葡萄球菌和大肠杆菌、白色念珠球菌和黑曲霉菌,按照常规的湿巾制备方法,得到婴幼儿手口湿巾。
参照美国药典(第21版)上微生物挑战性试验方法。将接入微生物的湿巾置于28℃下培养。在接菌的0、7、14、21和28天取样分析:准确称取3g湿巾液(取至加菌培养的湿巾)样品,加到含有玻璃小珠的灭菌锥形瓶内,加入27ml灭菌生理盐水,充分震荡混匀,此悬液为1∶10稀释液;然后再用灭菌生理盐水按10倍依次稀释。按平板倾注法计数受试品中含菌量,细菌培养是37℃下24h~48h,霉菌培养为28℃下3~5天。此实验用以评判防腐剂的有效与否。评判标准为:当每克样品中一次接菌(1×106细菌和1×105霉菌)后,在第14天存活菌量减少至不高于起始浓度的0.1%,以后逐渐减少,在28天为0。符合标准为防腐剂有效(通过测试),不符合为防腐剂无效(不通过测试),表3以不加防腐剂的湿巾作为空白对照。将实施例1和对比例1进行微生物(细菌)杀灭试验,结果如下表2:

表2结果表明,对比例1和实施例1相比,由于缺少快速杀菌剂LAE的加入,杀菌速度大大降低,2天后的杀菌率仅达到40-51.8%,远低于实施例1的93.4-98.6%。而且表3的应用试验表明,对比例1不能通过婴幼儿手口湿巾的防腐挑战试验。实施例1和实施例2顺利通过微生物挑战试验。

实施例的婴幼儿手口湿巾抗微生物组合物1%的水溶液及应用例湿巾液的皮肤刺激性试验结果:
采用目前化妆品流行的鸡胚绒毛刺激法测试本发明的婴幼儿手口湿巾液的刺激性。按照上海日用化学品行业协会团体标准(T/SHRH 011-2018)“化妆品眼刺激性/腐蚀性的鸡胚绒毛尿囊膜试验”方法。绒毛尿囊膜(CAM)是一个呼吸性膜,包围在鸡胚周围。由于鸡胚尿囊膜表面血管丰富,可以看作一个完整的生物体,本试验利用孵化的鸡胚中期绒毛尿囊膜血管系统完整、清晰和透明的特点,将一定量受试物直接与鸡胚尿囊膜接触,作用规定时间之后观察绒毛尿囊膜毒性效应指标(如:出血、凝血和血管融解)的变化,给予评分,计算数学平均值用于评估受试物的眼刺激性。
按以下公式计算ES平均分值,按表4判断刺激分类。

本发明按照此标准规范进行鸡胚绒毛尿囊膜试验,实施例的婴幼儿手口湿巾抗微生物组合物1%的水溶液及应用例湿巾液的皮肤刺激性试验结果,表5结果表明ES值在3-4之间,属于无刺激性范围。

本发明采用食品级及准食品级防腐杀菌剂,配合绿色助防腐剂,得到了毒理安全,防腐效果可靠,无刺激性的婴幼儿手口湿巾抗微生物组合物,应用于婴幼儿手口湿巾中,达到了理想的预期目标。
Claims (3)
1.一种婴幼儿手口湿巾抗微生物组合物,该抗微生物组合物包括如下组份及质量百分比:防腐剂8%~15%;快速杀菌剂3%~8%;助防腐剂20%~30%;多功能助剂40%~60%;溶剂12-25%;上述各组份质量百分比之和为100%;所述防腐剂为苯甲酸;所述快速杀菌剂为月桂酰精氨酸乙酯盐酸盐;所述助防腐剂为山梨坦辛酸酯;所述多功能助剂为1,2-己二醇;所述溶剂为1,3丁二醇。
2.根据权利要求1所述的一种婴幼儿手口湿巾抗微生物组合物,其特征是:该抗微生物组合物包括如下组份及质量百分比:防腐剂:8%~10%;快速杀菌剂:6%~8%;助防腐剂:25%~30%;多功能助剂:40%~50%:溶剂:15-20%;上述各组份质量百分比之和为100%。
3.一种婴幼儿手口湿巾抗微生物组合物的制备方法,包括如下步骤:
①将快速杀菌剂搅拌溶解在多功能助剂和溶剂中,继续搅拌加入防腐剂溶解;
②将助防腐剂溶解在步骤①混合物中,过滤,即得到浅黄色透明婴幼儿手口湿巾抗微生物组合物,其pH值为3-3.5;
所述防腐剂为苯甲酸;所述快速杀菌剂为月桂酰精氨酸乙酯盐酸盐;所述助防腐剂为山梨坦辛酸酯;所述多功能助剂为1,2-己二醇;所述溶剂为1,3丁二醇。
